[A new technique for collection the cerebrospinal fluid from rat (authors transl)].

An attempt was made to collect the cerebrospinal fluid (CSF) of rat using a capillary phenomenon technique. Experiments were carried out in 76 rats anesthetized with pentobarbital sodium (Nembutal, Abbot. 50 mg/kg IP). The midline incision in the occipital region was made from 0.5 cm anterior to interauricular line of 4 cm in length. Exposing the atlanto-occipital dura mater by separating the nuchal muscles, the CSF was collected with a heparinized microtube (MICROPET. Bection, Dickson and Company) through a small hole made by a 18G syringe needle. Thus, 0.05-0.05 ml of the CSF could be readily obtained without mixture of the blood in 64 cases out of 76 trials, 84%. This simple technique is a useful way to get a small amount of CSF from little animals, such as rats.

[A case with bilateral internal carotid and left vertebral artery occlusion (author's transl)].

Since Moniz reported the obstruction of the extracranial internal carotid artery in 1937, there have been many reports of extracranial arterial occlusion. The authors experienced a case of 40-year-old woman who had minor neurological deficit such as attack of black out, disturbance of the bilateral visual acuity, concentric narrowing of the bilateral field and disturbance of the bilateral auditory acuity, in spite of complete obstruction of both internal carotid arteries and the left vertebral artery, and possible obstruction of the right vertebral artery. The angiograms showed well developed collateral circulation. The cause of the obstruction was considered to be angitis following otitis media and meningitis in her childhood. The symptoms were alleviated by drugtherapy.

A case of intracranial osteochondroma (author's transl).

The authors reported a case of intracranial osteochondroma. A 52-year-old woman was admitted to Tokyo Women's Medical College hospital with seven years history of right visual disturbance and frontal heavy feeling. Neurological examination revealed merely right optic nerve impairment. A large mottled calcification in X-ray craniogram and a localized homogenous high density area illuminated by EMI scan in the right parasellar region were characteristic in diagnosis of osteochondroma. On April 21, 1976 a right fronto-temporal craniotomy was performed and the tumor tissue was removed partially. The pathological examination confirmed the diagnosis of osteochondroma (ossifying chodroma).

[Growth hormone, prolactin, LH and FSH secretion in tissue culture of pituitary adenomas (author's transl)].

We have performed tissue culture of pituitary adenomas (5 acromegalies, 7 non-acromegalies) and measured GH, PRL, LH and FSH in the media of tissue culture by radioimmunoassay (RIA) for 30-40 days. In addition, GH, PRL, LH and FSH were measured in the plasma and tumor tissue by RIA. GH concentration in the culture media was markedly high in all cases with acromegaly and high also in 2 out of 7 non-acromegaly cases (case 6 and 7). GH concentration in the media rapidly decreased almost as a straight line on a semilogarithmic scale until the 30-40 in day, when GH level became less than 10 ng/ml. PRL concentration in the culture media was high in 5 patients (3 acromegalies, 2 non-acromegalies). PRL concentration regressed more slowly than GH, but still remained higher than 10 ng/ml in all of the patients on the 25-40th day when the studies were discontinued. LH and FSH concentrations in the culture media was high in two patients. One patient was a pituitary adenoma secreting LH and FSH, and another case was a false high secreting one, possibly contaminated with gonadotropin in normal pituitary gland at surgical operation. LH and FSH concentrations rapidly decreased as that of GH. A rare case of primary LH and FSH secreting pituitary adenoma was demonstrated.

Purification and partial characterization of two glycoproteins in bovine peripheral nerve myelin membrane.

Two major glycoproteins of bovine peripheral nerve myelin were isolated from the acid-insoluble residue of the myelin by a procedure involving delipidation with chloroform/methanol (2:1, v/v) and chromatography on Sephadex G-200 column with a buffer containing sodium dodecyl sulfate. The separation patterns of the proteins on the gel were affected considerably by the dodecyl sulfate concentration in the elution buffer. At above 2% dodecyl sulfate concentration in the elution buffer, the glycoproteins, could be separated clearly on the gel and were purified. The purified proteins, the BR protein (mol. wt. 28 000) and the PAS-II protein (mol. wt. 13 000), were homogeneous on dodecyl sulfate-polyacrylamide gel electrophoresis. The NH2- terminal amino acids of the BR and the PAS-II proteins were isoleucine and methionine, respectively. The BR protein contained glucosamine, mannose, galactose, fucose and sialic acids and the PAS-II protein contained glucosamine, mannose, galactose, fucose and glucose. Neither the BR protein nor the PAS-II were a glycosylated derivative of a basic protein of bovine peripheral nerve myelin, a deduction based on the results of amino acid analysis. The two major glycoproteins were observed commonly in the peripheral nerve myelin of cows, pigs, rabbits and guinea pigs, using dodecyl sulfate-polyacrylamide gel electrophoresis.

3H-thymidine autoradiography of the CSF cells in cases of non-neoplastic disease.

