Immunohistochemical detection of biotin-labeled murine monoclonal antibody in nude mice transplanted with human colon cancer.

The tissue localization of biotin-labeled murine monoclonal antibody (MAb) A7 directed against human colonic cancer was examined in tumor-bearing nude mice following intravenous administration. MAb A7 did not react with any murine visceral organs. At 24 hr after biotin-labeled A7 injection, the tumor was stained by the avidin-biotin peroxidase complex method, but no staining was observed after injection of biotin-labeled normal mouse Ig G in the control group. Biotin-labeled MAb was found to be suitable for the detection of the xenografted tumor of nude mice. Our purpose was to provide a new method concerning the distribution of biotin-labeled MAb in vivo.

Gene amplification and overexpression of EGF receptor in squamous cell carcinomas of the head and neck.

Tumours of the head and neck were examined for gene amplification and expression of the epidermal growth factor (EGF) receptor by Southern blot and Western blot analyses. The EGF receptor gene was found to be amplified in four (19%) of 21 squamous cell carcinomas. The EGF receptor was overexpressed in eight (53%) of 15 squamous cell carcinomas examined, including all four tumours showing gene amplification. No amplification or overexpression of the EGF receptor gene was detected in any of nine malignant or eight benign tumours of other types of the head and neck. The tumours showing amplification and/or overexpression of the EGF receptor gene (8/15) were all identified histologically as well differentiated squamous cell carcinomas, whereas none of the histologically less differentiated squamous cell carcinomas (0/9) showed amplification and/or overexpression of the EGF receptor gene. Within our sample set, no correlation was evident between amplification and/or overexpression and the clinical stage or tumour site. Our results support the possible involvement of gene amplification and overexpression of the EGF receptor in a subclass of squamous cell carcinomas of the head and neck.

Lexical agraphia in the Japanese language. Pure agraphia for Kanji due to left posteroinferior temporal lesions.

A new syndrome of 'pure agraphia for Kanji' is described in 3 Japanese subjects with lesions in the left posteroinferior temporal region. Kanji (ideogram or morphogram) can be compared with orthographically irregular or ambiguous words in some European languages, since it is impossible to write Kanji characters unless each one of them is learned and memorized. In contrast, Kana (phonogram or syllabogram) words are comparable with orthographically regular words or nonsense words, because the Kana writing system depends on strict phonological rules (almost one-to-one correspondence between syllable and syllabogram). We conclude that 'lexical agraphia' reported in European languages can also be observed in the Japanese language where it is expressed as 'pure agraphia for Kanji'. 'Lexical agraphia' is a useful concept with general application regardless of language system.

Lichenoid drug eruptions due to nandrolone furylpropionate (Demelon).

A case of lichenoid drug eruption due to nandrolone furylpropionate (Demelon) is reported. A 71-year-old man with aplastic anemia developed widespread erythema and reticulation with violaceous papules and pigmentation after receiving intramuscular injections of Demelon weekly for 2 months. The eruption gradually resolved after drug withdrawal, leaving reticular pigmentation. A biopsy specimen of the skin lesion showed lichenoid-type reactions including hydropic or colloid degeneration of the basal cells, sometimes with satellite necrosis, and a band-like subepidermal lymphocytic infiltrate. Most of the mononuclear cells stained strongly for helper/inducer T lymphocytes (Leu 3a).

Transcription mapping of glycoprotein I (gpI) and gpIV of varicella-zoster virus and immunological analysis of the gpI produced in cells infected with the recombinant vaccinia virus.

