[Effect of a single gonadotropin injection on the uterus of prepuberal gilts].

Histological studies were conducted into the uteri and macroscopic studies into the ovaries of 64 gilts, aged between 180 and 220 days, following administration of 400 I.U. of pregnant mare serum gonadotrophin (PMSG) with 200 I.U. of human chorionic gonadotrophin (hCG) or 500 I.U. of PMSG with 250 I.U. of hCG, in order to test the action of one single injection of PMSG and hCG on the reproductive organs of prepuberal gilts. Ovulation was achieved in something between 75 and 100 per cent of all test animals which were treated in four experimental groups. The mean ovulation rates, following injection of 500 I.U. of PMSG with 250 I.U. of hCG, were between 17.2 and 28.6 (superovulation). The changes induced to the uterus by gonadotrophin injection were similar to those resulting from spontaneous onset of puberty, with the effectiveness of 500 I.U. of PMSG with 250 I.U. of hCG having been stronger than that of 400 I.U. of PMSG with 200 I.U. of hCG. However, histometric examinations have shown that uterus developed with induced cycle was not fully identical with uterus development with spontaneous onset of sexual maturity. Only in about 30 per cent of all animals tested was an induced cycle spontaneously followed by another cycle. Uterus growth was stimulated by gonadotrophin injection even in animals with no further cyclic activity.

[Control of puerperium in swine under conditions of extremely shortened nursing method. 1. Results of histological studies in postpartum involution and regeneration of the uterus under conditions of extremely shortened nursing period and medicamentous control of puerperium].

Uterus tissue samples were taken from 31 sows which had been under regimes of extremely shortened nursing periods and differentiated medication to control their puerperium. The samples were taken following laparotomy during the first postpartum oestrus as well as in subsequent phases of the cycle and histologically tested for puerperal involution and regeneration. The endometrium and myometrium of sows in oestrus were found to be in a condition which widely corresponded to normal oestrus condition of the uterus wall, when their piglets had been weaned in the third postpartum week. However, untreated sows in oestrus, whose piglets had been weaned extremely soon, that is in the second postpartum week, displayed numerous signs of inadequate postpartum involution and regeneration. Oxytocin was found to work well in speeding up regeneration of both the endometrium and myometrium. Intramuscular injection of twice nine International Units per die, from the first through ninth days postpartum, helped to complete largely all regeneration processes up to the first oestrus, when piglets had been weaned in the second postpartum week. Oral antibiotic medication (chlortetracycline) and parenteral application of vitamins failed to produce any substantive and histologically recordable effect on puerperal involution processes.

[Control of swine parturition. 1. Results of clinical, histological and hematological studies of parturition induction in swine using prostaglandin F2 alpha].

Parturition was induced to 23 sows on the 112th day of pregnancy by intramuscular injection of 7.5 mg PGF2 alpha (Dinoprost-Upjohn Co.). Onset of parturition was achieved within 27 hours and 52 minutes +/- four hours and 54 minutes in 21 animals (91.3 per cent). Average litter size was 11.33 piglets, with parturition time per litter having been five hours and 45 minutes +/- two hours and 39 minutes. Stillbirths accounted for 6.3 per cent. The average birth weight, 1.245 g +/- 264 g did not differ with significance from that of the controls. The physiological process accompanying parturition is described in greater detail. Unobstructed re-intergration with further reproduction of animals which had received PGF2 alpha treatment is established with hard evidence on the basis of clinical examination of oestrus, ovulation, conception, and farrowing rates as well as by histological examination of the endometrium. Alterations in the blood state recorded from animals with PGF2 alpha treatment close to full term were identical with those recorded from untreated animals.

[Control of parturition in swine. 2. Study results of parturition synchronization in swine with the prostaglandin analog cloprostenol (I.C.I. 80.996: Estrumate)].

Premature parturition was induced in 115 of 121 sows (95%) that had been given injections of 150 microgram or 200 microgram of "Estrumate", a prostaglandin analogues, on the 112th and 113th days of gestation. The average time that elapsed between injection and parturition of those 115 animals was 27.2 +/- 7.7 hours, with no difference being observed between gilts, on the one hand, and old sows, on the other. The time of gestation of the treated animals was shortened with significance from 114.6 +/- 1.6 days for the controls to 113.2 +/- 0.4 days. The average duration of parturition proper was 3.7 +/- 2.2 hours, the average litter being 11.2 +/- 3.3 piglets. The total number of piglets born was 1,320, with stillbirths accounting for 4.17%. The average weight at birth, 1,330 +/- 210 g, and the weight at weaning did not differ from weight figures recorded from the controls. The courses of parturition and puerperium were in agreement with normal biological standards. The sows treated were tested for their blood counts close to full term and for their endometrial histomorphology after weaning, with no side effects of "Estrumate" treatment being established. The behaviours of progesterone, 17-beta oestradiol, and cortisol in serum of "Estrumate"-treated sows were investigated, and congruence was established between spontaneous and prostaglandin-induced parturitions, which was in agreement with international literature. The conclusion is that 150 micrograms of 'Estrumate", administered to swine, after the 11th day of gestation has very good labour-inducing effects and ensures normal physiological parturition and piglet development.

