Design of an interface to allow microfluidic electrophoresis chips to drink from the fire hose of the external environment.

An interface design is presented that facilitates automated sample introduction into an electrokinetic microchip, without perturbing the liquids within the microfluidic device. The design utilizes an interface flow channel with a volume flow resistance that is 0.54-4.1 x 10(6) times lower than the volume flow resistance of the electrokinetic fluid manifold used for mixing, reaction, separation, and analysis. A channel, 300 microm deep, 1 mm wide and 15-20 mm long, was etched in glass substrates to create the sample introduction channel (SIC) for a manifold of electrokinetic flow channels in the range of 10-13 microm depth and 36-275 microm width. Volume flow rates of up to 1 mL/min were pumped through the SIC without perturbing the solutions within the electrokinetic channel manifold. Calculations support this observation, suggesting a leakage flow to electroosmotic flow ratio of 0.1:1% in the electrokinetic channels, arising from 66-700 microL/min pressure-driven flow rates in the SIC. Peak heights for capillary electrophoresis separations in the electrokinetic flow manifold showed no dependence on whether the SIC pump was on or off. On-chip mixing, reaction and separation of anti-ovalbumin and ovalbumin could be performed with good quantitative results, independent of the SIC pump operation. Reproducibility of injection performance, estimated from peak height variations, ranged from 1.5-4%, depending upon the device design and the sample composition.

Effect of inhibitors of Na+/H+-exchange and gastric H+/K+ ATPase on cell volume, intracellular pH and migration of human polymorphonuclear leucocytes.

1. Stimulation of chemotaxis of human polymorphonuclear leucocytes (PMNs) with the chemoattractive peptide fMLP (N-formyl-Met-Leu-Phe) is paralleled by profound morphological and metabolic alterations like changes of intracellular pH (pHi) and cell shape. The present study was performed to investigate the interrelation of cell volume (CV) regulatory ion transport, pHi and migration of fMLP stimulated PMNs. 2. Addition of fMLP to PMNs stimulated directed migration in Boyden chamber assays and was accompanied by rapid initial intracellular acidification and cell swelling. 3. Inhibition of the Na+/H+ exchanger suppressed fMLP stimulated cell migration, accelerated the intracellular acidification and inhibited the fMLP-induced cell swelling. 4. Step omission of extracellular Na+ caused intracellular acidification, which was accelerated by subsequent addition of gastric H+/K+ ATPase inhibitor SCH 28080, or by omission of extracellular K+ ions. In addition Na+ removal caused cell swelling, which was further enhanced by fMLP. 5. H+/K+ATPase inhibitors omeprazole and SCH 28080 inhibited stimulated migration and blunted the fMLP-induced increase in CV. 6. Increasing extracellular osmolarity by addition of mannitol to the extracellular solution caused cell shrinkage followed by regulatory volume increase, partially due to activation of the Na+/H+ exchanger. In fMLP-stimulated cells the CV increase was counteracted by simultaneous addition of mannitol. Under these conditions the fMLP stimulated migration was inhibited. 7. The antibacterial activity of PMNs was not modified by Hoe 694 or omeprazole. 8. Western analysis with a monoclonal anti gastric H+/K+ ATPase beta-subunit antibody detected a glycosylated 35 kD core protein in lysates of mouse and human gastric mucosa as well as in human PMNs. 9. The results indicate that fMLP leads to cell swelling of PMNs due to activation of the Na+/H+ exchanger and a K+-dependent H+-extruding mechanism, presumably an H+/K+ ATPase. Inhibition of these ion transporters suppresses the increase in CV and precludes PMNs from stimulated migration.

Characterization of sodium-selective optode membranes based on neutral ionophores and assay of sodium in plasma.

Plasticized poly(vinyl chloride) membranes that incorporate a highly lipophilic sodium-selective neutral ionophore (ETH 4120) and novel H(+)-selective chromo-ionophores, lipophilic isologs of Nile Blue, are used in the competitive binding of Na+ and H+ for a reversible, optical determination of sodium activities in buffered solutions at different selected pH values. These optode membranes are used to assay total sodium concentrations in pH-buffered human blood plasma.

A new double-barrelled, ionophore-based microelectrode for chloride ions.

A new Cl- selective microelectrode based on the ionophore 5,10,15,20-tetraphenyl-21H,23H-porphin manganese(III) chloride is presented which discriminates better against HCO3- and several organic anions than electrodes containing the Corning 477913 ion-exchanger. Using a redesigned construction procedure, fine-tip double-barrelled microelectrodes were produced which had slopes of -52.4 +/- 0.6 mV (SE, n = 24), resistances of about 7.10(11) omega and a selectivity coefficient log KpotCLHCO3 of -1.40 +/- 0.03. Some electrodes showed a small unexplained sensitivity to pH greater than 7.6. When used to puncture cells of isolated S3 segments of rabbit renal proximal tubule during perfusion with HCO3- Ringer solution, the electrodes gave a membrane potential of -69.8 +/- 1.5 mV and an intracellular Cl- activity, [Cl-]i, of 35.3 +/- 2.6 mmol/l. Upon switching bath and lumen perfusions to Cl- -free solutions the "residual" [Cl-]i dropped to 1.20 +/- 0.03 mmol/l, while in similar measurements with ion-exchanger electrodes the "residual" [Cl-]i dropped only to 10.9 +/- 0.5 mmol/l. These observations demonstrate the superiority of the new electrode and prove that previously determined high [Cl-]i values in Cl- -free ambient solutions reflect interference problems rather than non-exchangeable intracellular chloride.

