OBJECTIVE: Dental anatomy is one of the most important basic courses in the education of dentistry. The deep understanding of both the external and internal morphologic characteristics is very important to the teaching, research and clinical practice of dentistry. METHODS: In the present study, 32 permanent teeth from the skull specimen of a young man were individually embedded in black fluid resin, each in special containers. They were ground by a numerically-controlled grinding machine at intervals of 0.2 mm per layer. A distinct outline of the tooth could be seen on every section. Black and white photographs were taken and scanned into a computer by a film-scanner to obtain sequences of two-dimensional images of the tooth sections. After pattern recognition, all of the images of each tooth were piled up at intervals of 0.2 mm by the technique of computerized reconstruction. RESULTS: The three-dimensional stereo tooth models were built up from two-dimensional data. On the basis of those models, computer graphic techniques were used to highlight, smooth, and shade the teeth. These digital teeth and dentitions could function as a series of computerized teaching models. Their high resolution and accuracy could meet the basic demands of dentistry. CONCLUSIONS: The present study developed new techniques of model preparation, image input, and 3-D reconstruction. These digital teeth and dentitions provide an important foundation for the application of computer imaging, CAD/CAM, computer-assisted instruction, and virtual reality in dentistry.
Volatile organic compounds (VOCs) emitted from indoor materials was a major cause of indoor air pollution. The characteristics of VOCs emission was an important part of research programs on indoor air quality. The technology of test chambers with exactly controllable conditions has been successfully used in studies of VOCs emissions. The technology could be used to model the chamber VOCs concentration level vs time profile C(t), which could in turn be used to estimate the sample emission rate vs time profile R(t). The emission models of VOCs from indoor materials were presented in this review. The principal of emission process, parameters and the applications of emission models were introduced. The application of diffusion model, dilution model and vapor pressure (VP) model were limited due to the existence of sink effect. Sink model is the most promising model at present.

By the techniques of reconstruction, the 3D stereo tooth models were retrieved from 2D digital data. RESULTS: On the basis of those 3D data, computer graphics techniques were used to realize the lighted, smoothed and shaded teeth. CONCLUSION: Such a series of data teeth might be applicated as a corner stone of the computer aided instruction (CAI) in stomatology.

We have sequenced the 5'-flanking region of the Drosophila melanogaster norpA gene and characterized its promoter. The potential promoter region, which was deduced from the determination of the transcription start point (tsp), lacks a distinct TATA box sequence. Deletion analysis of the promoter region suggests that the minimal promoter necessary for efficient expression of the gene is located between -138 (PstI) and +278 relative to the tsp. Within this minimal promoter region, at least two downstream regulatory elements responsible for the stimulation of gene expression seem to exist in the DNA fragments between +44 and +121 and between +214 and +278. Among these, the DNA fragment between +44 and +121 affects promoter activity more dramatically (about 6-7 fold). This DNA fragment contains the consensus promoter element previously reported to be important for photoreceptor cell-specific expression, and this promoter element seems to be working in the norpA gene expression.
Advanced glycation end products of bovine serum albumin (AGEs-BSA) exhibited biphasic effects on the proliferation of cultured rabbit vascular smooth muscle cells (VSMCs) in terms of [3H]thymidine incorporation and cell number count; a stimulatory effect was observed at 1-10 micrograms/ml and an inhibitory effect at more than 20 micrograms/ml, while it inhibited [3H]thymidine incorporation even at 1-10 micrograms/ml in cultured bovine vascular endothelial cells (VECs). Transient activation of p42 mitogen-activated protein kinase (MAPK) with a peak at around 5 min and a subsequent sustained phase was induced by AGEs-BSA in VSMCs, but not in VECs. The dependence of MAPK activation on AGEs-BSA dose was correlated with that of VSMCs proliferation.
