Pyridinoline cross-links as markers for primary and secondary bone tumors.

BACKGROUND: Determination of hydroxylysylpyridinoline (HP) and lysylpyridinoline (LP) in urine is a promising method to determine bone resorption. This method is independent of gender, diet and kidney function (creatinine clearance > 25 mL/min). METHODS: The diagnostic efficacy of HP and LP was assessed in the urine of adult patients suffering from primary malignant bone tumors (n = 24), bone metastases (n = 38) and soft tissue sarcoma with additional osseous involvement (n = 13). The values were compared with those obtained from 543 healthy controls (aged 15 to 65 years). RESULTS: Results clearly exhibited a highly significant increase in HP values (57.75 nmol/mmol creatinine) in adult tumor patients (aged 15 to 65 years) in all three subgroups in comparison with the control group values (22.23 nmol/mmol creatinine) (p = 0.001). Although the LP fraction is more specific for bone than HP, the values of LP from all subgroups of the adult tumor patients were less distinctly but still significantly increased (p = 0.008). Regarding the HP:LP ratio, tumor patients exhibited a markedly increased average molar HP:LP ratio (12.0:1) in comparison to controls (6.6:1). CONCLUSION: Determination of HP and LP in urine appears to offer clinical utility for the detection of primary and secondary bone neoplasms. Prospective, longitudinal studies will be necessary to evaluate whether the HP:LP ratio is indicative of a relapse of the tumor and consecutively may be adopted in the follow-up of patients with neoplasms.

[Diagnosis of muscle function in hereditary muscular dystrophies].

The evaluation of musclefunction im myopathies has been done upto now only by clinical trial. But in contrast in the field of medicine in sports has been proved the method of dynamometry of the muscles for a longer time which are necessary for objectivation of musclefunktion in myopathies. In this paper we present first results of health children and children suffering from Duchenne myopathy.

Diacylglycerol species in microsomes and mitochondria of normal and dystrophic human muscle.

The concentration of 1.2-diacylglycerol in mitochondria and microsomes of normal human muscle was estimated to be 1.7 +/- 0.6 and 9.2 +/- 1.9 nmol X mg protein-1, respectively. The molecular species profile of 1.2-diacylglycerol was obtained using high pressure liquid chromatography which yielded 14 fractions. Corresponding studies were carried out for normal and dystrophic muscle. No significant difference could be seen between the 1.2-diacylglycerol patterns in normal muscle mitochondria and in microsomes. There was a radical shift in the 1.2-diacylglycerol pattern in both dystrophic organelles, mainly with a decrease in 16:0/16:0 species. In dystrophic mitochondria the total level of 1.2-diacylglycerol was higher.

[Electron microscopical findings in the sural nerve of twins with metachromatic leukodystrophy. An attempt to classify the findings according to distinctive phases of the disease (author's transl)].

Sural nerve biopsies taken from a pair of twins with metachromatic leukodystrophy were investigated by electron microscopy. The morphological findings showed distinct alterations. In the first twin the particularity was a large number of granules containing sulfatides. In the second twin, however, the main finding was a damage of myelin sheaths evidently caused by an excessive and morphologically as well as chemically defective myelin formation. According to the grading given by Ulrich the first case corresponds to the stadium one, in which a storage of sulfatides dominates, whereas case two showing a severe damage of myelin sheaths is characteristic of the second stadium of metachromatic leukodystrophy.
