Liver lesions of visceral larva migrans due to Ascaris suum infection: CT findings.

The purpose of this study was to analyze computed tomographic (CT) findings of hepatic lesions due to Ascaris suum infection. CT of the liver in three patients, all of whom had immunoserologically confirmed A. suum infection, were retrospectively reviewed. Twenty-five lesions were identified in total. Two radiologists analyzed CT findings in a consensus fashion, with particular interest in the margin, shape, and location of the lesions. Hepatic lesions were ill-defined (22 of 25), small (3-35 mm; average, 11 mm), and nodular (18 of 25) or wedge (three of 25) in shape. Most were located in periportal (16 of 25) or subcapsular (six of 25) regions. Hepatic nodules due to visceral larva migrans of A. suum were located mainly in periportal or subcapsular regions, which may represent periportal eosinophilic granuloma, its pathologic feature. The results were considered to represent the pathophysiology of this entity.

Expression of the c-myc and c-fos genes as a potential indicator of late radiation damage to the kidney.

PURPOSE: To evaluate proliferative response according to c-myc and c-fos gene expression as an early indicator of late radiation damage to the kidney. METHODS AND MATERIALS: c-myc and c-fos gene expression was measured in the irradiated kidney of C3H/HeSlc mice using reverse transcriptase-polymerase chain reaction (RT-PCR) 24 h after 9, 12, and 15 Gy as well as 2 and 7 days after 15 Gy. In a second experiment, 12 Gy was administered to the right kidney plus the lower half of left kidney. The mice underwent left nephrectomy 24 h after, and the correlation between increased gene expression and elevation of the blood urea nitrogen (BUN) level representing late functional damage was investigated. RESULTS: c-myc and c-fos expression increased in 1 of 10 controls and in 8 of 10 and 7 of 10 mice that received 15 Gy, respectively. Expression increased dose-dependently from 9 to 15 Gy. In mice having an increased BUN level, 7 of 10 and 8 of 10 were positive for c-myc and c-fos, respectively. All 6 mice having an unchanged BUN level were negative for c-fos, while 5 were negative for c-myc. CONCLUSION: Measurement of c-myc and c-fos gene expression is potentially an early indicator of late radiation damage to the kidney.

Effects of aging on muscle T2 relaxation time: difference between fast- and slow-twitch muscles.

RATIONALE AND OBJECTIVES: To determine whether the T2 relaxation time of skeletal muscle is affected by aging and to compare the effects of aging between fast- and slow-twitch muscles in a human study. To investigate the mechanisms of age-related changes in T2 relaxation time in an animal (mouse) study. METHODS: T2 relaxation times of the soleus (slow-twitch, rich in type I fiber) and gastrocnemius (fast-twitch, rich in type II fiber) muscles were examined in 59 healthy human subjects, 22 to 76 years of age, by clinical magnetic resonance imaging. In mice, T2 relaxation times, fat ratios, and extracellular space ratios (extracellular space/intracellular plus extracellular space) of the spinalis (fast-twitch, rich in type II fiber) muscles were also examined (group of 7 old mice, 24-26 months; group of 7 young mice, 8-10 weeks). RESULTS: In the human study, the T2 relaxation time of the gastrocnemius muscle increased significantly with aging (r = 0.53, P < 0.01) while that of the soleus muscle did not. In the animal study, the T2 relaxation time of the spinalis muscle was significantly longer (P < 0.05) and the extracellular space ratio of the spinalis muscle significantly wider (P < 0.01) in old than in young mice. No significant difference in fat ratio was observed between old and young mice. A significant, positive correlation was seen between the extracellular space ratio and T2 relaxation time (r = 0.84, P < 0.01). CONCLUSIONS: The T2 relaxation time of fast-twitch muscle increases with aging, due mainly to increased extracellular space, reflecting age-related type II fiber atrophy.

Radicicol binding to Swo1/Hsp90 and inhibition of growth of specific temperature-sensitive cell cycle mutants of fission yeast.

A panel screening using cdc mutants of Schizosaccharomyces pombe identified radicicol as a potent growth inhibitor of certain mutants at the permissive temperature. The strains sensitive to radicicol were cdc7, cdc11, and cdc14, all of which are defective in early septum formation. Cytokinesis but not nuclear division of these mutants was inhibited by radicicol, but that of cells with the wild-type background was not. A biologically active derivative of radicicol with a biotin moiety at the C-11 position bound Swo1, an Hsp90 homologue in S. pombe. Increased Swo1 expression partially suppressed radicicol sensitivity of cdc14 and almost completely rescued morphological abnormalities in cdc14 and cdc7 cells induced by radicicol at the permissive temperature. On the other hand, the increased Swo1 expression did not restore septum formation at the nonpermissive temperature. These results suggest that Swo1, as a molecular chaperone, plays a role in stabilizing these temperature-sensitive proteins at the permissive temperature or in activating the cytokinesis signaling cascade.

Radicicol binds and inhibits mammalian ATP citrate lyase.

