Research related to the herbicide buvinol, especially for possible carcinogenicity.

The Budapest Chemical Works (Hungary) has conducted a study on a new herbicide called 'Buvinol', containing 2,4,5-trichlorophenoxyethanol (TCPE). In long-term carcinogenicity testing, the oral administration of the maximum tolerated dose of TCPE (70 mg/kg), containing 0.1 ppm 2,3,7,8-tetrachlorodibenzo-para-dioxin, increased the incidence of liver tumours in male mice, while smaller doses (1/10 and 1/100) did not. Dioxin in the dose range used (0.1--10 ppm) did not influence tumour frequency. Exposure of workers to TCPE during its production or use should be controlled and reduced.

[Classical and modified intramedullary nailing in osteomyelitis pseudoarthroses].

13 cases of osteomyelitic pseudarthroses following injury were treated by the authors, by means of intramedullary nailing. 5 of these patients already cured are introduced in detail in the article. Intramedullary nailing in the classic manner and its modification, using bolts as well, were performed alike. The authors have a favourable impression after these series. No septic complication occured. Intramedullary nailing is a good method in the management of this serious sequel to fracture treatment.

Study of the antitumoral activity of S-carbamoyl-L-cysteine derivatives in animal experiments.

The antitumoral activity of three amino acid derivatives, S-carbamoyl-L-cysteine, S-ethyl-carbamoyl-L-cysteine, and S-chloroethyl-carbamoyl-L-cysteine, was studied. The ethyl and chloroethyl derivatives had a pronounced curative effect on certain animal tumors. It is interesting in the case of S-carbamoyl-L-cysteine that N-ethylization of the substance resulted in increased chemotherapeutic effectivity with no simultaneous change of the toxicity. According to various studies this effect is different in many respects from that of the known cytostatics, and a possibility of a specific selective or surface activity is raised.

Carcinogenic bioassay of the herbicide, 2,4,5-trichlorophenoxyethanol (TCPE) with different 2,3,7,8-tetrachlorodibenzo-p-dioxin (dioxin) content in Swiss mice.

The carcinogenic effect of a herbicide trichlorophenoxyethanol (TCPE) was investigated. The substance always contains 2,3,7,8-tetrachlorodibenzo-p-dioxin (dioxin) as a trace contaminant. Outbred Swiss-H/Riop mice were treated for 1 year with different doses of the various admixtures of the two compounds and those of dioxin alone to determine whether their carcinogenic effects are dose-dependent. Observation time after TCPE treatment was 1 and 2 years. The maximum tolerable dose of TCPE (70 mg/kg) with a dioxin concentration of 7.10(-6) mg/kg doubled the number of liver tumours as compared to the controls. The occurrence of liver tumours decreased at the administration of lower doses of the two compounds. No change in tumour frequency could be detected when the level of dioxin as a trace contaminant was raised, either (10 ppm). According to our preliminary results, dioxin has a more toxic than carcinogenic effect. The use of TCPE as a herbicide in accordance with the proper factory regulations and the consumption of products treated with it does not seem to bear a carcinogenic risk for the consumer.

Ten years domestic salt fluoridation in Hungary.

Results are presented regarding the caries reduction achieved over a 10 yr period using 250 mg F per kg of domestic salt, and compared with data from a fluoridated water area and a control group. The frequency of caries-free children in the 4 to 6, 7 to 11 and 12 to 14 age range in the experimental group rose from 8.06%, 4.74% and 1.43% to 46.75%, 43.98% and 7.92% respectively, and dmft and DMFT indices dropped from 5.35, 3.62 and 6.60 to 2.80, 1.45 and 3.65. There was no significant change in the percentage of caries-free children or in the indices of the control group. The ratio of dmft and DMFT to teeth and the differences in the extent of caries among specified teeth are also presented. The data support the assumption that fluoridated salt prevents dental caries both in deciduous and permanent dentitions. The results were similar as those obtained in a water fluoride area. The optimum and tolerated F intake water and salt ingestion, F intake from foods and the urinary F levels are also discussed.

Fluorine content of vegetables and fruits.

Twenty-six species of vegetables grown in Szeged, in its environment and at Szabadkigyós were examined for their F content. The mean F content of the most important staple products is 0.30 mg/kg raw weight or less. When vegetables of the same kind were examined, the results were mostly different. The highest mean F content was obtained for spinach (0.55 mg); potato yielded less (0.15 mg), and the lowest was obtained for cucumber (0.06 mg). The mean F content of the 13 fruit species was 0.10 mg/kg. The highest F content was obtained for pears (0.23 mg). Only traces of fluorine were observed in sweet cherry, cherry and water melon.

Fluorine content of foods and the estimated daily intake from foods.

The F content of rice, rye, bread, wheat flour, rolls and wheat bread was found to be: 0.42, 0.31, 0.28, 0.28 and 0.22 mg/kg raw weight, respectively. Among foods of animal origin the F content of fish was the highest: mackerel in oil 7.28 mg, and carp 2.02 mg/kg. Mutton contained 0.71, meat tissues of pig 0.36, other meats and meat tissues less than 0.36, while egg only 0.12 mg/kg raw weight. Eight different dishes of soups, nine cooked vegetables and sauces, and ten other dishes of cooked food prepared with F-poor water contained: 0.18, 0.20 and 0.26 mg/kg, respectively. Calculations were made to determine the amount of F ingested daily with foods. Mean F ingestion of the population was found to be 0.314 mg daily with a food supplying 3000 calories.

Effect of dianhydrodulcitol (DAD) on the primary immune response of normal and tumor bearing rats.

The effect of dianhydrodulcithol (DAD) administered in a dose of 5 mg/kg on the number of plaque-forming cells (PFC) in spleens and on the titre of hemolytic antibodies in the sera of rats immunised with sheep red blood cells (SRBC) have been examined. The compound elicited a short-lasting suppressive effect when applied after immunisation; the number of PFC decreased two days after treatment while 5 days after drug administration increased as compared to the untreated animals. Treatment prior to immunisation proved to be ineffective. The titre of circulating antibodies showed changes similar although of lesser extent. Five days following treatment marked infiltration of plasmacytes was present in the axillary lymph nodes. Rats bearing transplantable Guérin tumor showed similar changes in immune response as the tumor free animals following DAD treatment.

[Consumption of drinking water by workers in a steel foundry during working hours in Hungary].

In a former paper the authors had investigated the fluorine intake per kilogram body weight and its extreme possibilities. This study includes 97 workers of a steel foundry. The amount of liquid ingested during working hours was measured, for 30 persons in summer during 90 days and 67 persons in winter during 201 days. Inside and outside temperatures were recorded. The mean age was 40,02 years. The mean intake of drinking water was, in summer 3732 ml (+/- s 1176 and +/- sx 123 ml) and in winter 1835 ml (+/- s 684 and +/- sx 48 ml). The mean value was 2422 ml and the extreme values were 500 and 8500 ml. The calculation of the individual mean values showed however, that the water intake follows an irregular pattern of distribution. About 11% consumed more than 41, and about 2% more than 51 per day continuously. In this study, there are those who consume habitually little, more and a lot of drinking water. With the results, the authors are examining the possibilities of the maximum fluorine intake.
