Aggressiveness of SJL/J lymphomas correlates with absence of H-2Ds antigens.

SJL/J lymphomas emerged as H-2Ds-negative variants upon transplantation of their respective H-2Ds-positive spontaneous tumors. Of nine spontaneous SJL/J lymphomas examined, only two contained detectable numbers of H-2Ds-negative tumor cells. Of seven successfully transplanted tumors, however, four emerged as predominantly H-2Ds-negative tumors by their second transplantation. Long term SJL/J lymphoma lines, RCS 5 and SJL/SJL, were also found to be H-2Ds-negative. The expression of H-2Ks and I-As were maintained on all tumor lines examined. H-2Ds-negative tumor cells were consistent stimulators of syngeneic lymphoproliferation, whereas tumors maintaining H-2Ds expression varied in their stimulatory capacity. Transplantation times required to achieve morbidity in syngeneic mice correlated with tumor cell expression of H-2Ds. Tumors maintaining H-2Ds expression were less aggressive and required 57 to 150 days to produce morbidity compared with highly aggressive H-2Ds-negative tumors that required only 7 to 10 days at one-quarter of the cell dose (10(7) cells). Growth curve analysis based on spleen weights (corrected for body weight) indicated that three types of SJL/J lymphomas could be identified based on their expression of H-2Ds and ability to stimulate syngeneic lymphoproliferation: 1) indolent, nonstimulatory H-2Ds-positive tumors; 2) indolent, stimulatory H-2Ds-positive tumors; and 3) highly aggressive, stimulatory H-2Ds-negative tumors.

Dominant susceptibility effect on the murine corneal response to Pseudomonas aeruginosa.

Natural host resistance to Pseudomonas aeruginosa corneal infection is regulated by two complementing dominant genes PsCR1 and PsCR2 (RS Berk, MA Leon, LD Hazlett. Infect Immun 26:1221-1223, 1979). In this study we have demonstrated a third dominant gene, which determines susceptibility to P. aeruginosa-induced eye damage. This gene was designated as PsCS. The F1 progeny from matings between the resistant DBA/2J strain and the susceptible strain C57BL/6J and C3H/HeJ displayed the susceptibility phenotype. Backcross and F2 studies using the C3H/HeJ and DBA/2J strains suggested the presence of two linked PsCS loci.

Regulation of experimental autoimmune thyroiditis: influence of non-H-2 genes.

Experiments were conducted to investigate the non-H-2 genetic effects on experimental autoimmune thyroiditis (EAT). Strains having C3H or BALB background in general produced higher autoimmune responses to mouse thyroglobulin (MTg) than the B10 or A strains. Comparisons of C3H and B10 congenic strains carrying similar H-2 haplotypes demonstrated that the C3H congenics had significantly higher MTg antibody titres and more severe thyroid damage, even when the strains carry the low responder H-2 haplotypes. These observations show that non-H-2 gene(s) influences EAT, in addition to genes in the MHC.

Intra-H-2 recombination in the mouse. III. I-Region characterization of haplotypes H-2at2, H-2at3 and H-2a2.

Serological characterization of three K-S interval recombinant strains, TBR2 (H-2at2), TBR3 (H-2at3) and AIR 1 (H-2a2) was performed using anti-H-2, Ia, Ss and Slp antisera. The data presented here reveal that the crossover events in both TBR2 and TBR3 occurred between the I-A and I-E subregions. In both cases, the H-2K and I-A subregions were derived fron the H-2t1 of chromosome, while the I-E, S and H-2D regions were derived from the H-2b chromosome (KsAkEbSbDb). The H-2a2 chromosome resulted from a crossover event between the H-2a1 and H-2i9 chromosomes. Ia and Ss typing of AIR 1 suggested that the K to I-E regions originated from H-2a1 and the S and D regions originated from H-2i9 (KkAkEkSbDd).

Intra-H-2 recombination in H-2b/H-2t1 heterozygotes in the mouse I. Four cases of crossing over between the S And D regions.

An examination of 1009 backcross animals produced from H-2b/H-2t1 heterozygotes resulted in the detection of four cases of intra-H-2 recombination. Three of the recombinants received the K region from H-2b and the D region from the H-2t1 parenteral chromosome. The fourth recombinant received a K region from the H-2t1 parental chromosome and the D region from H-2b. Ss typing revealed that crossing over occurred between the S and D regions of the H-2 complex in all four cases. Three of the four recombinants were developed into inbred strains and assigned the haplotype designations H-2i8, H-2i9 and H-2at1.
