[Cellular glucocorticoid reception in man and differences in type of reaction to stress].

Three groups of individuals were investigated, which differed in constitution and reaction to stress. Group I comprised randomly selected individuals. Groups II and III were formed of highly qualified athletes, weight lifters and marathoners who matched those from Group I. Studies of the hormonal profile revealed significant differences in the baseline cortisol levels between Groups II and III. Evaluation of parameters of glucocorticoid reception by cultured individuals' fibroblasts also showed significant differences between Groups II and III. Employment of complex quantitative characteristics of potential cell sensitivity to the effect of physiological glucocorticoid concentrations, and of the resting biological effect of this hormone, characterising the efficacy of glucocorticoid reception in postreceptor and glucocorticoid-dependent processes in somatic cells, allowed for adequate qualitative evaluation of glucocorticoid-dependent reactions to stress on the cellular level which is indicative of the stress reaction type of the organism as a whole. Thus, genetically determined variability of cellular parameters was first revealed, the parameters being directly related to the ontogenetic formation of the stress reaction type. It is biologically understandable that this variability correlates with human resistance to long-term psychosomatic stress.

[In vitro movement of human embryonic fibroblasts with normal and anomalous sets of chromosomes].

Migration of diploid postnatal and embryonic diploid and aneuploid cells was studied using a modified method of investigation of leukocyte migration under agarose. The method permits to study migration of fibroblasts and other proliferating cells. Embryonic fibroblasts were shown to move faster, than postnatal fibroblasts. Cells with trisomy 7, 9 and C, and triploid cells were found to move slower than diploid cells. Locomotor disturbances are supposed to be the basis of impairment of morphogenesis in chromosomal anomalies in man.

[Cytotoxic action of the chemical substances found as admixtures in medical immunobiological preparations].

The cytotoxic action (CTA) of chemical substances contained as admixtures in medical immunobiological preparations on human diploid cells has been studied. Such chemical substances as rivanol and merthiolate in admissible concentrations show the highest degree of CTA. The results obtained in this investigation indicate that different concentrations of chemical substances may produce equal CTA; thus, thiolate in toxic in a dose of 0.8 microgram/ml; the same CTA is produced by aluminium sulfate in a concentration of 500 micrograms/ml. Small doses of chemical substances, producing no explicit manifestations of the cytotoxic effect, may have latent CTA determined by additional methods of investigation. CTA may be manifested as lethal, sublethal and latent cell lesions. In working out regulations on the test for CTA it is expedient to indicate admissible residual amounts of chemical substances contained in finished medical immunobiological preparations, considering that these amounts must be incapable of producing CTA in cell cultures. The conclusion has been made on the expediency of denoting small amounts of chemical substances capable of producing latent CTA as tentatively tolerable doses.

Collagen and fibronectin synthesis by trisomic and triploid fibroblasts from human spontaneous abortuses.

Collagen and fibronectin synthesis by trisomic and triploid fibroblasts derived from human spontaneous abortuses was studied. It was demonstrated that the level of fibronectin and collagen production in fibroblasts with trisomy 7, trisomy 9, and triploidy was reduced as compared with diploid cells. A correlation between this observation and an increased rate of intracellular 14C-procollagen degradation was also established for the anomalous strains. No difference in hydroxylation of 14C-proline residues in alpha 1(I) and alpha 2(I) collagen chains and no fluctuation in the collagen type (I): type III ratio was found in the strains with the abnormal karyotypes. It was concluded that differentiation of the abnormal fibroblasts was impaired. The data also favour the hypothesis that the deficiency of the fibroblasts in producing proteins may account for a variety of anatomic abnormalities of embryos.

[Determination of the amount of somatic cell glucocorticoid receptors as one of the prospective approaches to the individual prognosis of glucocorticosteroid therapy].

