Studies on the activity of selected lysosomal exoglycosidases in patients with systemic lupus erythematosus.

The activity of selected lysosomal hydrolases ( (beta-galactosidase, beta-N-acetylglucosaminidase, beta-xylosidase, alpha-mannosidase, alpha-fucosidase) was determined in the blood serum of patients with systemic lupus erythematosus (SLE). An increased activity of all the enzymes studied was found as compared to control subjects. Presumable mechanisms of the observed changes are discussed. It is suggested that the studied enzymes may be responsible for the desintegration of the connective tissue ground substance in SLE.

Serum glycoproteins and synovial fluid in seropositive and seronegative rheumatoid arthritis.

An increased level of glycoproteins (seromucoid, protein-bound hexoses, protein-bound hexosamines, protein-bound sialic acids) was found in the blood serum from patients with rheumatoid arthritis (RA) as compared with healthy individuals. Higher glycoprotein content was observed in the seropositive RA patients than in the seronegative ones. Higher levels of glycoproteins was also observed in the synovial fluid from seropositive RA patients than in the seronegative ones.

Activity of lysosomal enzymes in the heart muscle of healthy and hydrazinophtalazine-treated rats after isoprenaline injury.

Heart damage was produced by isoprenaline injection in healthy and hydrazinophtalazine-treated rats. The activity of beta-glucuronidase, beta-acetyloglucosaminase and acid phosphatase was determined in the heart muscle, liver and blood serum. The activity of the studied enzymes increased in the seventh day of experiment and after two weeks after injection was normalized. The increase in the enzyme activity in hydrazinophtalazine-treated animals was significantly lower than in healthy animals.

Effect of gamma radiation on tissue elastin content and serum elastolytic activity in rats.

The elastin content of aorta, heart, skin and lungs as well as the serum elastolytic activity were determined in rats exposed to radiation. It was found that a single irradiation of rats with gamma rays (500 r) caused a decrease of the elastin content in all examined tissues. The serum elastolytic activity in the irradiated rats was increased. It is suggested that elastin degradation following radiation may be caused by changes in its molecular structure and possibly, due to increased serum elastolytic activity.

Age-related changes in proteoglycans of human intervertebral discs.

The age-related changes in glycosaminoglycan composition of human intervertebral discs were studied. A large number of tissue samples taken from 301 individuals at autopsy or at operation of prolapsed discs were investigated. It was found that the aging process is accompanied by a progressive decrease in GAGs content, both in annulus fibrosus and in nucleus pulposus. The GAG: collagen ratio significantly decreases. The KS and Ch-6-S were found to be the most abundant GAGs in both parts of the disc and their amounts decrease during the aging. A distinct, age-related increase of KS/Ch-S and Ch-6-S/Ch-4-S ratios was observed. Significant quantitative differences between the GAGs contents in normal and prolapsed discs were found. The nuclei pulposi of prolapsed discs contained less GAGs, especially KS and Ch-6-S, in comparison to normal tissue of the same age. Also, the age-related decline of GAGs: collagen ratio is more distinct in prolapsed discs. It seems that these differences in GAGs composition may be important in the mechanism of predisposition to disc prolapse.
