Ultrastructural analysis of central serotoninergic neurons immunolabeled by silver-gold-intensified diaminobenzidine chromogen. Completion of immunocytochemistry with X-ray microanalysis.

Serotonin immunoreactive structures of the rat central nervous system (CNS) were detected by the recently developed silver-intensified peroxidase-antiperoxidase complex (SI-PAP) method at both the light and electron microscopic levels. The silver postintensification of the diaminobenzidine (DAB) chromogen increased the sensitivity of the original PAP method, resulting in a very Golgi-like appearance of serotonin-immunopositive neuronal elements. The metallic silver and gold deposited onto DAB-labeled organelles, filling out the whole immunoreactive neuron, assures the easy tracing of thin neuronal processes far from the cell body. At the ultrastructural level, metallic grains were seen over immunolabeled structures only, proving the specificity of the silver method. In neurons of the dorsal raphe nucleus, free ribosomes, endoplasmic reticulum, and granules (80-100 nm in diameter) were labeled. Immunoreactive, e.g., serotoninergic, dendrites were seen to receive afferent terminals. The increased electron density of the intensified immunolabel facilitates the ultrastructural recognition of even weakly labeled profiles, while its metallic components (Ag and Au) provide a base for X-ray analysis of the immunolabeled biological specimen.

The odontoblast process and its branches in human teeth observed by scanning electron microscopy.

Healthy teeth were fixed, resin-impregnated and freeze-fractured. Specimens were critical point-dried and examined by scanning electron microscopy. Odontoblast processes were observed in the inner-, middle- and peripheral dentine. The processes in the inner-third dentine closely approximated tubule wall and could be found in most of the tubules. Some tubules contained a thin fibre besides the odontoblast process. In the middle third of dentine, the processes became thinner, their appearance was cable-like with lateral branches and a wide periodontoblast space could be seen. Processes were seen only in a few tubules of outer-third dentine in both young and aged subjects. They divided into longitudinal and lateral branches and seemed to be connected with one another.

Ultrastructural damage of crural muscle in post-thrombotic syndrome.

The fine structure of crural muscles in 11 patients suffering from post-thrombotic syndrome was investigated. The contractile elements and the mitochondria were mainly involved in the pathological changes. Glycogen granules in increased number and strong lipid vacuolization were shown. The focal occurrence of damages has to be emphasized. Beside, ultrastructural changes are induced by hypoxia and by other factors. It can be established that limited functional capacity of the lower extremities in post-thrombotic syndrome is due not only to chronic haemodynamic insufficiency but to the serious fine-structural damage.

Ultrastructural consequences of reperfusion of the ischaemic myocardium.

Ultrastructural changes following ischaemic myocardial injury have been investigated in dogs. After 15 to 120 min global normothermic ischaemia the hearts were reperfused in a special heart-lung model for 60 min and then subjected to electron microscopic examination. The degree of morphological alterations and the duration of ischaemia were approximately interrelated. The ischaemic hearts showed characteristic changes such as mitochondrial, sarcolemmal and myofibrillar defects (swelling, disruption of cristae, loss of matrical density and contractile bands, myofibrillar rupture and lysis, blurred Z-lines). The hypercontractions are no artifacts but consequences of irreversible cellular injury and the observed stiffness during reperfusion is a good marker of the myocardial damage suffered.

Adult-onset rapidly progressive spinal muscular atrophy of shoulder girdle with gammopathy.

The paper reports on clinical and pathologic findings in a 51-year-old man who presented with rapidly progressive spinal muscular atrophy associated with relatively early respiratory paralysis. Clinical diagnosis was based on typical EMG, and bioptic and enzyme findings. In the CSF protein and cell content were normal, the IgG concentration was high, the IgG/Alb quotient increased, intrathecal synthesis of IgG was evident, and oligoclonal gammopathy was found. All of these findings generally appear in neuroallergic diseases. Total loss of anterior-horn cells, demyelination of fasciculus cuneatus and spinothalamic pathways, as well as round Lafora-body like inclusions, preponderantly in the white matter, were observed with light microscopy. Electron microscopic examination revealed reoviruses in the glia cells of the damaged motor zone.

Ultrastructural changes of the subendocardium in ischemic and cardioplegic states before and after reperfusion.

