Development of gastric somatostatin-like immunoreactivity in response to corticosterone acetate and dietary changes in young rats.

The effects of dietary changes from breast milk to solid food and corticosterone acetate administration on somatostatin-like immunoreactivity (SLI) in the gastric antrum and corpus were studied in young and adult rats. At 7 days of age, a single dose of corticosterone acetate (250 mg/kg of body weight) was injected in one group of rats, and a single dose of physiological saline was injected in another group. Each group was divided into two subgroups; one was fed only rat breast milk until 25 days of age and the other weaned (solid food alone) at 21 days of age. Gastric antrums and corpora were removed at various ages for assay of SLI concentrations by radioimmunoassay. In non-corticosterone treated rats, antral and corpus SLI increased gradually until 20 days of age, and after changing from breast milk to solid food on day 21 antral SLI increased 2-fold to the adult level, whereas corpus SLI remained constant. In corticosterone-treated rats, antral and corpus SLI on day 11 was 5-fold that on day 7, and thereafter remained constant before changing from breast milk to solid food; after weaning (solid food alone) antral SLI increased again to reach the adult level at 25 days of age whereas corpus SLI remained constant after weaning. Prolonged breast milk feeding alone did not influence antral or corpus SLI levels in either corticosterone-treated or nontreated rats as compared to levels on day 20.(ABSTRACT TRUNCATED AT 250 WORDS)

[Endoscopic administration of OK-432 in gastric cancer].

This paper describes the morphological and histological changes of gastric cancer following OK-432 injection into and around the lesion under endoscopic observation. Of 10 cases of unresectable gastric cancer, 7 revealed morphological changes and 3 a significant reduction of tumor size. In one of these cases, all the tumor cells disappeared. In 7 cases of resected gastric cancer preceded preoperatively by OK-432 administration, no obvious morphological changes were observed. However, histologically, marked infiltration of inflammatory cells was observed at the site of injection, especially in the submucosal layer. In regional lymph nodes, a lymphocyte predominance pattern which may have resulted from augmentation of T-cell activity was observed in many cases. The results suggested that local OK-432 injection therapy does not have such a marked direct effect on tumor cells, but may be useful as a means of prevention against invasion into the submucosal layer and regional lymph nodes.

Development of antral gastrin-like immunoreactivity and pituitary CCK8/gastrin-like immunoreactivity in rats.

Gel filtration of antral extract of adult rats revealed gastrin-17 and gastrin-34. Gel filtration of anterior pituitary extract showed CCK8 and gastrin-17, whereas posterior pituitary extract showed only a CCK8. Antral gastrin-like immunoreactivity (G-LI) increased after milk feeding and especially after weaning was started. Changes in diet may exert a profound influence on the ontogenic development of antral G-LI, but not pituitary CCK8/G-LI.

Gastrin/cholecystokinin-like immunoreactivity in human blood cells.

In the gastrin and/or cholecystokinin-like immunoreactivity (G/CCK-LI) elution patterns of blood cells in human adults, erythrocyte (RBC) elution pattern has three peaks which are coeluted with gastrin-34 (G34), gastrin-17 (G17) and Vt, and polymorphonuclear leukocyte (PMN) and mononuclear cell (MNC) elution patterns have four peaks which are coeluted with Vo, G34, G17 and Vt. The content of G/CCK-LI in RBC is 1.20 +/- 0.54 f mole/10(8) cells (mean +/- SD). That in PMN and MNC is 1.44 +/- 0.67 p mole/10(8) cells and 1.67 +/- 0.76 p mole/10(8) cells, respectively.

Beta-endorphinlike immunoreactivity and somatostatinlike immunoreactivity in normal gastric mucosa, muscle layer, and adenocarcinoma.

beta-Endorphinlike immunoreactivity and somatostatinlike immunoreactivity were detected in the mucosa and muscle layer of normal gastric antrum and corpus and in moderately differentiated adenocarcinoma derived from the antral mucosa. The concentration of beta-endorphinlike immunoreactivity in the normal gastric tissues was 4-15 pmol/g wet wt tissue; this value varied from 9.64 to 241.39 pmol/g wet wt tissue (81.38 +/- 37.82 pmol/g wet wt tissue) in adenocarcinomas. The concentration of somatostatinlike immunoreactivity was 18-25 pmol/g wet wt tissue in normal gastric mucosa, whereas it was 1-2 pmol/g wet wt tissue in adenocarcinomas. Gel exclusion chromatography of beta-endorphinlike immunoreactivity revealed two peaks corresponding to beta-endorphin and beta-lipotropin. In normal mucosa and in the whole layer of antrum, the major peak was eluted near the position of beta-lipotropin, and the minor broad peak was eluted near the position of beta-endorphin. In contrast, in adenocarcinoma, beta-endorphinlike immunoreactivity was eluted broadly at the position of beta-endorphin and the other smaller peak was at the position of beta-lipotropin. Gel exculsion chromatography of somatostatinlike immunoreactivity also showed different patterns between antral mucosa and adenocarcinoma. This study revealed the presence of the opioid peptide, beta-endorphinlike immunoreactivity, not only in normal gastric tissue but also in adenocarcinomas with highly increased concentration and different elution patterns in combination with decreased concentration of somatostatinlike immunoreactivity.

Influence of diet on the development of gastrin- and somatostatin-like immunoreactivity in the stomach of rats.

The effects of changes in diet on antral gastrin-like immunoreactivities and antral and corporal somatostatin-like immunoreactivities were studied in three groups of young male rats. Group 1 rats were weaned physiologically, group 2 rats received only breast milk, and group 3 rats were fed milk until 20 days of age and then were switched abruptly to laboratory chow. In group 1, antral gastrin-like immunoreactivity and somatostatin-like immunoreactivity increased gradually from 13 days of age to adults. In group 2, antral gastrin-like immunoreactivity did not increase, and antral and corporal somatostatin-like immunoreactivity increased until 20 and 25 days of age, respectively. In group 3, there were three- to fivefold increases of antral gastrin-like immunoreactivity and somatostatin-like immunoreactivity after changing to laboratory chow. Dietary changes had no effect on the concentration of corporal somatostatin-like immunoreactivity. These results suggest that the developmental patterns of gastric gastrin-like immunoreactivity and somatostatin-like immunoreactivity are closely related to diet; laboratory chow may play an important role in the development of these gut hormones.

Scirrhous argyrophil cell carcinoma of the stomach with multiple production of polypeptide hormones, amine, CEA, lysozyme, and HCG.

Sixteen argyrophil cell carcinomas in 59 gastric scirrhous carcinomas were examined histologically, ultrastructurally, and immunohistochemically for polypeptide hormones, CEA, lysozyme, and HCG. In nine of these 16 tumors, polypeptides such as gastrin, somatostatin, and glucagon were demonstrated. Six of these nine tumors contained all three hormones, and three of these six tumors also had argentaffin cells. In all of these 16 tumors CEA were observed. Eight of them had CEA, lysozyme, and acid mucin synchronously. Of the above six tumors containing three peptides, three produced focal HCG. Ultrastructurally, several types of secretory granules were noted. Histologically, these 16 tumors showed poorly differentiated adenocarcinomas or signet ring cell carcinomas. Macroscopically, generalized type was 11 and localized type five. No hormonal syndrome was detected in any of the patients. It was suggested that these scirrhous argyrophil cell carcinomas of the stomach with the multifunction originate from totipotent immature cells of endodermal origin.