[Protein quality of flatbread and extruded dry flatbread made from full-grain rye groats].

The effect of various processing methods used for the production of dry flatbread from ground whole rye on the nutritional protein quality were investigated by means of the nitrogen balance method in growing rats. The following products were tested: untreated ground whole rye, baked crisp bread, extruded flatbread without and with the addition of 2.5% and 5.0% of saccharose. The apparent protein digestibility (PD) of the 5 samples varied between 69% and 72% and was neither affected by the method of processing nor by the difference in sugar content. Compared to the untreated raw material (NPU = 41%; BV = 59%) net protein utilization and biological value in crisp bread baked by the traditional method (NPU = 35%; BV = 48%) and in extruded flat bread with 5% sugar (NPU = 35%; BV = 51%) were significantly reduced (P less than 0.05). In extruded flatbread without and with 2.5% sugar, NPU and BV of the raw material were essentially maintained. The results indicate that in comparison with the baking process the extrusion of flatbread is more suitable to protect the protein quality. The addition of more than 2.5% sugar caused protein damage in extruded flatbread, presumably as a result of Maillard reactions.

[Effect of various extrusion conditions on the protein quality of soybean and whole rye meal].

Using a Creusot-Loire twin-screw extruder (Type BC 45), ground soy bean and whole rye meal were extruded under various processing conditions (12% and 18% water content in the raw material, 129 degrees C and 165 degrees C product temperature in the case of soy bean, 12% and 18% water content, 165 degrees C product temperature in the case of rye, screw speed of 150 R/min, for all samples). The nutritional protein quality (apparent protein digestibility PV, net protein utilization NPU, and biological value BW) of the extrudates was determined by the nitrogen balance technique in growing rats. Compared to the untreated raw material no decrease in PV or NPU was noted for extrudates made from soy bean or rye. Extrusion of ground soy bean at 165 degrees C and 18% water content caused a significant decrease of BW from 68% to 64%. The lower water content (12%) induced a slight improvement of PV in soy extrudates and of NPU and BW in rye extrudates. The results indicate that the extrusion at low water content and moderate temperature can safely be used to process ground soy bean and whole rye meal without and damage to the nutritional protein quality.

The presence of steroids in larval and mature brook lampreys (Lampetra planeri Bloch).

The presence of steroids was investigated by radioimmunoassay - mass spectrometry in serum and homogenates of gonads and presumed adrenocortical (pro- and opisthonephric) tissue of larval and mature brook lampreys (Lampetra planeri Bloch) of both sexes. Pregnenolone was detected in all tissue samples studied whereas androstenedione could be identified only in presumed adrenocortical tissue from male and female mature animals as well as in testis. Testosterone was present in testis of adult lampreys and in sera of larval and adult animals.

Spectrophotometric analyses of hydroxysteroid dehydrogenase activity in presumed steroid-producing tissues of the brook lamprey (Lampetra planeri bloch) in different developmental stages.

Hydroxysteroid dehydrogenase (HSD) activity was determined spectrophotometrically on homogenates of the pronephros, the opisthonephros, and the gonads of metamorphosing, immature, and mature adult male and female brook lampreys (Lampetra planeri Bloch), employing pregnenolone, dehydroepiandrosterone, androsterone, epiandrosterone, and 3 beta,17 beta-dihydroxy-5 alpha-androstane as substrates and NAD or NADP as coenzymes in October, December, February, and April. With NADP a higher HSD-dependent HSD activity was detected in presumed adrenocortical tissue (PAT) homogenates with pregnenolone and 3 beta,17 beta-dihydroxy-5 alpha-androstane and in gonad homogenates only with 3 beta,17 beta-dihydroxy-5 alpha-androstane. With 3 beta,17 beta-dihydroxy-5 alpha-androstane as substrate the highest enzyme activities were measured in PAT and gonad homogenates from animals of all investigated life stages. A varying intensity of HSD activity in the examined tissue homogenates probably dependent on stages of the individual development was observed.

Histochemical and spectrophotometric demonstration of hydroxysteroid dehydrogenase activity in the presumed steroid producing cells of the brook lamprey (Lampetra planeri Bloch) during metamorphosis.

Pronephric, opisthonephric, ovarian, and testicular tissue rich in presumed steroid producing cells taken from the brook lamprey during metamorphosis was investigated by histochemical and spectrophotometric methods. Histochemical results seemed to provide evidence for hydroxysteroid dehydrogenase activity using pregnenolone as substrate. Spectrophotometric evidence for hydroxysteroid dehydrogenase activity was obtained from presumed adrenocortical tissue, ovarian and testicular homogenates using pregnenolone, dehydroepiandrosterone, androsterone, and 3 beta, 17 beta-dihydroxy-5 alpha-androstane as substrates.

Iron deficiency in the rat. Physiological and biochemical studies of muscle dysfunction.

Work performance on a treadmill has been evaluated in normal and iron-deficient rats. Anemia was removed as a variable by adjusting the hemoglobin of all animals to the same concentration. At a hemoglobin compatible with normal work performance, iron-deficient animals showed a marked impairment of running ability as compared to control animals. Iron therapy corrected the disability within 4 days. Concentrations of the cytochrome pigments and myoglobin, and rates of oxidative phosphorylation with pyruvate-malate, succinate, and alpha-glycerophosphate as substrates were all reduced in mitochondrial preparations from skeletal muscle of iron-deficient rats, but only the rate of phosphorylation with alpha-glycerophosphate as substrate increased significantly and in parallel with the recovery in work performance of the iron-deficient rats treated with iron dextran.