[Trials of rut induction with prostaglandin F2alpha in gilts].

The possibility of oestrus induction by means of F-2-alpha prostaglandin was checked on nine gilts. The animals received intramuscular injections of 7.5 mg of 10 mg of PG F2alpha on the tenth or twelfth day of cycle. The sexual behaviours of the animals was followed up, and after slaughter followed macroscopic assessment of the ovaries as well as histomorphological and histochemical examinations of the uterus wall. The results have shown that no oestrus or ovulation could be induced by such injection on the tenth or twelfth day of cycle. Regression of corpora lutea and follicle growth five days following application on the twelfth day of cycle were somewhat faster than the same phenomena when observed following injection on the tenth day. However, oestrus and ovulation failed throughout to occur earlier than they would have occurred any way without any treatment. The cycle was not shortened by application of F-2-alpha prostaglandin.

[Histological findings on the uterus of gilts following synchronized ovulation].

Histological studies were undertaken with the view to testing uterus structure and function of gilts following synchronised ovulation by means of different PMSG doses. All proband groups received 500 I.U. HCG. All histomorphological, histochemical, and histometric checks revealed 500 I.U. PMSG to be too low a dosage, whereas the optimum amount was found to be between 750 and 1,000 I.U. PMSG. The uterine glands of all treated animals in all three groups were less favourably developed than those of the untreated controls. The best morphologico-histochemical pattern was observed following administration of 750 I.U. PMSG.

[Results of experimental studies to control birth in swine].

An experiment was applied to ovulation-synchronised gilts and old sows. Mean values of pregnancy time were almost one and the same. Variations between the 111th and 117th days were exhibited within the control group. The test animals, however, farrowed on the 109th, 110th, and 111th days within spans of three days (113th to 115th day of pregnancy), following intramuscular injection of 75 mg DXMS. Intramuscular injection of 5 mg prostaglandin F2alpha on the 112th day of pregnancy triggered parturition within 25-36 hours in ten of 13 treated sows, whereas 19 of 20 sows gave birth within 27.7 +/- 5.1 hours in response to injection of 7.5 mg PGF2alpha.

[Vaginal cytology studies on the determination of the onset of estrus cycle post partum in sheep].

State of vaginal cells and arborisation grade of cervical mucus were studied in 364 Merino ewes of meat type from the 7. day ante partum. The Pundel modified Shorr-straining method and the Papanicolaou-straining method were used for differentiation of the cells (casal-, parabasal-, intermediate-and superficialcells). Near the 44., 54. and 70. day post partum a maximum of superficialcells and simultaneously a high grade of cervical mucus arborisation in elongating cyclic intervals can be observed in lactating ewes. In conclusion an early weaning of lambs between the 40. and 50. day post partum is considered to be in aggreement with cyclus.

[Studies on improving the fertility of aged sows. 3. Results of histological studies on puerperal involution and regeneration processes in the endometrium of lactating sows and sows after extremely early weaning].

Involution and regeneration following parturition was studied in 48 sows allowed to suckle their piglets normally, or from which the piglets were taken away at 5-8 days of age. Samples of uterus were obtained by biopsy 4, 8, 11, 14, 17, 20, 30, 32 and 40 days after parturition. In lactating sows, endometrial changes similar to those of oestrus in a normal cycle were present 32 days after parturition. This fact seems to justify the removal of piglets at 21-28 days of age. After very early weaning (5-8 days), incomplete oestrus-like changes in the endometrium were seen on the 17th day after parturition. The observed involutionary and regenerative changes in the puerperal uterus suggest that early weaning after 10-12 days of suckling is feasible, but it may be advisable to use drugs that would accelerate uterine involution.

[Studies on improving the fertility of aged sows. 2. Results of histological studies on the structure of the endometrium in aged sows during estrous cycle].

Histomorphological and histometric techniques were employed to study cyclic processes in the endometrium. Specimens of uterus were obtained on the 1st, 5th, 8th, 13th, 17th and 21st days of the cycle. The effect of oestrogens, which are produced in large amounts during pro-oestrus, oestrus and to some extent in metoestrus, was reflected in proliferative processes in the epithelium, with increases in the cell population, thickness of the epithelium, and height of the superficial and glandular epithelium. During oestrus the subepithelial layer was broad and was rich in lymphocytes and neutrophile grandulocytes, while the lamina propria showed considerable oedema and hyperaemia. The action of gestagens during dioestrus led to an increase in glandular structures due to the development of more convolutions and a more even distribution of glands throughout the entire propria. There was evidence of active glandular secretion, which during the course of dioestrus resulted in a relatively low and single-layered superficial and glandular epithelium.