In order to determine the transcripts of gpI and gpIV of varicella-zoster virus (VZV), RNA was isolated from human embryonic fibroblast cells infected with VZV and Northern blot analysis was carried out using cloned DNA probes of unique short region including gpI and gpIV genes. The analysis of RNA revealed two discrete transcripts of 3.6 and 2.15 kilobases (kb) and three transcripts of 3.6, 2.9, and 1.6 kb which hybridized to DNA probes covering the gpI and gpIV region, respectively. Next, mRNAs were hybrid-selected, translated in vitro and the polypeptide products were immunoprecipitated with antibodies against these glycoproteins. The polypeptides with a molecular weight of 70,000 (70K) and 37K which were in vitro translational products of mRNA hybrid-selected with the DNA clone covering gpI and gpIV were detected using antibodies against gpI and gpIV, respectively. The result showed that the 70K polypeptide is presumably the translational product of 2.15 kb mRNA and the 37K polypeptide is that of 1.6 kb mRNA. DNA fragment encoding gpI or gpIV was inserted into vaccinia virus DNA and the recombinant viruses, mO74 (gpI) and mO39 (gpIV), were used for immunological analysis. In consequence, the gpI derived from cells infected with mO74 showed antigenic characteristics similar to those of gpI from VZV-infected cells as determined from the immunoprecipitation pattern, although the molecular weight of each polypeptide was different, and antibody produced in rabbits infected with recombinant virus had a high neutralizing activity, when the reaction was performed with complement. This suggested that gpI plays an important role for protection and recovery from VZV infection.

Effect of a bacteriocin-producing strain of Streptococcus sobrinus on infection and establishment of Streptococcus mutans on tooth surfaces in rats.

The effect of bacteriocin produced by Streptococcus sobrinus MT6223 on infection and establishment of Streptococcus mutans MT6222 was studied in specific pathogen-free rats. These strains were isolated from a carious lesion of a single subject. S. mutans MT6222 was found to be susceptible to the growth inhibitory action of S. sobrinus MT6223. When simultaneously inoculated into the oral cavity of rats, even a small inoculum (10(5) CFU) of S. sobrinus MT6223 completely inhibited colonization of S. mutans MT6222 on the tooth surface. Also, S. sobrinus MT6223 eliminated S. mutans MT6222 when MT6223 (10(8) CFU) was inoculated 2 days after the inoculation of 10(8) CFU cells of MT6222. Similar results were obtained in dental plaque samples from the tooth surface and the fissures of the upper molars at the end of the experiment. However, when S. sobrinus MT6223 (10(8) CFU) was inoculated 2 weeks after the inoculation of S. mutans MT6222 (10(8) CFU), MT6223 coexisted with MT6222. However, the plaque samples showed that MT6223 inhibited the establishment of MT6222 on smooth surfaces, but not in fissures. In addition, MT6223 protected against subsequent infection with MT6222. However, a nonbacteriocinogenic mutant of S. sobrinus MT6223 did not inhibit the infection and establishment of S. mutans MT6222.

Transient myocardial thickening in acute myocarditis--serial study by two-dimensional echocardiography.

A 13-year-old girl developed viral myocarditis complicated by pulmonary thromboembolism. Marked dilatation and reduction in the contractility of the left ventricle were noted in a two-dimensional echocardiogram on admission. The thicknesses of the left ventricle and the interventricular septum were both 8 mm at this time, but increased after the 9th day of the illness to reach 2 cm by the 15th day, with a concomitant reduction in the left ventricular cavity. The myocardial thickening gradually decreased and the ejection fraction improved on serial echocardiographic evaluations, and the wall thickness and ejection fraction returned to normal by the 32nd day. The myocarditis resolved without sequelae. The implications of this transient myocardial thickening are discussed.

Adenocarcinoma in Meckel's diverticulum: case report and literature review.

A case of adenocarcinoma arising in Meckel's diverticulum in a 55 year old man is reported, and a brief review of the literature is presented. The patient developed low abdominal pain and showed elevation of serum carcinoembryonic antigen (CEA) level. The tumour was located in the apical portion of the diverticulum and extended into the mesenterium. Histologically, the tumour was a well-differentiated adenocarcinoma arising in the Meckel's diverticulum. Immunohistochemical study showed that malignant cells were positive for CEA. The noteworthy feature of this case is the pre-operative elevation of serum CEA level and the immunohistochemical demonstration of CEA in the cancer cells.