Human CSF cells in cases of non-neoplastic disease of the central nervous system (CNS) were examined in vitro by 3H-thymidine autoradiography. Immediately after withdrawl by lumbar or ventricular puncture, the CSF was incubated in a sedimentation chamber at 37 degrees C for 1 hr with an admixture of 3H-thymidine at a concentration of 1-2 muCi/ml CSF. In a few cases the CSF withdrawn was incubated in a glass tube in the same condition as in a sedimentation chamber, and the CSF cells were collected by centrifugation. The CSF cells collected were fixed in methanol and the microautoradiographic procedure was performed. Labeled CSF cells were found in 21 cases out of 22. The average labeling index of the total nucleated cells was 0.22% with the highest labeling of 0.74%. Almost all the labeled cells were thought to be medium to large sized lymphocytes and monocytoids. Peripheral blood was examined by a similar method and the results were compared with those of the CSF. It may be noteworthy that thre exist DNA synthesizing cells in the CSF even in a non-neoplastic state of the CNS, although the number is not large.

A morphological study on vascular changes in the surgically resected tissues of temporal lobe epilepsy-with a special reference to the etiology.

(1) Twenty-one out of 23 cases surgically resected specimens of TLE presented the common histological findings; sclerotic changes of the tissues. And the other two cases encountered the small tumors which might be called "cryptic". (2) Since circulatory disturbance is generally accepted to explain the cause of TLE we reexamined vascular behaviors in detail and found the quantitative and qualitative abnormaltities which reached the high rate of 71% (17 out of 21 cases) except two cases of small tumors. Moreover, it is remarkably noteworthy that these vascular changes adjoin the sclerosed parenchymal tissue. (3) It may be said that vascular changes have exerted some influences on the formation of inferomedial temporal sclerosis from the standpoint of the histological findings, though they are alone not fully sufficient to explain the etiology of TLE.

Correlation between myocardial and plasma concentration of digitoxin and its metabolites with special reference to digitoxin radio-immunoassay.

According to our prevous study, the metabolites of digitoxin show nearly dentical affinity with digitoxin antibody. The present study reveals relatively constant ratio of heart to plasma in total 3H-counts, and the concentration of metabolites are ver similar between the two throughout the time investigated which indicates digitoxin concentration of plasma as determined by radioimmunoassay reflects well that of myocardium, although it may contain metabolites.

A female case of the HCG-producing ectopic pinealoma associated with precocious puberty.

A female case of precocious puberty associated with HCG-producing ectopic pinealoma was reported. The patient, a 5-year-old girl, was referred to the hospital because of headache and choked discs. Physical examination revealed normal physical growth with breast enlargement. Endocrinological study revealed a high plasma HCG concentration of 1192 ng/ml with a normal FSH level. None of HCG, LH and FSH did respond to the LH-RH test. A partial resection of the tumor and an external X-ray irradiation relieved the symptoms and breast enlargement subsided with a remarkable decrease in the plasma HCG level. Histological examination revealed two-cell-pattern pinealoma and electron microscopic findings showed abundant secretory granules in the dark cells. HCG content in the tumor was as high was 400 ng/mg of acetone dried tumor tissue, but no FSH was detectable. Hitherto, all of the reported cases of precocious puberty associated with pineal tumors have been exclusively boys. A normal level of plasma FSH concentration with a somewhat elevated prolactin level might be a contributory factor for the development of precocial sexual development in the present case.

Prostaglandin action on the main pulmonary artery and portal vein of the rabbit.

The effects of prostaglandins on electrical and mechanical properties of the rabbit pulmonary artery and portal vein were investigated and compared with those recorded from the guinea pig. In the pulmonary artery, PGE1 and PGE2 (10(-9)-10(-6) g/ml) had no effect on mechanical properties, but PGF2alpha (10(-8) g/ml) produced tonic contraction, while in the portal vein, PGE1 and PGE2 caused relaxation of the tissues. When the tissue of the pulmonary artery contracted from pretreatment with noradrenaline (10(-7) g/ml), PGE1 and PGE2 experienced partial, incomplete, relaxation, while PGF2alpha enhanced the mechanical response produced by noradrenaline. At concentrations of 10(-7) g/ml, PGE1 and PGE2 hyperpolarized the membranes in both smooth muscle tissues while PGF2alpha depolarized them. These agents decreased membrane resistance at all membrane potential levels to values below those measured in Krebs solution. PGF2alpha (10(-6) g/ml) applied simultaneously with procaine (1.4-2.7 x 10(-3) g/ml) markedly enhanced the mechanical responses of both tissues beyond the effects evoked by PGF2alpha alone. However, noradrenaline (10(-8) and 10(-7) g/ml) applied simultaneously with procaine (1.4 x 10(-3) g/ml) markedly suppressed the mechanical responses evoked by noradrenaline alone. It is concluded that PGE1 and PGE2 brought about relaxation of both tissues (vasodilation), while PGF2alpha brought about contraction (vasoconstriction). The excitatory action of PGF2alpha (10(-5) g/ml) and noradrenaline (10(-7) g/ml) on the electrical and mechanical responses appear as the same phenomena, i.e., depolarized membrane, decreased membrane resistance and contraction, although the mechanisms producing the above phenomena differ.
