The lung-damaging effect of intratracheally administered cellulose was studied by biochemical and histological methods. Cell count, protein, phospholipid, lactate dehydrogenase and acid phosphatase were determined in bronchoalveolar lavage fluid 1, 3 and 7 days after intratracheal instillation. Histological tests were performed after days 1, 3 and 30. In vitro, cellulose did not damage the macrophage cells. In vivo, interstitial oedema as well as the initial signs of inflammation could be detected in the lung after the first day. Inflammation after 1 week could be noted, partly interstitial and partly intra-alveolar and intrabronchial. In the bronchoalveolar lavage fluid, protein, lactate dehydrogenase, acid phosphatase, phospholipid and cell count were enhanced after days 1 and 3. After 1 month, the developing bronchioalveolitis is fibrous in character. Contrary to the in vivo study, cellulose did not damage rat peritoneal macrophages.
The pulmonary toxicity of two potential environmental pollutants was studied in rats 1, 7 and 30 days after a single intratracheal instillation of lead nitrate and Dithane M-45 (mancoceb), either individually or in various combinations. The cell count, protein, phospholipids and lactate dehydrogenase level were determined in the bronchoalveolar lavage fluid, as were the protein, phospholipids and acid phosphatase contents in the lung tissue. Lead nitrate and Dithane M-45 induced acute inflammation reactions with different features. The effects of mixtures of lead nitrate and Dithane M-45 were found to be different from those of the individual components.
To discover the antigenicity-producing mechanism of acetylsalicylic acid, the interaction of this drug and relevant salicylic acid with human serum albumin (HSA) has been studied by means of nuclear magnetic resonance (NMR) spectroscopy. The determination of spin-lattice relaxation rates (1/T1) of some protons have revealed that one HSA molecule can bind acetylsalicylate and salicylate up to 80 and 290 molecules, respectively. The hydrolysis rates of acetylsalicylate were greatly enhanced in the presence of HSA, especially when the drug/HSA mole ratio was small. Thus, the esterase-like activity of HSA was verified. This activity of HSA was effectively inhibited by salicylate; the effect was ascribed to the stronger binding affinity of salicylate toward HSA as compared with that of acetylsalicylate. Based on these results, the antigenicity-producing mechanism of acetylsalicylate and salicylate has been discussed.
In this short overview, features of the two suprachiasmatic nucleus (SCN)-independent oscillators are described. One is restricted daily feeding (RF)-associated oscillator and the other is methamphetamine-dependent oscillator. We found that both oscillators have free-running period of circadian range, and express themselves in the special state of energy supply or energy deficiency. The expression of the RF-associated oscillator depends heavily on the catecholaminergic input from the medulla, whereas the methamphetamine-dependent oscillator appears to depend on the sympathomimetic effects of the drug. It is plausible that overt rhythms of both oscillators are complex, mainly because of the interaction of hourglass principle with circadian clock mechanism.
Effects of dietary consistency were examined on the development and persistence of meal anticipation in adrenocortical activity in rats under restricted daily feeding (RF), in which food supply was restricted to a fixed time of day. Restricted daily solid meal produced the anticipatory hormone peak in 8 days, whereas restricted daily liquid meal produced the peak in 12 days. The developed hormone peak was less prominent by liquid meal than by solid. The anticipatory corticosterone peak developed by solid meal persisted throughout the period examined (3 weeks) after the termination of RF, whereas the peak developed by liquid meal disappeared after the first week. It is concluded that liquid meal attenuates the meal anticipation in the adrenocortical activity in rats under RF.
We report a case of a 78-year-old woman with cerebellar infarction who developed intracerebral bleeding during the magnetic resonance (MR) examination. The intensity of the hematoma was heterogeneous corresponding to the heterogeneous density shown on CT study that immediately followed the MR examination.

