[Biosensors for the determination of phenol and benzoate on the basis of Rhodococcus cells and enzyme extracts].

An amperometric biosensor for determination of phenol, cresol, benzoate and 2-methyl-4-chlorophenol using Rhodococcus-cells and enzyme extracts of Rhodococcus has been developed. The influence of cultivation of Rhodococcus-cells and preincubation of the biosensor with desired substrate on sensibility and specificity was been investigated. In relation to cultivation and preincubation the Rhodococcus sensor was high specific to benzoate or phenol and cresol. A linear range was obtained for phenol and benzoate up to 80 mumol and for 2-methyl-4-chlorophenol up to 400 mumol. The biosensor using enzyme extracts show a higher specificity, it is but necessary NADPH. A further disadvantage is the little measuring range of this sensor.

Benzoates

Amperometric determination of ammonium ions with a microbial sensor.

A sensor for NH+4 ions has been developed, which consists of immobilized micro-organisms (Bacillus subtilis, Pseudomonas aeruginosa, Trichosporon cutaneum) in combination with an electrochemical transducer. This sensor is based on the measurement of acceleration of respiration after addition of NH+4 in the presence of glucose. The physiological background of this signal and its connection with NH+4 ion uptake and/or metabolism is discussed. The response time of the sensor is about 5-10 s for NH+4 ions. A linearity was observed between 0.005 and 0.15 mmol dm-3 NH+4 ions. The sensitivity of the sensor remained almost constant for 12 days. The sensor was used to determine NH+4 ions in a microbial fermentation broth.

Bacillus subtilis

Prediction of the risk of hereditary retinoblastoma, using DNA polymorphisms within the retinoblastoma gene.

Using molecular cloning, we earlier isolated the "retinoblastoma gene"; mutations or deletions at this locus are associated with the hereditary predisposition to some human cancers, especially retinoblastoma and osteosarcoma. To develop diagnostic tests for such a predisposition, we identified restriction-fragment-length polymorphisms (RFLPs) within the retinoblastoma gene and tested their usefulness in predicting the risk of cancer in 20 families with members who had hereditary retinoblastoma. We were able to make predictions in 19 of the 20 kindreds. In 18 kindreds, we demonstrated a consistent association of marker RFLPs with the mutation predisposing to retinoblastoma. In the 19th kindred, there may be a lack of cosegregation of the DNA polymorphisms within the gene and the site of the mutation predisposing to retinoblastoma. However, there is uncertainty about the clinical diagnosis of the retinal lesion in a key member of this kindred; if the lesion is not a retinoblastoma, there is no discrepancy between the DNA polymorphisms and the retinoblastoma trait. We conclude that it is feasible and clinically useful to use these DNA polymorphisms to determine the risk of cancer.

Formation of some extracellular enzymes during the exponential growth of Bacillus subtilis.

The formation of the exoenzymes, neutral and alkaline proteinase as well as alpha-amylase of Bacillus subtilis, is characterized by the same time course. The exoenzyme formation starts in the exponential phase of growth by an excess of C and N sources. We assume that carbon metabolism of pyruvate is responsible for the exoenzyme formation during this growth phase. The proteinase formation at the transient and/or stationary phase of growth is related to amino acid limitation.

Formation of extracellular neutral proteinase and the stringent response in Bacillus subtilis.

The kinetics of extracellular neutral proteinase synthesis by an isogenic stringent (IS58) and a relaxed (IS56) strain of B. subtilis were compared. The specific enzyme formation rate by the stringent strain was higher than that of the relaxed one. Norvaline addition (1 mg/mL) induced the formation of pppGpp and ppGpp, respectively, as well as the appearance of extracellular neutral proteinase activities in cultures of the stringent strain IS58 and a strain with high proteinase production (ZF-178) only. These correlations support the suggestion that (p)ppGpp are involved in the regulation processes responsible for production of extracellular neutral proteinases by B. subtilis.

Bacillus subtilis

An electrochemical method for determination of cell respiration.

The respiration of microorganisms was determined with a sensor composed of microorganisms and an oxygen electrode. The microorganisms (suspensions of about 100 microliters) are sedimented by centrifugation in a special tube on a very thin paper layer (diameter 4 mm). This paper layer is sandwiched between a polyethylene membrane and a dialysis membrane. A linear relationship is obtained between the current and the cell mass of Bacillus subtilis (between 0.05-0.35 mg dry weight per electrode preparation). When an assimilable substrate is added to the measuring solution the respiration rate is increased. It is possible to determine very fast changes in the respiration rates (1-5 sec). The application of this electrode system for investigations of the physiological state of cells during the growth cyclus is demonstrated.

Bacillus subtilis

Extending formant transitions may not improve aphasics' perception of stop consonant place of articulation.

Synthetic speech stimuli were used to investigate whether aphasics' ability to perceive stop consonant place of articulation was enhanced by the extension of initial formant transitions in CV syllables. Phoneme identification and discrimination tests were administered to 12 aphasic patients, 5 fluent and 7 nonfluent. There were no significant differences in performance due to the extended transitions, and no systematic pattern of performance due to aphasia type. In both groups, discrimination was generally high and significantly better than identification, demonstrating that auditory capacity was retained, while phonetic perception was impaired; this result is consistent with repeated demonstrations that auditory and phonetic processes may be dissociated in normal listeners. Moreover, significant rank order correlations between performances on the Token Test and on both perceptual tasks suggest that impairment on these tests may reflect a general cognitive rather than a language-specific deficit.
