Characterization of Yersinia pseudotuberculosis serogroups O9, O10 and O11; subdivision of O1 serogroup into O1a, O1b, and O1c subgroups.

In this study, three new antigens (O9, O10 and O11) of Yersinia pseudotuberculosis are described. The O1 antigen is further subdivided into O1a, O1b and O1c. The methods used to prepare specific antisera for O-antigen identification are also described. Furthermore, the H antigens of these new serogroups are determined and their biochemical characteristics analysed. The antigenic formulae of the known serogroups within the basic antigenic scheme introduced by the authors' laboratories are presented.

[The distribution and kinetics of 123I-MIBG in normal human hearts].

123I-metaiodobenzylguanidine (MIBG) myocardial scintigraphy was performed in twelve normal human subjects, and the distribution and clearance of MIBG were estimated. In PLANAR studies, heart to mediastinum (H/M) and lung to mediastinum activity ratio (L/M) were 2.68 +/- 0.45 and 1.82 +/- 0.23 in initial images, and 2.75 +/- 0.44 and 1.55 +/- 0.13 in delayed images. The clearance of MIBG in the lung was more rapid than that in the heart, 41.6 +/- 4.1 vs. 29.8 +/- 3.2 (%), therefore MIBG cardiac profiles were more clarified in delayed images. In SPECT studies, the regional relative uptake (RRU) in anterior, septal, inferior and lateral wall were 100.0 +/- 0, 93.9 +/- 6.9, 85.0 +/- 10.0 and 104.9 +/- 8.3 (%) in initial images, and 100.0 +/- 0, 96.8 +/- 9.6, 79.4 +/- 8.3 and 99.1 +/- 7.9 (%) in delayed images, respectively. The RRU in inferior wall was significantly lower than those in the other walls. The regional clearance rate (RCR) in these four walls were 25.1 +/- 4.6, 22.5 +/- 8.2, 29.7 +/- 8.3, 29.3 +/- 3.4 (%), respectively. The RCR in inferior and lateral wall were significantly larger than those in anterior and septal walls. The RRU in basal, mid and apical portion were 100.0 +/- 0, 104.5 +/- 3.3, 98.9 +/- 12.1 (%) in initial images, and 100.0 +/- 0, 103.9 +/- 4.5, 96.8 +/- 15.2 (%) in delayed images, respectively. The RRU in mid portion was significantly higher than that in basal portion.(ABSTRACT TRUNCATED AT 250 WORDS)

Characterization of an amorphous and soluble hemagglutinin from Yersinia pseudotuberculosis.

Yersinia pseudotuberculosis which were screened out depending on auto-agglutination and Ca2+ dependency, were examined for their production of hemagglutinin (HA), and its purification and characterization were performed. The HA with a broad reactivity with various mammalian erythrocytes was recovered from the culture supernatant of these strains grown at 37 C but not 25 C. HAs from two strains, R148R and T1040, were purified by salt precipitation, gel filtration and anion-exchange chromatography by HPLC. Both purified HAs were cysteine-deficient acidic protein with an apparent molecular weight in the range of 15,000 to 16,000. N-terminal amino acid sequences of the first 25 residues were found to share 12% identity with that of afimbrial adhesin from enterotoxigenic Escherichia coli 2230. Immunoelectron microscopy and immunodiffusion test with polyclonal antiserum raised against the purified R148RHA demonstrated that the HA was associated with the amorphous aggregates which were detached from bacteria. These results suggest that the HA of Y. pseudotuberculosis belongs to a third type of HA produced by the yersinial species.

Detection of porcine rotavirus in stools by a latex agglutination test.

We developed a simple agglutination test for the detection of porcine rotavirus in stools from pigs with diarrhea. The virus was detected with high sensitivity and specificity by a slide agglutination test using latex particles coated with antibody against the porcine rotavirus strain OSU (LA-antiOSU). The agglutination of LA-antiOSU with OSU on a glass slide was evident macroscopically within 2 min. The sensitivity of this latex agglutination (LA) test was four times higher than that of the electron microscope method. The LA test is available for the rapid diagnosis of porcine rotavirus infections.