Bombesin-like peptides, including the mammalian homologue gastrin-releasing peptides, are highly expressed and secreted by neuroendocrine cells in prostate carcinoma (PCa) tissues and are likely to be related to the progression of this disease. In the present study, we show that bombesin enhances the migration of androgen-independent PCa cells (PC-3) in vitro, while not affecting their adhesion to extracellular matrix proteins. The bombesin-increased motility of PC-3 cells occurs through its receptor, and, as shown with inhibitors, it likely requires activation of both protein tyrosine kinases (PTKs) and protein kinases C (PKCs). Because the focal adhesion kinase pp125FAK plays a key role in adhesion/motility and is highly expressed in advanced PCa, we examined whether in PC-3 cells bombesin signal transduction triggers the tyrosine phosphorylation of this PTK and of associated integrins and signaling proteins likely to be present in focal adhesion plaques. pp125FAK tyrosine phosphorylation was stimulated by bombesin and mimicked by PKC activation with the tumor-promotor phorbol 12-myristate-13-acetate (PMA). Moreover, this effect of bombesin on pp125FAK tyrosine phosphorylation requires the presence of both active PKC and cytoskeleton integrity since this signal was abolished by down-regulating PKCs induced by prolonged PMA treatment or by PKC inhibition with GF 109203X, as well as by disruption of the cytoskeleton with cytochalasin D. We also show that bombesin increases the tyrosine phosphorylation of a 95-kDa protein (pp95) which was co-immunoprecipitated with the alpha v and beta (3 and 5) subunits, forming integrin receptors with alpha v in PC-3 cells. The protein pp95 is distinct from the endogenously tyrosine-phosphorylated beta3 subunit. In addition, upon bombesin treatment, the beta1, beta3 and beta5 integrin subunits co-immunoprecipitated with pp125FAK and major phosphotyrosine (pY)-containing proteins of 125 and 68-70 kDa, likely corresponding to pp125FAK and paxillin. Together our data suggest that, in addition to PKC activation, tyrosine phosphorylation of pp125FAK and integrin-associated proteins may play an important role in bombesin signaling, triggering the processes of PCa cell motility and invasion.
We have isolated and characterized a genomic clone encoding the Drosophila melanogaster transcription factor IIB (TFIIB). The coding region of the TFIIB gene is interrupted by three short introns. The 5'-flanking region of the gene lacks the typical TATA box sequence like those of other known genes encoding the general transcription factors. In addition, the 5'-flanking region of the gene contains several common DNA sequences present in Drosophila TBP and TFIIS genes, suggesting the common regulation mechanism of gene expression. RNA blot analysis revealed that the gene expresses 1.6 kb, 1.3 kb and 1.2 kb mRNAs throughout development and in adults. Deletion analysis of the promoter region shows that the minimal promoter necessary for efficient expression is located between -698 (PstI) and +60 relative to the transcription start point. Within this minimal promoter region, the upstream regulatory element responsible for the stimulation of gene expression may exist in the DNA fragment between -698 (PsfI) and -351 (StuI).
BACKGROUND: The objective of this study was to characterize the calcium mobilizing activity of bombesin-related neuropeptides in PC-3 cells. METHODS: Intracellular free Ca2+ concentration ([Ca2+]i) was measured following the administration of bombesin-related neuropeptides using spectrofluorometric monitoring of PC-3 cells loaded with Fura2/AM. RESULTS: Individual neuropeptides induced a rapid rise in [Ca2+]i immediately after stimulation in similar fashion. When two different peptides were administered simultaneously, the [Ca2+]i changes were similar to that produced by each peptide alone. However, simultaneous addition of tyr4-bombesin 100 nM and 2% serum resulted in [Ca2+]i increase greater than either alone. Sequential stimulation by same or different peptides, was not able to induce another response. However, 6 hrs after the first stimulus, a second stimulus was able to induce a similar response. CONCLUSIONS: The data indicate that all members of the bombesin-related family of neuropeptides are capable of triggering calcium mobilization in PC-3 cells via a similar signalling pathway. PC-3 cells become acutely desensitized to additional stimulation of neuropeptide. This desensitization is selective and reversible, requiring approximately 6 hrs for recovery.
This paper describes a cost-effective technique that optimally utilizes all available diagnostic studies for three-dimensional treatment planning. A simulator unit modified to produce cross-sectional images (simulator-CT unit) is used to create a reference data set with the patient in the treatment position. Registration software (qsh) brings other diagnostic studies into agreement with this reference data set. Two cases are presented as examples of the use of this technique. Registration of abdominal scans from the same patient demonstrates the warping of a nontreatment position study to the treatment position. The second case is based on paired data sets through the head, in which the diagnostic study was obtained by using a gantry tilt to follow the base of the skull and to avoid sections passing through the teeth. The registration software provides a method for combining diagnostic studies into a single "master" data set. The success of the transformation depends on the operator's ability to identify corresponding anatomic landmarks for different data sets and on the magnitude of the variation in the patient's position from one procedure to the next. Limitations in image quality and the number of cross-sections obtainable from a simulator-CT unit can be partially overcome by using the described technique. Thus, the information contained in nontreatment position diagnostic tests can be used accurately for treatment planning at limited cost.
