Specific phosphorylation of pig liver initiation factor eIF-2 by the N-ethylmaleimide-treated hemin-controlled translational inhibitor.

The specific phosphorylation of pig liver initiation factor 2(eIF-2) by the N-ethylmaleimide (NEM)-treated hemin-controlled translational inhibitor (HCI) from rabbit reticulocytes was investigated. The inhibitor phosphorylated the serine residue of the alpha subunit of eIF-2 (eIF-2 alpha) and 1 mol of phosphate was incorporated into 1 mol of eIF-2 alpha by the inhibitor on maximal phosphorylation, even when eIF-2 was pretreated with alkaline phosphatase prior to phosphorylation. The 32P-labeled eIF-2 alpha was subjected to tryptic digestion and the tryptic digest was analyzed by two-dimensional peptide mapping on a cellulose thin-layer sheet. After 94 h digestion, the autoradiograph of the peptide map showed a single 32P-labeled band with a molecular weight of approximately 1,200. These findings suggest that one specific serine residue of pig liver eIF-2 alpha was phosphorylated by the NEM-treated HCI.

[Case of histopathological findings of choriocarcinoma in stomach cancer].

A case of gastric cancer which contained a part of choriocarcinoma was reported in this study. The patient was a 64-year-old man, who was operated on with the diagnosis of gastric cancer in the prepyloric region. The postoperative histological examination revealed that typical choriocarcinoma was noted in a part of the adenocarcinoma, which occupied almost the entire portion of the surgical specimen. In addition, the precise localization of human chorionic gonadotropin (hCG) in the portion of choriocarcinoma was proved by the indirect immunoperoxidase stain (PAP method). The patient showed the gynecomastia clinically. The concentration of postoperative serum hCG was elevated and testicular abnormality was not noted clinically.

Changes in antibody titers in cattle infected clinically and subclinically with bovine leukemia virus.

Changes in titers of antibodies to protein and glycoprotein antigens of bovine leukemia virus (BLV) were investigated by immunodiffusion in two groups of cattle. In one group of cattle (10 head) which developed enzootic bovine leukosis during the observation period of 3 years, seven showed a gradual increase in antibody titers following a period of level antibody titers that persisted for several months to several years. The titer was finally eight to 32 times higher than the initial titer. The remaining three showed little change in titer. Of the 10 animals with tumors, three showed a decrease in the titer when they were in extremis. In contrast, in the second group of cattle (110 head), which were infected with BLV subclinically, the change in the titers during 1 and 3 years ranged from no change to four times the lowest titer that was observed during the period. Of the cattle which had a low level of antibody to one of the BLV antigens, a few became negative reactors to the respective antigen.

Regional variations in the incidence of end-stage renal failure in Japan.

CONTEXT: Despite recent medical advances, the number of patients beginning dialysis annually is increasing in both the United States and Japan. The ethnically homogeneous population of Japan presents an opportunity to study the presence of factors other than race/ethnicity that might contribute to incidence of end-stage renal disease (ESRD). OBJECTIVE: To determine if and where regional differences exist in ESRD in Japan. DESIGN, SETTING, AND SUBJECTS: Analysis of data reported by the Japanese Society for Dialysis Therapy based on the annual number of patients with ESRD beginning maintenance dialysis therapy in all 47 prefectures of Japan from 1982 to 1998. MAIN OUTCOME MEASURES: Mean annual ESRD incidence and increasing rate of ESRD in each of 11 predefined areas making up the entire country. RESULTS: Incidence of ESRD increased approximately 3-fold in Japan during the study years, from 81.3 per 1 million in 1982 to 237.6 per 1 million in 1998. Significant regional differences were found in both measures. The mean (SEM) annual ESRD incidence (P<.01) and increasing rate of ESRD (P<.01), respectively, were significantly different across Japan. Koshinetsu (140 [11] per 1 million and 9.1 [0.6] per 1 million/y) and Hokuriku (141 [12] per 1 million and 9.7 [0.5] per 1 million/y) were the areas with the lowest incidence and increasing rate of incidence, while Okinawa (188 [17] per 1 million and 13.4 [0.6] per 1 million/y) and Kyushu (179 [15] per 1 million and 12.0 [0.6] per 1 million/y) were the areas with the highest incidence and increasing rate of incidence. CONCLUSIONS: We found definite and significant regional differences in incidence and increasing rate of incidence of ESRD in Japan. Further analyses are needed to identify factors that contribute to these regional differences and thereby improve strategies for treatment of renal disease. JAMA. 2000;284:2622-2624.
