Antitumor effect of lysine-isopeptides.

Isopeptides (ϵ-peptides) of lysine, with a given Mw and low polydispersity (10-400 units), were synthesized to study the relationship between their chemical structure and biological effect. The designed compounds were of high purity, low polydispersity and high stereochemical purity. The effect of the compounds was tested on a human erythroleukemia cell line (K-562) and on four transplantable mouse tumors (L1210 lymphoid leukemia, P38 macrophage derived tumor, Ehrlich ascites carcinoma, Lewis lung tumor /LLT/). In case of the L1210 and P388 tumors and the Ehrlich carcinoma, survival of the animals was used as an indicator of the effect. In case of the Lewis lung tumor, the number and size of metastases in the lung and/or liver of treated and untreated mice were used as indicators. The polymers of polymerisation degree 80-120 (Mw 10.2-15.4 KD) showed the strongest antiproliferative effect both on K562 cells and the tumors growing in vivo. This effect was manifest with a significantly higher survival rate as compared to the control (L1210, P38, Ehrlich ascites), furthermore, by a decrease in the number and size of liver and lung metastases (LLT).

[Detection of minimal residual diseases in B-cell tumors using PCR specific for the immunoglobulin heavy chain gene].

In B-cell non-Hodgkin's lymphomas (NHL), clonal rearrangement of the immunoglobulin heavy chain (IgH) gene provides a useful marker for the detection of minimal residual disease (MRD) after treatment. To explore clinical usefulness of polymerase chain reaction (PCR) analysis of clonal IgH gene rearrangement in the detection of MRD a follow up study of 10 patients with B-cell NHL have been performed. At the time of diagnosis, tumor DNAs were PCR-amplified using sense primer specific for the heavy chain variable region (VH) and antisense primer specific for the heavy chain joining region (JH) of the IgH gene. The clonal rearrangement of IgH gene detected by PCR was used as clonal marker to determine MRD after treatment. In three cases, where clinical remission was not achieved, clonal IgH gene rearrangement was detected after the treatment. In seven cases, clinical remission was achieved after induction therapy but the PCR analysis revealed clonal IgH gene rearrangement in three of the cases. In all of the three cases, where MRD was detected by PCR, clinical relapse developed after 7-28 months of the therapy. In all cases that have relapsed, the IgH gene rearrangement was identical at the time of initial diagnosis and at the relapse. This study demonstrates that PCR analysis of clonal IgH gene rearrangement is a useful method to monitor and detect MRD before clinical relapse.

[Mesangiocapillary glomerulonephritis].

Mesangiocapillary glomerulonephritis (MCGN) accounting for 15 percent of adult glomerulopathy has not received proper attention in the Hungarian medical literature. The present study offers a detailed description of the procedures to be used for distinguishing its three types, which is only possible by histological, mainly electron microscopic examination of kidney biopsy samples. Type I represents subendothelial deposits; Type II is the "sausage-like" dense deposits localised on the lamina densa of the glomerular basal basement membrane; Type III is the mixed type. There is no significant difference in their clinical appearance. They occur most frequently as "coloured nephrosis" (the syndrome of nephrosis is associated with haematuria and/or hypertension). The course of the disease is characterised by periodicity, and in 50 percent of the cases kidney failure may develop. The therapy for MCGN has not been established. The administration of steroid, cyclophosphamide and anticoagulant can have a favourable effect.

Humoral leukocyte adherence inhibition (H-LAI) follow-up trials on malignant process in mice.

Humoral antitumor response of Lewis lung tumor-bearing mice was measured by humoral leukocyte adherence inhibition (H-LAI) test. The reactivity of serum against tumor antigens prepared from primary tumor and lung metastases could be revealed at the first day after injection of tumor cells. On the other hand, the macroscopical appearance of the primary tumors and lung metastases was observed after 5 and 11 days, respectively. These findings suggest that the immune reaction of the host could be detected by H-LAI test earlier than the tumor can manifest itself. The antigens prepared from primary tumors and metastases seemed to be very similar, however, the metastasis antigen had additional determinants as detected by the H-LAI technique. Comparison of two tumor lines, the original Lewis lung tumor (LLT) and its variant with high metastatic capacity (LLT-HH), showed high similarity between their antigens as tested in H-LAI system by cross-reaction probes.

[Comparative study of the consumption of cariogenic food in monozygotic and same-sex dizygotic twins].