This paper demonstrates examples of successful reconstruction of partial-thickness eyelid defects using the expanded forehead flap. Two patients are presented: one in whom cicatrical ectropion was present and the other in whom there was an absence of ectropion. In both patients primary grafting was executed just after the injury was sustained. By the time the patients were referred for reconstruction, mismatch in the quality of the skin and scars along the border of the graft left the area of primary repair to be impaired aesthetically as well as functionally. Reconstruction of the upper eyelid using the expanded forehead flap resulted in excellent approximation of the native tissue. Aesthetic as well as functional capacities were considered in this approximation. The advantage of this technique is that it offers a larger amount of tissue without compromising the aesthetic or functional similarity to the native eyelid.
Cellular autophagy in the cells of distal tubular segments (DT cells) of the kidney cortex in streptozotocin-diabetic rats was evaluated by quantitative electron microscopy. Five days after streptozotocin administration, volume and numerical densities of autophagic vacuoles (AVs) in DT cells were reduced by 56 and 57%, respectively. The correction of the diabetic state by insulin injection reversed the inhibition of cellular autophagy. Volume and numerical densities of AVs increased by 97 and 53%, respectively. Endogenous insulin replacement by islet transplantation showed the same effect on cellular autophagy. Volume and numerical densities of AVs increased by 82 and 34%, respectively, as compared with sham-operated diabetic animals. The data show that, during diabetic kidney hypertrophy, the cellular autophagy is inhibited in DT cells to the same extent as in proximal tubular cells, suggesting that DT cells contribute in a balanced manner to hypertrophic growth of the kidney cortex. Similarly, DT cells are involved in the catabolic reaction, i.e., stimulation of autophagy, after metabolic correction of the diabetic state by insulin.
The improperiety of the location of implants would cause many difficulties to prosthetic procedure. The function, appearance and long-term health of implant denture would then be effected. Dental stents were used to resolve the above problems for 30 cases of implant denture involved 74 implants in present study. A radiographic stent with given diameter sphere can be used to eliminate distortion and determine the bone quantity. A surgical stent can express the prosthetic plan to surgeons during operation directly in the mouths of patients. It is very helpful to the surgeons to take account of both the condition of jaw bone and the future denture making. Holes, tunnels and windows on surgical stents can be used as the guidances of drilling positions and directions. The surgical stents may also be a useful reference to surgeons to make bone graft, do some changes in surgical plan and even cancel the implant denture therapy when the situation of jaw bone significantly differ from what the expectancy.

I have developed an efficient method for transfecting Drosophila S2 cells using DDAB, a cationic liposome reagent. The optimized DDAB method resulted in a 10 times or greater increase in transfection efficiency compared with the conventional calcium phosphate method which has been essentially the only way for transfecting S2 cells.
In an attempt to clarify the role of platelet-activating factor (PAF) in the pathogenesis of hepatic injury induced by galactosamine (GalN) plus lipopolysaccharide (LPS), effects of WEB 2086 (PAF receptor antagonist) on hepatic injury in vivo as well as on neutrophil adherence to hepatic endothelial cells in vitro have been investigated, as we have recently clarified the role of neutrophils in this experimental model of hepatic injury. Although an enhanced serum TNF-alpha level after GalN-LPS administration was not reduced by WEB 2086, hepatic injury and hepatic neutrophil accumulation in the liver after GalN-LPS administration were attenuated by WEB 2086. An in vitro study revealed that an enhanced neutrophil adhesion to hepatic endothelial cells by stimulation with the sera that were collected from the GalN-LPS-treated rats, was reduced in the presence of WEB 2086 in a dose-dependent manner. In addition, LPS, TNF-alpha, and PAF were found to enhance the neutrophil adherence to hepatic endothelial cells, which was reduced in the presence of WEB 2086. These results suggest that PAF play an important role in the GalN-LPS induced hepatic injury and that PAF receptor antagonist reduces the neutrophil adherence to hepatic endothelial cells in the liver.