The authors discuss the importance of the content of receptors to glucocorticosteroid hormones in somatic cells to predict the effectiveness of glucocorticosteroid therapy. Prognostically important is the detection of a relatively low amount of receptors in cells when administration of corticosteroids at high doses might be most probably ineffective. Any human somatic cells are representative (with respect to the receptor content) for other cell types therefore investigation of the number of glucocorticoid receptors in lymphocytes (the quickest result) or in skin biopsy specimens (less traumatic for a patient) seems most effective.

[Chromosome imbalance and cellular proliferation potential in vitro].

The effect of chromosome unbalance on the cell proliferative potential (cloning efficiency) was studied. The proliferative potential of the cells obtained from spontaneous abortions with chromosome unbalance appeared drastically reduced as compared with the proliferative potential of the cells from medical abortions. Chromosome unbalance in postnatal cells did not lead to diminution of the proliferative potential as compared with control.

Disturbances in collagen synthesis in trisomic cells from spontaneously aborted embryos.

Collagen synthesis in cells with trisomy 7 and 9 derived from human spontaneous abortuses was found to be lower (5.06% and 5.53% respectively) than in the control diploid cells (8.80%). The ratio of collagen types (I/III) in trisomic strains did not differ from the control data while the amount of the degraded procollagen in trisomic cells was increased.

[Variability of alkaline phosphatase activity in diploid human cells in vitro].

Experiments were made with 19 strains obtained from different tissues (skin, lungs, muscles) of 8-10-week-old medical abortions and skin biopsies of healthy donors to study the manifestations of alkaline phosphatase (AP) activity in human diploid cells in vitro. Based on the data obtained it is concluded that AP activity is marked by demonstrable intra- and interstrain variability. The spectrum of "AP activity" trait variability is broader for transformed cells than for human diploid cells.

[Individual variability in the proliferative potential of human diploid fibroblasts in vitro].

The rate of individual variability of proliferative potential determined by the percentage of colonies containing 16 and more cells (the cloning efficiency) was studied 16 embryonic and 15 postnatal strains of human diploid fibroblasts. It is highly probable that these differences are hereditary in nature and determined by the genotype of the given cell line. This conclusion is based on the fact that the cloning efficiency is a criterion marking each individual cell line and is well reproducible in a series of experiments.

[Simultaneous activity of nucleolus organizer regions of human and Chinese hamster chromosomes in somatic cell hybrids].

In an interspecific human-Chinese hamster hybrid that retains 13 and 85.6% of the chromosomes of each parental complement, activity of nucleolus-organizing regions (NOR) of both type chromosomes is observed in 18.9% of the cells. Interspecific chromosomal associations are also noted. Unlike the parental lines of Chinese hamster cells, the hybrids show the associations of the NOR of Chinese hamster chromosomes. In hybrid cells, there occurs partial suppression of NOR activity in human and Chinese hamster chromosomes, while the NOR of the 3d chromosome of the Chinese hamster is completely suppressed.

[Isolation of hybrid clones of human-rodent somatic cells].

Mutant cells of Chinese hamster or mouse were fused with normal embryonic human cells. The fusion was induced by polyethylene glycol. The Eagle medium containing hypoxanthine, aminopterine, thymidine, glycine, and ouabain served as a selective medium. The parent cells were completely destroyed in the selective medium. The hybrid cells formed colonies which were isolated and reproduced for cytogenetic analysis. The cytogenetic analysis has confirmed the hybrid origin of the isolated clones.

[Prenatal diagnosis of the cri-du-chat syndrome in the case of balanced 5p--; 18p+ translocation in the mother].

The case studied is taken to emphasize the high risk of anomalous progeny being delivered from couples, who are the balanced translocation carriers. The "cri du chat" syndrome was diagnosed in a woman carrier of the balanced reciprocal translocation t(5, 18) (5p13; 18p11) during two successive pregnancies: the diagnosis was postnatal in the first pregnancy, and prenatal in the second. The prenatal diagnosis of the 5p--syndrome was made in amniotic fluid cell culture and verified in fetal skin culture obtained through fetal biopsy. A wider network of prenatal diagnosis services, primarily for couples carrying balanced chromosome rearrangements, could greatly contribute to the task of preventing severe hereditary diseases.