The eating habits of the 111 monozygotic and 67 like-sexed dizygotic twins were compared with respect to cariogenic foods. The food habits were recorded using a seven-day dietary history with the weekly frequency of the cariogenic foods examined. The biometrical analysis showed that the eating habits of the monozygotic pairs were more similar than that of the dizygotic twin pairs. The differences were statistically significant. Based on these findings, it seems that genetic factors play a role in the development of the food habits.

Comparative study on Lewis lung tumor lines with 'low' and 'high' metastatic capacity. I. Growth rate, morphology and resistance to host defence.

A highly metastatic variant (LLT-HH) of liver metastasizing Lewis lung tumor (LLT) has been selected. Comparative studies were made on proliferation rate, morphological characteristics and host cell interaction of these two lines. Increased metastasis formation seemed to result from the selection of cells with increased resistance to nonspecific host effector cells.

The physico-chemical properties of tumor cells with different metastatic potential.

Authors investigated the total and surface sialic-acid content and elpho mobility of Lewis lung lines with low (LLT-parent) and high (LLT-HH selected) metastatic capacity. They pointed out, that the total and surface sialic-acid content of LLT-HH primary tumor cells is higher than in the cells of the LLT line. Despite the increase of sialic-acid on the cell surface, the electrophoretic mobility of the cells was unchangeable. The changes in sialic acid content are the markers of the metastasizing cells, but not of the highly metastatic cell lines.

Transmission and scanning-electronmicroscopy of experimental liver metastases derived from intrasplenically growing primary tumor.

Liver metastasis formation from intrasplenically growing Lewis lung tumor was studied with transmission and scanning electron microscopy. Tumor cells arrested in the sinusoids formed desmosome-like junctions with endothelial cells and crossed the endothelial lining at intercellular gaps and by transcellular diapedesis. The metastatic foci had no newly formed vessels, and the stroma was provided by non-parenchymatous liver cells. Morphology suggested different outcome for tumor cell-host cell interactions at different stages of tumor growth. The host cell activity to destroy tumor cells was present only at the early stage but disappeared later, when the tumor cells were ready to phagocyte normal cells. The reason for this could be the immunosuppressive effect of the primary tumor. Results emphasize the importance to study metastasis formation in primary tumor-bearing hosts.

Increased metastatic capacity of Lewis lung tumor cells by in vivo selection procedure.

Lewis lung tumor cells from liver metastases originally obtained from an intrasplenic tumor, and lung metastases obtained from an intramuscular transplant, were repeatedly passaged in the corresponding transplantation sites (spleen, intramuscular). Cells from liver metastases injected into the spleen gained an increased metastatic capacity. The same phenomenon was observed with lung metastatic cells injected intramuscularly, but to a lesser degree. In all passages metastases occurred only in the organs receiving the venous blood from the primary site. The enhanced metastasis formation may be a result of a selection of tumor cells resistant to host cytotoxic cells and/or of selection of tumor cells 'seeding' successfully in target organs.

[Misleading self-inflicted gunshot wound].

One deceiving case of suicide with firearm is reported. In the heart area three entrance-wound were seen. At first sight these were the injuries to account for the death. At the autopsy in the mouth another entrancewound was, found. The death occurred due to gunshot into the mouth.

Comparative study on Lewis lung tumor lines with 'low' and 'high' metastatic capacity. II. Cytochemical and biochemical evidence for differences in glycosaminoglycans.

The enhanced metastatic capacity of an in vivo selected Lewis lung tumor line (LLT-HH) was correlated with changes in cell-associated glycosaminoglycans (GAG) using ultrastructural cytochemistry, flow cytometry and biochemistry. The increase in highly sulphated GAG content on the cell membrane of LLT-HH cells compared to the parent LLT cells was demonstrated cytochemically. Using in vitro [3H]glucosamine labelling of GAG components it was shown that the LLT-HH cells were characterized by a high production of heparan sulphate while the parent LLT line had a high hyaluronic acid-chondroitin sulphate production. The high metastatic phenotype is accompanied by an altered production of cell-associated GAGs.

Chemotherapeutic sensitivity of liver metastases from intrasplenically-growing Lewis lung tumor.

A recently developed metastatic tumor model was used to study the therapeutic response of liver metastases derived from intrasplenically growing LLT. Treatment was performed on the day following surgical removal of the 'primary tumor'. The life-span of tumor-bearing animals and the number and volume of liver metastases were measured. Cyclophosphamide and 13324 (a new bifunctional nitrosoureido derivative) proved to be most effective. Some other drugs (5-FU, MeCCNU, Lycurim) showed a temporary regression in the formation of macrometastases without influencing the life-span. Adriamycin was slightly more effective given i.p. than i.v.
