Due to the action of light and oxidation some drugs applied to and developed on a thin layer give a colour reaction. As a result of photooxidation artefacts may develop directly on the thin layer.
For colour identification of drugs on a thin layer bromine vapours were used. The investigated drugs were divided into those which after bromination gave a colour reaction and those which did not display this reaction.
In order to assess which drugs give a colour reaction on TLC after chlorination and subsequent heating, more than 110 drugs were investigated. It was revealed that the investigated drugs can be divided into four groups: positive after short-term chlorination positive after prolonged chlorination, positive after prolonged chlorination and heating to 150 degrees C, and negative.
Many a drug was identified by thin layer chromatography when using tetramethyldiaminodiphenylmethan (TDM) as a universal reagent subsequently to chlorination. This non-carcinogenic compound yields a characteristical tint with all the drugs and their metabolites.
For detection of some selected drugs on a thin-layer sprinkling with water was used. White, well defined spots develop on a wet background, which after drying can be further identified by a specific colour reaction or extract for photometric identification.
The authors give an account of practical experience with chromatography of drugs using HPTLC. As an indicator berberin hydrochloride or sulphate proved useful. The sandwich method was satisfactory as regards reproducibility of Rf values.
The authors discuss detection of drugs and other substances during thin layer chromatography by means of an aqueous solution of potassium permanganate. Ninety drugs and other substances were divided into three groups: a) strongly reacting, b) weakly reacting, c) not reacting. It was revealed that all compounds containing non-aromatic C = C bonds or oxidizable sulphur react very intensely. Other drugs containing readily oxidized groups react also.

Authors succeeded in proving an insecticide Lannate 90 WS in coffee grounds by mass spectrometry and thin-layer chromatography . The compound was later found in a table located in the building where intoxication had occurred and more persons had suffered from a sudden disease with similar signs previously.
