Characterization and purification of the membrane-bound ATPase of the archaebacterium Methanosarcina barkeri.

Membrane-bound ATPase was found in membranes of the archaebacterium Methanosarcina barkeri. The ATPase activity required divalent cations, Mg2+ or Mn2+, and maximum activity was obtained at pH 5.2. The activity was specifically stimulated by HSO3- with a shift of optimal pH to 5.8, and N,N'-dicyclohexylcarbodiimide inhibited ATP hydrolysis. The enzyme could be solubilized from membranes by incubation in 1 mM Tris-maleate buffer (pH 6.9) containing 0.5 mM EDTA. The solubilized ATPase was purified by DEAE-Sepharose and Sephacryl S-300 chromatography. The molecular weight of the purified enzyme was estimated to be 420,000 by gel filtration through Sephacryl S-300. Polyacrylamide gel electrophoresis in sodium dodecyl sulfate revealed two classes of subunit, Mr 62,000 (alpha) and 49,000 (beta) associated in the molar ratio 1:1. These results suggest that the ATPase of M. barkeri is similar to the F0F1 type ATPase found in many eubacteria.

[The evaluation of single administration of cisplatin in non-small cell carcinoma of the lung].

The effects of chemotherapy by single administration of Cisplatin were studied in 26 patients with non-small cell carcinoma of the lung. There were 22 males and 4 females with a median age of 63 years. 21 cases were epidermoid carcinoma, 4 cases were adenocarcinoma and 1 case was atypical carcinoid. Cisplatin 20 mg/m2 with hydration and diuresis was given daily for 5 consecutive days. The course was repeated every 3 weeks. Partial response was observed in 4 patients (3 epidermoid carcinoma and one atypical carcinoid). The response rate was 19%. Side effects induced by Cisplatin were gastrointestinal toxicity including vomiting, nausea and appetite less, bone marrow toxicity and renal damage. These were not so severe, and reversible. Gastrointestinal toxicities were controlled successfully by corticosteroids. In 12 patients gastrointestinal toxicities were not observed. We conclude that Cisplatin is effective for non-small cell carcinoma, especially for epidermoid carcinoma. Hematologic toxicities of Cisplatin were not so severe. Therefore, combination chemotherapy including Cisplatin would improve the quality of life of patients suffering from non-small cell carcinoma of the lung.

Lymphatic system or hydro-oncotic forces. Which is more significant in drainage of pleural fluid?

Artificial pleural fluids, containing plasma protein from 0.1 to 9.0 g per 100 ml, indocyanine green (ICG) and para-aminohippurate (PAH), were injected into the pleural cavity of dogs. The concentration of these two indicators were measured at a regular time interval, and the lymphatic and non-lymphatic flows and PAH clearance were calculated by Stewart's and Takashina's equations. When the protein concentration in the pleural fluid was less than 4 g per 100 ml, the pleural fluid volume decreased at a rate of 0.20 ml/kg/hour. The decrease was due to pleural fluid drainage by the lymphatic flow, because the inward and outward non-lymphatic flows due to hydro-oncotic forces balanced each other and the net non-lymphatic flow was essentially zero. When the protein concentration was 6 g per 100 ml or more, the decrease of pleural fluid was remarkably attenuated, because of an increase of the inward non-lymphatic flow due to an increase of protein concentration in the pleural fluid. Even in these situations, the lymphatic flow was kept at the rate of 0.20 ml/kg/hour, similarly with the former conditions. From these results, it may be concluded that the lymphatic system is a principal factor, and that factors governed by hydro-oncotic elements have additive effect to that of the lymphatic system in the regulation of pleural fluid dynamics.

Combination therapy using 9,3"-di-O-acetyl-midecamycin with beta-lactam antibiotics against Pseudomonas aeruginosa infection.

The combined effect of beta-lactam antibiotics and 9,3"-di-O-acetylmidecamycin were examined in 10 patients diagnosed as having chronic respiratory tract infections and complicated urinary tract infections. Patients were initially treated with 3 to 10 g/day of beta-lactams as intravenous infusions or by oral route for a certain period, after which period they were treated concurrently with 0.3 to 0.9 g/day of 9,3"-di-O-acetylmidecamycin by oral route. Through this combination therapy eradication of P. aeruginosa was obtained in 2 cases and remarkable decrease in another 2 cases out of the 10 cases. This combination therapy was judged to be effective in these 4 cases. Of the remaining cases, P. aeruginosa was eliminated before the start of the combination therapy in 3 cases, and no change in bacterial count was noted after the combination therapy in the other 3 cases.

Airway obstruction in achalasia.

A man with a mediastinal mass shadow detected on chest roentgenogram was diagnosed as having achalasia by barium swallowing study. Megaesophagus caused by achalasia produced airway obstruction. Maximum expiratory-inspiratory flow volume curve identified that peak flow rate decreased in early expiration, and the ratio of expired to inspired flow at 50 percent of vital capacity was below 0.9, aggravated by 250 ml water intake. Deformation of the tracheal shape was proved easily.

Glutamate decarboxylase from barley embryos and roots. General properties and the occurrence of three enzymic forms.

Glutamate decarboxylase in extracts of barley has a Km value for L-glutamate of 22 mM and is activated by the addition of pyridoxal phosphate by up to 3.5 times. Sucrose-density-gradient experiments indicate the presence of two enzyme forms with molecular weights 256000 and 120000. The lower-molecular-weight form appears to be relatively inactive and spontaneously associates to the higher-molecular-weight form on storage. The enzyme is inhibited by thiol reagents and the distribution of activity on density gradients is altered in favour of the lower-molecular-weight form by the presence of 2-mercaptoethanol. After removal of the 2-mercaptoethanol spontaneous association to the higher-molecular-weight form occurs. The presence of oxygen in the extraction buffer and in the water during imbibition leads to a relative increase in the higher-molecular-weight form compared with situations where oxygen is excluded. In contrast, glutamate decarboxylase in extracts of 3-day-old barley roots has a Km value for L-glutamate of 3.1 mM and is activated up to 10% by addition of pyridoxal phosphate. The root enzyme occurs as a single species with molecular weight 310000 and this is unaffected by 2-mercaptoethanol although thiol reagents do act as weak inhibitors. The molecular weight is also unaffected by the presence or absence of oxygen in the extraction buffers.