[Two cases of adult Down's syndrome presenting parietal low uptake in 123I-IMP-SPECT].

We performed 123I-IMP-SPECT to patients with Down's syndrome who were supposed to have neuropathological findings of dementia of the Alzheimer's type. In case 1, a 52-year-old man with dementia, bilateral posterior parietal low uptake and right temporal low uptake were noted. Uptake was markedly lower in the right hemisphere. Case 2, a 38-year-old man not exhibiting clinical features of dementia, presented slightly decreased uptake in bilateral posterior parietal areas. We conclude that the patterns of abnormally decreased uptake in these cases of Down's syndrome are similar to those in dementia of Alzheimer's type, and that there is a possibility to detect the predemented stage of Down's syndrome by 123I-IMP-SPECT.

Selection of variants of avian infectious bronchitis virus showing tropism for different organs.

Avian infectious bronchitis virus strain Kagoshima-34 isolated from the kidneys of a chicken that died of nephrosis/nephritis lost its nephropathogenicity during intratracheal passage in SPF chickens. The resultant virus acquired stronger respirotropism but reduced tropism for kidneys. On the other hand strain Tottori-2 isolated from the trachea of a chicken suffering from severe respiratory disease did not lose its respirotropism after serial intravenous passage in SPF chickens. The serological properties of the passaged virus were investigated by virus neutralisation test. The antibody titres of both strains of virus fluctuated with progressive passage. The serological properties of the virus isolated from respiratory organs were not necessarily the same as those of the isolates made from the kidneys.

Investigations into resistance of chicken lines to infection with infectious bronchitis virus.

Although nine inbred and partially inbred lines of chickens were equally susceptible initially to infection with infectious bronchitis virus (IBV), the outcome of that infection varied considerably between the lines. A more detailed study of a line which was resistant (line C) and one which was highly susceptible (line 15I) confirmed that both lines could be infected initially with IBV but showed that the respiratory tract epithelium of line C chicks recovered more rapidly than that of line 15I chicks. Whereas more virus could be recovered from tissues of line 15I chicks following IBV inoculation, explants of tissues from each line inoculated in vitro appeared equally susceptible. The possibility of a different immunological response to IBV infection in the two lines is discussed and it is clear that further work is required to elucidate the mechanism involved in the differing susceptibility of the inbred chicken lines to inoculation with IBV.

Screening test of stimulants in human urine utilizing headspace gas chromatography for field test.

An accurate and simple screening method of stimulants in human urine using headspace gas chromatography utilizing heat of dissolution of potassium carbonate was developed. A 4.9-g portion of potassium carbonate was put into the vial prior to sending to the field, and a 5-ml aliquot of urine, suspected of containing stimulants and internal standard components was pipetted. After the vial was sealed and shaken by hand, 1 ml of its headspace gas was taken by disposable syringe and injected into the gas chromatograph. A compact gas chromatograph device with flame ionization detector and fused silica capillary column was developed for this experiment. Detection limits of methamphetamine and amphetamine were 1.0 micrograms/ml and 1.5 micrograms/ml, respectively.

Special features of distribution of Yersinia pseudotuberculosis in Japan.

The incidence of Yersinia pseudotuberculosis isolation from humans, animals, water, and pork in Japan is presented on the basis of a review of the literature and serotyping of 633 strains referred to our laboratory. Most of the strains belonged to serogroup 4b, followed in frequency of isolation by serogroups 3, 1b, 5b, 5a, and the others, whereas serogroup 1a has not been detected. Although strains were isolated from 11 species of animals, none were isolated from birds.

Intracerebral pathogenicity for chickens of avian influenza viruses isolated from free-living waterfowl in Japan.

The pathogenicity for chickens of 91 strains of avian influenza A virus isolated from such free-living waterfowl as whistling swan, pintail, tufted duck, mallard and black-tailed gull in Japan was tested. The majority of the virus strains infected and were pathogenic for the chickens. The virulence of these viruses seemed not to be as high as that of fowl plague virus. There were no significant differences in the intracerebral index score among the viruses belonging to the same subtype, irrespective of year of isolation or host.

