Analysis of malignancy-associated DNA changes in the nuclei of buccal epithelium in the pathology of the thyroid and mammary glands.

The object of the investigation reported in this paper was to study, from the point of view of statistical and geometric theory of pattern recognition, the DNA optical density distribution peculiarities in the interphase nuclei of buccal epithelium present in the pathology of the thyroid and mammary glands. Two new indices to characterize this distribution (ratio of modal class volumes and relief index) are proposed. It is shown that in malignant neoplasms of the thyroid and mammary glands the changes in the nuclei of buccal epithelium are characterized by an increase in the optical density of DNA over a range from 0.15 to 0.30 in conventional units of measure, as compared with its values in benign pathological processes. The sensitivity of the proposed criterion for diseases of the thyroid gland is equal to 76.2% and the specificity is equal to 85.8%. For diseases of the mammary gland (excluding IDLC) we have discovered that the sensitivity of the method is equal to 94.29% and its specificity equal to 90.91%. In diseases of the mammary gland (including IDLC) we have discovered that the sensitivity of the method is equal to 71.42% and its specificity is equal to 90.91%.

[The determination of beta-casein gene expression and the assessment of hormonal homeostasis in gynecomastia patients and in breast cancer in men].

beta-Casein genes expression in breast epithelium was studied in male patients with various forms of gynecomastia and cancer. Blood serum levels of pituitary, sex and glucocorticoid hormones were assayed in 29 patients with gynecomastia and 22 cases of breast cancer, and in 25 of them beta-casein genes expression was evaluated additionally. Activation of the above genes was established in the tissues studied. Their level proved to be in a correlation with that of prolactin.

[Familial-populational studies of endometrial cancer: segregation and genetic dispersion analyses].

A clinico-genealogic investigation was carried out in 216 patients with endometrial cancer. Familial accumulation of endometrial and other cancer incidence was established. The segregation rates appeared to be lower than those expected from simple Mendelian models (2-11%). A multifactorial nature of endometrial cancer in overall susceptibility to the disease was found to be at 61%. A genetic correlation analysis showed endometrial cancer to share common genes with breast and gastric cancer in females. Tables of recurrent risk of the disease for relatives were prepared to be used in medico-genetic counseling.

Ornithine decarboxylase activity and polyamine content in adenocarcinomas of human stomach and large intestine.

Ornithine decarboxylase (ODC) activity and differential polyamine composition have been studied in macroscopically normal mucosa, adjacent mucosa, and neoplastic tissue of 95 patients with adenocarcinomas of stomach and large intestine. In tumors, we found increased ODC activity and polyamine content as compared with surrounding mucosa. ODC activity in macroscopically normal tissue of patients with tumors of stomach and large intestine increased as the disease progressed. An inverse relationship was observed between ODC activity in adenocarcinomas and differentiation.

[Pathognomonic signs of human papillomavirus infection in diagnostic material from the uterine cervix].

Etiological factors and mechanisms of pretumorous and tumourous processes in the female genitalia are presented in light of current concept of viral cocarcinogenesis. The utility of cytoscreening for the detection of the papilloma-virus infection in the uterine cervix mucosa is founded. Detailed cytological characteristics of cells in the uterine cervix mucosa with a cytopathic effect (coilocytosis, the presence of two or many nuclei, vacuolisation of cytoplasm, nuclear atypia) are presented. A selective localization of the human papilloma virus in the transformation zone of the uterine cervix is determined. The frequency of cells with cytopathic effect is 80.2%.

[Recurrent endometrial cancer after combined radiation therapy].

Data of a patients are reported who developed recurrence of highly differentiated cancer of the endometrium 20 years after combined. At that time the patient harboured also a borderline mucinous cystadenoma of the ovary with malignization foci. An analysis of the lymphocyte karyotype in the peripheral blood revealed a 14% increase of aberrant cells.

[Use of genetic analysis methods in examining patients with endometrial cancer].

An analysis of clinical and family history of 36 patients with endometrial cancer revealed clustering of tumors in some families. Cytogenetic studies of peripheral lymphocytes showed increased counts of aberrant cells in patients with endometrial cancer as against their values in normal women and patients with endometrial hyperplasia.

[Quantitative characterization of nuclear polymorphism of the epithelial cells in fibroadenomatosis and cancer of the breast].