Six different biotinylated radicicol derivatives were synthesized as affinity probes for identification of cellular radicicol-binding proteins. Derivatives biotinylated at the C-17 (BR-1) and C-11 (BR-6) positions retained the activity of morphological reversion in v-src-transformed 3Y1 fibroblasts. Two radicicol-binding proteins, 120 and 90-kDa in size, were detected in HeLa cell extracts by employing BR-1 and BR-6, respectively. The 90-kDa protein bound to BR-6 was identified to be Hsp90 by immunoblotting. The 120-kDa protein bound to BR-1 was purified from rabbit reticulocyte lysate, and its internal amino acid sequence was identical to that of human and rat ATP citrate lyase. The identity of the 120-kDa protein as ATP citrate lyase was confirmed by immunoblotting. Interaction between BR-1 and ATP citrate lyase was blocked by radicicol but not by herbimycin A that interacts with Hsp90. These results suggest that radicicol binds the two proteins through different molecular portions of its structure. BR-1-bound ATP citrate lyase isolated from rabbit reticulocyte lysate showed no enzymatic activity. The activity of rat liver ATP citrate lyase was inhibited by radicicol and BR-1 but not by BR-6. Kinetic analysis demonstrated that radicicol was a non-competitive inhibitor of ATP citrate lyase with K(i) values for citrate and ATP of 13 and 7 microm, respectively.

Primary non-Hodgkin's lymphoma of the lacrimal sac: a case report and a review of the literature.

BACKGROUND: Primary non-Hodgkin's lymphoma of the lacrimal sac is extremely rare, with most reported cases representing secondary involvement of a systemic malignancy. METHODS: The clinical record of a 70-year-old female who presented with epiphora and swelling of the lacrimal sac area is described. A review of the literature of patients with primary lacrimal sac lymphoma also is presented. RESULTS: Computed tomography demonstrated a lacrimal sac mass involving the nasolacrimal canal. Histopathologic examination of a biopsy specimen revealed a diffuse large cell lymphoma. Using immunohistologic staining, the tumor was a B-cell type, similar to those described in the literature. The patient was treated with 50 gray of irradiation and chemotherapy with cyclophosphamide, doxorubicin, vincristine, and prednisone. The patient remained free of lymphoma 26 months after initial treatment. An ocular examination was unremarkable except for epiphora. CONCLUSIONS: Radiotherapy and/or chemotherapy can treat localized lymphoma of the lacrimal sac successfully.

Cororubicin, a new anthracycline antibiotic generating active oxygen in tumor cells.

In the course of our screening for antitumor antibiotics which show biological activities by generating active oxygen in tumor cells. Micromonospora sp. JY16 was found to produce a new anthracycline antibiotic designated cororubicin. The structure of cororubicin was elucidated as shown in Fig. 5 on the basis of NMR spectral analysis. Cororubicin generated superoxide radicals in tumor cells and showed cytotoxicity, which was reduced by addition of an antioxidative agent, dithiothreitol (DTT).

[The policy and role of government in community dental health care in the twenty-first century].

The level of dental treatment in Japan is highly rated in the world. However, Japanese prevalence of dental caries in children is relatively high for industrialized countries. The purpose of this paper is to suggest some strategies for community-based dental health promotion in Japan for the twenty-first century, based on a review of the results of an evaluation in Niigata prefecture. The suggestions are as follows; 1. Legislation shoulde be adopted for community-based dental health promotion. 2. Adoption of fluoride usage as an effective preventive method for dental caries should be given priority. 3. Primary dental health services should be provided by municipalities to its inhabitants, and the role for such services by the health center and prefectural government should be clearly defined. 4. Adequate staffing-dental personnel, dentists and dental hygienists-should be secured by municipal governments. 5. A system for evaluation of efficacy of preventive services should be established.

Basic studies on visible light-cured resin as a denture base. Part 17. Transverse and tensile strengths of repaired denture base resin using a trial repair resin.

Trial production of a visible light-cured repair resin (powder/liquid type, TPL) as a denture base resin was carried out using cyclophosphazene monomer, and then the transverse and tensile strengths of the repaired specimens were examined. As a control, Triad Gel (TRI) was used. In the transverse strength test, TPL showed values of 507-470 kgf/cm2, which were about double those of TRI. As for tensile strength, TPL showed values of 329-268 kgf/cm2, and higher values were obtained in comparison with TRI, especially after 30 days of water immersion, when the values were doubled. The durability of TPL was favorable in comparison with TRI.

Basic studies on visible light-curing resin for denture base. Part 8. The hardening depth of denture relining material after penetration of light through the denture base resin.

A study was performed to examine how the duration of irradiation at the time of relining with visible light-curing material influences the depth of hardening of the relining material, in relation to the thickness of the denture base placed between the denture and the relining material. It was found that for a range of denture base thickness of 1-2.5 mm, a desirable degree of hardening could be obtained using an irradiation time of 20 s for relining material with a thickness of up to 4 mm used for the basal surface.

[Effectiveness of multidisciplinary treatment combined with immunotherapy in esophageal cancer--analysis of 287 cases of carcinoma of the thoracic part of the esophagus in Kyushu and Yamaguchi districts].

The effectiveness of surgical adjuvant immunochemotherapy was studied on 287 patients with thoracic esophageal cancer in the Kyushu and Yamaguchi districts. Pre- and postoperative subcutaneous administrations of BCG, 10 mg once a week and an oral administration of PSK, 3 g a day, for as long as possible after surgery were instituted. The control group was designated as the one which neither PSK nor BCG was given. Most patients received chemotherapy and/or radiotherapy. Significant elongation of survival time was observed in immunotherapy patients with stage III and in those with non-curative resection, both having no lymph node metastasis.