Two types of suppressor T cells that inhibit delayed-type hypersensitivity to Mycobacterium intracellulare in mice.

Mycobacterium intracellulare Mino grows progressively in the organs of C57BL/6 mice. When C57BL/6 mice were injected with strain Mino, very weak or almost no delayed-type hypersensitivity (DTH) to the specific antigen, M. intracellulare purified protein derivative, was observed. The DTH response was enhanced by cyclophosphamide pretreatment, indicating that some suppression mechanism(s) might be involved in DTH. In cell transfer experiments it was shown that two types of suppressor T cells, afferent type (TS-Aff) and efferent type (TS-Eff), are induced in the spleens of Mino-infected mice. TS-Aff were detected from the early stage to the late stage of infection. TS-Eff were detected only in the late stage of infection. These two types of suppressor T cells were both Thy-1 positive and Lyt-2 positive; however, TS-Aff seemed to be I-J positive, and TS-Eff seemed to be I-J negative. Cross-reactivity in suppression was observed between Mino and Mycobacterium bovis BCG but not between Mino and sheep erythrocytes. DTH to Mino and BCG was not cross-reactive. The possible role of suppressor T cells in M. intracellulare infection is discussed.

Prostaglandins augment muscarinic responses in the rabbit cecum.

Effects of prostaglandin E2, F2 alpha, and synthetic thromboxane A2 (PGE2, PGF2 alpha, and STA2, respectively) on electrical and mechanical responses of the rabbit cecum were investigated. Transmural electrical stimulation evoked an excitatory junction potential (EJP) and contraction, events that were inhibited by 1 microM atropine or 0.3 microM tetrodotoxin. Indomethacin (up to 30 microM) modified neither the membrane potential nor the muscle tone but did inhibit amplitudes of the EJP and the twitch contraction. In the presence of 30 microM indomethacin, PGE2 (below 1 nM) had no effect on the membrane potential or muscle tone, while PGE2 (above 10 nM) contracted the cecal tissues, without depolarization. PGE2 enhanced the twitch contraction and restored the EJP in the presence of 30 microM indomethacin. Acetylcholine (0.1-3 microM) depolarized the membrane, but in the presence of 30 microM indomethacin, this depolarization was inhibited. PGE2 (0.1-100 nM) prevented these inhibitory actions of indomethacin. PGF2 alpha (0.1-100 nM) had weaker actions than PGE2 while STA2 (0.1-100 nM) had no effect on muscarinic responses. Thus muscarinic responses are augmented by primary prostaglandins.

Effect of uridine 5'-diphosphate on cryogenic brain edema in rabbits.

This study was undertaken to examine the effect of uridine 5'-diphosphate, administered intravenously or intraperitoneally, on cold injury-induced brain edema in rabbits. Bolus injection or continuous intravenous infusion of uridine 5'-diphosphate 26 hours after a lesion was established had adverse effects, such as increased intracranial pressure and lowered systolic arterial blood pressure and cerebral perfusion pressure for approximately 10-29 minutes, but these parameters did not change appreciably from 29 minutes to 3 hours after administration. Intraperitoneally administered uridine 5'-diphosphate did not affect these parameters appreciably during 3 hours. Thus, the intravenous administration of uridine 5'-diphosphate is harmful under neurosurgical conditions. In contrast, 10 mg/kg/day i.p. uridine 5'-diphosphate pretreatment and posttreatment, beginning 24 hours before and continuing until 24 hours after the insult, significantly reduced neurologic abnormalities, Evans blue extravasation, water content in the injured gray matter, and intracranial pressure without affecting water content in the white matter. Intravenous dexamethasone pretreatment and posttreatment in this setting significantly reduced only neurologic abnormalities. However, there were no significant differences between intraperitoneal uridine 5'-diphosphate and intravenous dexamethasone effects on cold-injured brain.