A method for quantitative analysis of epithelial cell nuclear polymorphism was suggested, viz. identification of general statistical population using Petunin's criterion. This criterion was employed to assess heterogeneity of visible surface of interphase epithelial cell nuclei and to assay nuclear DNA level in fibroadenomatous hyperplasia and cancer of the breast. Heterogeneity index (h), alongside with other parameters, appeared useful for quantitative assessment of the disease: heterogeneity index values ranging 0.1-0.4 point to pronounced heterogeneity of epithelial cell nucleus surface and DNA level, and are suggestive of malignant transformation of tissue, whereas benign proliferation of the epithelium is usually characterized by 0.4 less than h less than or equal to 0.9.

[Clinico-morphologic manifestations of the regression of a primary focus of malignant melanoma of the skin].

The general laws and clinico-morphologic peculiarities of the regression of cutaneous melanoma in 4 patients, which occurred spontaneously or under the influence of the local use of non-conventional methods of therapy are presented. It is shown that resorption of primary neoplasm is not the ultimate outcome of the tumour process, and because of the development of metastases, the prognosis remains unfavourable. Regression of a primary focus of cutaneous melanoma is estimated as one of the manifestations of the progression of tumour growth.

[Quantitative changes in the karyotype during hyperplastic processes in cancer of the endometrium].

Cytogenetic examination of scrapings obtained from 42 patients with uterine bleeding was performed. A decrease in diploid cell level matched by a rise in that of aneuploid and aberrant cells was seen both in carcinoma and adenomatous hyperplasia of the uterine mucosa. A correlation was established between the level of aneuploid and aberrant cells, on the one hand, and degree of endometrial hyperplasia, on the other. Changes in karyotype in hyperplastic lesions of the endometrium are thought to be a factor predisposing to cancer development.

[Determination of the individual sensitivity of human stomach cancer cells to fluoropyrimidines by the cytomorphological criteria of heterotransplants in diffusion chambers].

An optimal morphologic complex of parameters for evaluation of individual sensitivity of heterotransplants to two anti-cancer drugs--5-fluorouracil and ftorafur has been developed on the basis of clinical and experimental material and using 76 gastric malignant tumours, 1320 heterotransplants in diffusion chambers (DC) in vivo which were implanted to 660 animal recipients. A simplified preclinical evaluation method of the individual sensitivity of the human gastric tumour to fluoropyrimidines is recommended based on dynamic level of mitotic activity in DC in vivo heterotransplants.

[Complex cytomorphological determination of the degree of anaplasia of breast cancer cells].

Complex study of cytological, morphometric and cytogenetic indices of mammary carcinoma in 92 women and an evaluation of survival of 196 patients depending on the level of tumour anaplasia. The correlation is shown between the tumour anaplasia signs including an interphase nuclei morphometry, X-chromatin content and mitotic index on the one hand and the degree of clinical malignancy confirmed by the results of 5-years survival on the other.

[Cytomorphological and endoscopic parallels of precancer and early cancer of the cervix uteri].

Complex clinico-endoscopic and morphological study of the uterine cervix pathology is performed in more than 20 000 women since 1962 up to now. The possibility of correlation between the results of endoscopic tests and those of cytomorphological study of the uterine cervix and cervical channel pathological processes is shown. The limitations of the endoscopic analysis are noted. Endoscopical-morphological classification is suggested and tactics of the patient examination with the risk, pretumourous conditions and early carcinoma is grounded.

[Current problems of oncogenetics].

Data available in literature on clinical oncogenetics are reviewed. Pretumour and tumor processes with dominant, recessive and multifactorial inheritance are analyzed in particular. A possibility of application of cytogenetic indices to diagnose initial forms of tumour process and determine the level of anaplasia of developed malignant tumours is suggested. The role of Ukrainian oncologists in oncogenetic studies with the view of comprehending the nature of malignancy and expanding diagnostic possibilities in practical oncology is shown.

[Comparative study of the DNA content and cross-sectional area of the interphase nuclei of epithelial cells in stomach polyps and cancer (based on gastric biopsy data)].

Cytospectrophotometric measurement of DNA level of interphase nuclei versus their cross-reaction area was carried out in mucosal cells of the stomach. The material included gastric bioptates from healthy subjects and patients with single and multiple adenomatous polyps of the stomach as well as gastric cancer. Tumor cells, and to a lesser degree, those of multiple polyps exhibited certain changes in DNA content: histograms showed several peaks and variations in DNA level increased, the latter being unmatched by a rise in DNA content per nucleus. A correlation was established between DNA content and cross-reaction area of interphase nuclei. Therefore, both parameters may serve for diagnosis.