We aimed to explore whether and how the pineal influences the adrenal medulla, the area postrema and the brain water content; and is influenced by sham pinealectomy (SPX) in its structures and melatonin secretion. Quantitative morphological methods were mainly used in rats and golden hamsters. Experimental results showed: (1) Pineal effects on the A cell system of the adrenal medulla were inhibitory to the synthesis and secretion of epinephrine into the perivascular space and stimulatory to the opioid synthesis, and augmented by SPX. Time-of-day changes were also influenced by SPX and PX. (2) The sizes of a part of the nuclei of nerve and glia cells in the area postrema and the brain water content increased by SPX depending on the presence of the pineal, suggesting the pineal's stimulatory effects on these following SPX. (3) SPX caused morphological changes suggesting an increased activity of pinealocytes. However, corresponding consistent increases in plasma melatonin levels were not seen following SPX. Possible interpretations of these observations were discussed.
Plasma melatonin rhythm in humans was investigated: its stability, relationship to the sleep-wake rhythm, and response to light. The so-called day-to-day variation of reference phases of plasma melatonin rhythm was within 1.4 h when blood was sampled at 1-hour intervals. Therefore, a change in phase beyond this value is regarded as a phase shift of melatonin rhythm in individuals. Plasma melatonin rhythm was spontaneously desynchronized from the sleep-wake rhythm and probably regulated by the common circadian pacemaker which drives the rhythm in rectal temperature. When a bright-light pulse was applied, the melatonin rhythm produced a phase shift, but the amount of phase shift seems to be different for the ascending and descending phases of nocturnal melatonin rise. Finally, a partial entrainment was observed in a subject who developed a non-24-hour sleep-wake syndrome later, in which the plasma melatonin rhythm was free-running whereas the sleep-wake rhythm was apparently entrained by a 24-hour day-night alternation. It is concluded that the plasma melatonin rhythm is the best marker of the human circadian pacemaker so far available.
We herein present a case of synchronous triple cancer, which was successfully resected in a curative manner. These cancers consisted of primary duodenal, pancreatic and lung cancers, which were diagnosed in an asymptomatic 74 year-old male, who was referred to our department on December 14, 1996. On admission, his laboratory data showed no abnormality, including tumor markers (CEA 1.0, CA 19-9 1.0, AFP 8.1 U/ml), but he did show an impaired pulmonary function (FEV1.0: 57%). Upper gastrointestinal endoscopy showed a smooth surfaced duodenal tumor measuring 4 cm in size. The second tumor was found at the head of the pancreas by computed tomography (CT), showing a hypervascular mass measuring 3.0 cm, along with neighboring multiple cysts. In endoscopic retrograde cholangiopancreatography (ERCP), marked mucous secretion was observed through the papilla, while a filling defect was found in the dilated pancreatic duct. In a routine chest X-ray, a third tumor, which measured 1.5 cm in diameter, was recognized in the right upper lobe of the lung, and a moderately differentiated squamous cell carcinoma was also detected by a percutaneous CT guided biopsy. The pancreatic and duodenal tumors were surgically resected by a pancreatoduodenectomy (Stage I) in January 1997 and, 5 months later, a lung tumor underwent partial resection (Stage I). This patient tolerated these surgical procedures well and presently leads a normal, healthy life after discharge. In summary, a successful resection of synchronous triple cancers, which has never been previously reported in this specific combination, is described.
Neuropathological findings were described in a 9-day-old female infant who died of the vitamin B12 non-responsive (mut0) type of methylmalonic acidemia (MMA). Widespread karyorhectic fragments of varying size and shape were noted throughout the brain, in particular densely accumulated in the cerebellar granular layers and the layer IV of the striate cortex. Bilateral or symmetrical necrotic foci were observed in various regions of the grey matter: Sommer's sector of the hippocampus, basal ganglia, thalamus, hypothalamus and brainstem. In the cerebral cortex small spongy necrotic foci were scattered mainly in the depths of gyri. Alzheimer type II astrocytes appeared in the preserved zone of the caudate nucleus. Myelinated nerve fibers in the brainstem were spongy or vacuolated, whereas peripheral myelin sheaths of cranial nerves were intact. Multiple hemorrhagic foci were noted in the cerebellum, predominantly the granular layers. The lymphoid tissue in the spleen and the thymus was hypoplastic. It may be difficult to explain exactly the mechanisms of the pathological changes observed here on routine light microscopy; the outcome of systemic ischemia/hypoxia before death cannot be completely ignored. But, it is suggested that widespread karyorhexis may occur selectively in specific cells (or cell groups), including immature neurons and other cellular components (glial and/or mesenchymal cells) among the patients with the mut0 type of MMA.