Crystal structure and molecular conformation of E-64, a cysteine protease inhibitor.

In order to elucidate the conformational characteristics of cysteine protease inhibitors contributing to their inhibitory activities, the conformation of E-64 (N-[N-(L-3-trans-carboxyoxiran-2-carbonyl)-L-leucyl]-agmatine), a potent inhibitor of papain, was determined by X-ray crystal structure analysis. The molecules were packed in the crystal through electrostatic forces and hydrogen bonding between the oppositely charged terminal groups and between the amide groups. Two crystallographically independent E-64 molecules both took a flattened and slightly curved structure, which is similar to that of loxistatin, a related cysteine protease inhibitor. Based on the present results, a possible inhibitory mechanism of E-64 is proposed, with reference to the binding mode observed in the crystal structure of papain-substrate analogue complex.

[Suppressor T cells against delayed type hypersensitivity to Mycobacterium intracellulare].

Mycobacterium intracellulare Mino strain (Mino) grows progressively in the organs of susceptible mice, such as C57BL/6, C57BL/10 or BALB/c. It is very difficult to induce acquired immunity against M. intracellulare in those susceptible mice. C57BL/6 (B6) mice show no or very weak delayed type hypersensitivity (DTH) to partially purified Mino antigens when sensitized with 10(7) living Mino subcutaneously. B6 mice pretreated with cyclophosphamide (CY) showed enhanced DTH to Mino, suggesting that suppressor mechanism exists in this system. Whether or not such a suppressor mechanism exists in the induction phase of DTH (A) and/or in the expression phase of DTH (B) was examined by cell transfer experiments. Recipients for testing (A) were B6 mice receiving CY 2 days before an immunization, and those for (B) were B6 mice receiving both CY and subcutaneous injections of Mino 3 weeks before the cell transfer. B6 mice which were intravenously injected with Mino a week or 6 weeks before preparing spleen cell suspension, were used as the donors. Single cell suspensions of the spleens were plated on tissue culture dishes and non-adherent cells were harvested after incubating for 60 min at 37 degrees C. These cells were used for cell transfer. In the induction phase experiment, recipient mice received the cells before sensitization, while they received the cells after sensitization in the expression phase experiment. DTH were markedly suppressed in the cell transferred groups as compared with the non-transferred groups. This activity disappeared by treatment of the transferred cells with antibody (alpha Thy-1 Ab) and complement.(ABSTRACT TRUNCATED AT 250 WORDS)

Immunochemical characterization of the antigen recognized by the murine monoclonal antibody A7 against human colorectal cancer.

The nature of the antigen recognized by the murine monoclonal antibody A7 (Mab A7) against human colorectal carcinoma was investigated using immunochemical and biochemical techniques. Binding activity of 125I-labeled Mab A7 was examined using various human cancer cell lines. Mab A7 gave the highly specific binding to colon cancer cell lines, SW1116 and WiDr, and gave only a very weak or no reactivity to gastric cancer cell lines, pancreas cell lines or lung cancer cell lines. SDS-polyacrylamide gel electrophoresis (SDS-PAGE) and immunoblotting of the extractable antigen from SW1116 showed a single band at approximately 45,000 dalton formed by 125I-labeled Mab A7. Treatment of SW1116 with sodium periodate, pronase and ficin resulted in the loss of antigenic activity. These data strongly suggest that the antigen recognized by Mab A7 is composed of glycoprotein. Competitive binding analysis to the surface of the colon cancer cell line using polyclonal anti-CEA and Mab A7 as well as immunoblotting analysis using monoclonal anti-CEA and Mab A7 suggested that the antigen recognized by Mab A7 was different from CEA. Moreover, this antigen was also found in surgical specimens of colorectal cancer patients and its molecular property was identical to the antigen extracted from SW1116.