Unprotected and preserved myocardial ultrastructures were analyzed in prepump and postreperfused states. The experiments were divided into three groups: group 1 - normal hearts functioning 60 min in a special heart-lung model; group 2 - hearts with 30, 60, 90 and 120 min of normothermic ischemia plus 60 min of recirculation; group 3 - hearts with 30, 60, 90 and 120 min of hypothermic cardioplegia plus 60 min of recirculation. Anoxic hearts could be set in motion, but after longer anoxia the symptoms of stone-heart regularly appeared. Reperfusion induced grave ultrastructural changes both in the energy system (mitochondrial edema, rupture and lysis of cristae) and myofibrils (hypercontraction, elongation, Z line anomalies). Cardioplegic hearts showed a dynamic recovery. However, after reperfusion, characteristic sporadic signs of ischemia could always be demonstrated.

X-ray microprobe analysis of chloragosomes of untreated and of EDTA-treated Lumbricus terrestris by using fresh air-dried smears.

The chloragosomes of Lumbricus terrestris were analysed by an electron microscope X-ray microphobe, using both air-dried smear preparations and conventional electron microscopic sections. In untreated worms the air-dried chloragosomes showed prominent peaks for Ca and P, distinct peaks for S and Zn, while Na, K, and Cl peaks were barely detectable. In air-dried smears the S peak was constantly more prominent than in EM sections, indicating that chloragosomes contain easily soluble S-compounds. By long-term CaNa2EDTA treatment of the worms, the Ca and P content of the chloragosomes could be significantly decreased, empirically proving, that the storage and mobilisation of chloragosomal Ca and P are closely related.

[Ultrastructural investigation on muscles of patients with talipes equinovarus (author's transl)].

Authors in course of soft tissue release of children with talipes equinovarus between the age of 9 months and 4 years performed a muscle biopsy in 23 cases from the so called club foot muscles and from the peronei. The results of electronmicroscopic investigation are described. The muscle is not as a hole affected, but the pathologic parts are scattered among the intact ones. Fatty degeneration was noted after long termed redression in plaster, together with the fat connective tissue also appeared. The most severe alterations were observed on the contractile elements, consistent seemingly with neuromuscular atrophy. The posterior tibial muscle and the peronei revealed the most obvious alterations. With rising age there was no deterioration of the picture. On basis of their results it is suggested, that for children starting to walk all kind of treatment involving immobilisation should be abandoned, and after operation a functional treatment with eventual electrotherapy introduced. This is in their opinion only realizable in frame of organized care.

Swelling of nuclei and decondensation of chromatin in a multinuclear model.

Rat liver nuclei were isolated in the presence of a nonionic detergent, and the soluble proteins were extracted from them using low-salt solution. The isolated nuclei were then embedded in a fibrin clot which was cut into 4 to 5 mm3 pieces and placed into an AMICON pressure cell in the presence and absence of the soluble nuclear proteins. When the soluble nuclear proteins were present and when their concentrations reached 40 mg/ml, the nuclei were significantly larger and their chromatin more decondensed than in the absence of added proteins. Our findings suggest that the soluble nuclear proteins play a role in the regulation of nuclear volume and the hydration of chromatin.

X-ray microanalytical studies on native myofibrils and mitochondria isolated by microdissection from honey-bee flight muscle.

The distribution of elements was studied by means of energy dispersive X ray microanalysis in the honey-bee flight muscle. Semi-quantitative analysis of bulk muscle specimen, isolated mitochondria and isolated myofibrils either in resting or stretched state was performed. High K peak was observed in the spectra of bulk muscle specimen resembling the spectra of myofibrils and high P peak resembling the spectra of mitochondria. In the myofibrils no substantial difference was found between the spectra of A bands and Z lines, but they contained much higher K peak than the spectra of I bands.

Effect of prostaglandin E2 on the permeability of the iris vessels to horseradish peroxidase in the rabbit.

Prostaglandin E2 (0.5 microgram in 10 microliter 10% ethanol) was introduced into the anterior chambers of rabbit eyes. Using the horseradish peroxidase method, it was shown under the electron microscope that the endothelial barrier of the iris vessels broke down. The peroxidase penetrated as far as the basis of the posterior epithelial cells, however, without entering their lateral intercellular spaces. The question of whether the effect of prostaglandins on the barrier was a direct effect or at least a partially indirect one, i.e., a haemodynamic action, is discussed.

The Z line of the flight muscle of honey-bee.

The size and arrangement of the hexagonal filament lattice in the Z line of the flight muscle of honey-bee is similar to the size and arrangement of the lattice of A filaments. Z filaments show a "tubular" structure and possess a seemingly empty inner core just like A filaments do. The Z filaments are connected with a thick network transversally. The I filaments are probably embedded in this transversal network on both sides of the Z line; they cannot be recognized inside the Z line. The filament lattice of the Z line does not exclude the possibility of the A filaments to be connected to the Z line through the connecting filaments, or continuously to pass from one sarcomer into the other.
