On the possibility of differential diagnosis at elevated erythrocyte sedimentation rate by analysis of the concentrations of blood plasma proteins--a model study.

An elevated erythrocyte sedimentation rate is generally regarded as an unspecific and mostly pathological indicator of inflammation or tumour. However, we have determined the concentrations of plasma/serum proteins that influence the erythrocyte sedimentation rate in numerous samples from several groups of patients with different diseases, including 2 forms of cancer. Equations have been developed by which the 1 h value of erythrocyte sedimentation rate can be expressed as the sum of disease-specific coefficients for each protein multiplied by the measured concentrations of the respective proteins. These equations are shown to be disease-specific with 64-93% probability. Such equations may thus form the basis for differential diagnosis.

"Cytomic" data system modules--modern electronic devices for flow cytometric data handling and presentation.

Flow cytometric investigations can be very complex when several parameters of cells are to be measured and cells must be sorted. On the other hand, many questions may be answered by a single-parameter analysis. The Cytomic modular data system can easily be adapted to such different needs of flow cytometry. It consists of four types of modules which can be used as stand-alone elements or can be combined into a larger data system: CYTOMIC 12, a one-and two-parameter analyzer; CYTOMIC 123, a module mainly provided for on-line three-parameter data uptake and processing; CYTOMIC SORT, a fully software-controlled sort module, where sort windows may be selected by a built-in light pen; and the CYTOMIC LIST list mode module, which is able to collect up to eight cell parameters unclassified on a 1.2-MB floppy disk. Each module built from Z80 microprocessor Eurocards has its own keyboard, display screen, power supply, analog-to-digital pulse height conversion equipment, and an intermodule data communication bus. Each module has a preprogrammed set of functions, which is specially tailored for flow cytometric applications. This kind of data system allows the user to start with a single module for one-and two-parameter analysis. When required, the system may successively be enlarged without losing the stand-alone feature of each module.

A new flow cytometric pulse height analyzer offering microprocessor controlled data acquisition and statistical analysis.

The instrument described is capable of storing up to 112 one parameter histograms (128 channels) or 16 one parameter and 3 two parameter histograms (64 X 64 channels). A low cost 10 Mhz oscilloscope displays the graphical and alphanumerical data. During data acquisition, the original pulses, an analogue rate meter and the growing histogram are displayed simultaneously on the screen. The instrument may be completely operated by an 18 element keyboard. The data processor allows calculation of integrals, mean values, standard deviations and coefficients of variation of operator selectable regions of the acquired histograms. Gaussian distributions determined by the statistical parameters may be compared directly with the measured distributions. Distribution curves can be normalized, shifted, added, subtracted or compared with artificial distributions which are generated by superimposition of distinct Gaussian curves. Exponentially fitted background curves can be subtracted from the measured histograms. A text line at the top of the screen displays the numerical values and explanatory remarks. The two parameter histograms may be rotated, observed from the sides or back and projected either on the x- or y-axes. The projections or selected lines or columns can be transferred to the one parameter range and treated statistically. Sequences of one parameter distributions may be transferred two parameter range and displayed as pseudo-two-parameter histograms.