Lack of correlation between haemagglutination and adherence to epithelial cells in Yersinia pseudotuberculosis.

Yersinia pseudotuberculosis was examined for its haemagglutinating activity and adherence to cultured epithelial cells (HEp-2) in relation to possession of a virulence (VW) plasmid and to growth conditions. VW-lacking (VW-) bacteria were isolated from ten VW+ strains of each serovar which, after they were grown on CFA plates at 37 degrees C, agglutinated the erythrocytes from five different species. In contrast to the bacteria possessing the plasmid (VW+) half of the VW- bacteria, grown on CFA plates at 37 degrees C, did not agglutinate any of the erythrocytes used and the other half agglutinated only human erythrocytes. Furthermore, when grown on CFA plates at 25 degrees C, neither VW+ nor VW- bacteria showed a haemagglutinating activity. When the bacteria were grown in CFA broth, only two strains grown at 25 degrees C did not agglutinate any of the erythrocytes tested. The VW+ and VW- bacteria of the remaining strains, grown either at 25 degrees C or 37 degrees C, showed relatively high haemagglutinating activity. Adherence to HEp-2 cells did not correlate with haemagglutinating activity in Y. pseudotuberculosis; the VW+ bacteria grown at 37 degrees C adhered to HEp-2 cells more efficiently than either the VW- derivatives or the VW+ bacteria grown at 25 degrees C, regardless of the growth medium. These results indicate that some of the haemagglutinins detected on Y. pseudotuberculosis are not involved in the adherence to HEp-2 cells.

Comparison of two strains of avian infectious bronchitis virus for their interferon induction, viral growth and development of virus-neutralizing antibody in experimentally-infected chickens.

Two field strains of avian infectious bronchitis virus were tested in specific pathogen-free chickens for interferon induction, growth and stimulation of virus-neutralizing antibody. Both strains induced interferon in lungs and trachea and neither strain induced it in kidneys. Strain Kagoshima-34, isolated from the kidneys of a chicken that had died of nephrosis/nephritis, induced interferon earlier than Strain Tottori-2, which was isolated from the trachea of a chicken with respiratory disease. Virus growth was generally more prolonged with Kagoshima-34, especially in the kidneys, but Tottori-2 persisted longer in the lungs.

Isolation of influenza A viruses from migratory waterfowls in San-in District, Western Japan in winters of 1980-1982.

In the two winters of 1980-1982, we surveyed migratory waterfowl of some species staying in San-in District, Western Japan for influenza virus at a few stations. From November 1980 to April 1981, only two strains of influenza virus, H13N1 and H11N6 subtypes, were isolated from 465 fecal samples from pintails but none from 255 samples from whistling swans nor from 625 black-tailed gulls. From November 1981 to March 1982, 17 viruses were isolated from 1156 fecal samples. Fourteen viruses, 10 H7N3, 2 H1N6 and 2 H3N8, were isolated from 459 feces samples from whistling swans. Two viruses, H13N3 and H13N6 subtypes, were isolated from 425 fecal samples from black-tailed gulls. A strain belonging to H1N3 subtype was isolated from 30 feces samples from mallards but no virus was isolated from 242 samples from pintails.

Enzymological characteristics of avian influenza A virus neuraminidase.

Neuraminidases of 18 strains of avian influenza A virus were examined by both colorimetric and fluorometric assays using fetuin and 4-methylumbelliferyl-N-Ac-alpha-D-neuraminide as substrates, respectively, to compare them with those of human influenza A and B viruses. The ratios of the neuraminidase activity of avian influenza virus measured by the colorimetric assay method to that measured by the fluorometric assay were distributed in the range of 2.4-20.3. The enzyme of avian influenza virus showed calcium-ion dependence in both assay methods. These results suggest that neuraminidase of avian influenza A virus is varies greatly from one strain to another in substrate specificity as compared with those of human influenza A and B viruses, and that some strains of avian influenza A virus have a neuraminidase with unique enzymological characteristics different from that of human influenza A virus as well as that of influenza B virus.
