Generation of a stable cell line producing high-titer self-inactivating lentiviral vectors.

To facilitate the generation of SIN lentivirus vector-producer cell lines, we have developed a novel conditional SIN (cSIN) lentivirus vector, which retains its SIN properties in normal target cells yet can be produced at high titers from tetracycline-regulated packaging cell lines. The design of the cSIN vector is based on replacing the vector U3 transcription regulatory elements with the Tet-responsive element, which allows vector production exclusively in cells expressing the synthetic Tet-regulated transactivator (tTA). In contrast minimal vector production ( approximately 200 IU/ml) is obtained in target cells that do not express the tTA, even in the presence of all HIV-1 proteins. Following transduction of the Tet-regulated SODk1 lentivirus vector-packaging cell line with the cSIN vector, high titers of cSIN recombinant vector (>10(6) IU/ml) could be generated, which efficiently transduced terminally differentiated neurons in normal rat brain.

Immune related genetic polymorphisms and schizophrenia among the Chinese.

Genetic association studies were conducted among two independent cohorts of Chinese ethnicity. The samples consisted of cases and unrelated controls, ascertained from Guangzhou, China, and Singapore. The studies were prompted by our earlier report of an association between schizophrenia and HLA DQB1 alleles (HLA DQB1*0602 and HLA DQB1*0303) in the Singapore sample. Polymorphisms of HLA DQB1 and flanking markers on chromosome 6p21.3 were investigated in the first part of the study. A significant negative association with HLA DQB1*0402 was detected in the Guangzhou sample (Odds ratio, OR 0.26, 95% confidence intervals, CI 0.1, 0.6; p < 0.02, corrected for multiple comparisons). Additional analysis of the Guangzhou and Singapore samples revealed associations at three other anonymous markers flanking HLA DQB1. In the second part of the study, three polymorphisms at the Interleukin-1 gene cluster (IL-1, chromosome 2q13-q21) were investigated in both cohorts, since associations with schizophrenia have been reported in another sample. Persuasive evidence for an association at IL-1 was not detected in either sample. Our results suggest a susceptibility locus for schizophrenia in the HLA region among the Chinese, but further clarification is necessary.

Trichodinid ectoparasites (ciliphora: Peritrichida) of some marine fishes from coastal regions of the Yellow Sea and Bohai Sea.

Five species of marine fishes, including two of the main maricultured fishes from coastal regions of the Yellow Sea and Bohai Sea, were examined for ectoparasitic trichodinids. A total of five species of trichodinids belonging to three genera, Trichodina Ehrenberg, 1830, Paratrichodina Lom, 1963 and Trichodinella Srámek-Husek, 1953 were reinvestigated following dry silver impregnation. These were: Trichodina rectuncinata Raabe, 1958, T. jadranica (Raabe, 1958) Haider, 1964, Paratrichodina globonuclea Lom, 1963, P. obliqua Lom, 1963 and Trichodinella lomi Xu, Song & Warren, 1999. Morphometric data and comparative descriptions of these trichodinids are provided along with details of their prevalence and intensity of infestation.

Nuclear factor kappaB is a critical determinant in N-methyl-D-aspartate receptor-mediated neuroprotection.

The role of a nuclear factor kappaB (NF-kappaB) in NMDA receptor-mediated neuroprotection is not known. A candidate sequence from the 5' flanking region of exon 3 of the rat brain-derived neurotrophic factor (BDNF) gene was used to show that exposure of rat cerebellar granule cells to 100 microM NMDA activated a specific DNA binding activity that was blocked by the NMDA receptor antagonist MK-801. Anti-p65 antibody or anti-p50 antibody 'supershifted' the DNA binding activity, suggesting that the DNA-protein complex was composed of p65 and p50 subunits. NMDA receptor-mediated neuroprotection was blocked when cerebellar neurons were transfected with a double-stranded oligonucleotide containing the BDNF gene NF-kappaB sequence. Furthermore, nuclear extracts prepared from neurons treated with NMDA and the double-stranded NF-kappaB oligonucleotide showed reduced DNA binding activity to the target sequence, supporting the idea that NF-kappaB may be involved in the transcriptional activation of the BDNF gene. To address this issue, we quantified the level of exon 3-specific BDNF mRNA. Relative to GAPDH mRNA levels and compared with untreated neurons, NMDA increased exon 3-specific BDNF mRNA twofold. In contrast, pretreatment of neurons with the NF-kappaB target DNA abolished the increase in BDNF mRNA following addition of NMDA. We also determined that BDNF itself induced an NF-kappaB DNA binding activity. Taken together, these data support a mechanism where NF-kappaB plays a critical role in NMDA-mediated neuroprotection.

Independent and synergistic effects of interleukin-18 and interleukin-12 in augmenting cytotoxic T lymphocyte responses and IFN-gamma production in aging.

Aged mice exhibit diminished CD8(+) cytotoxic T lymphocyte (CTL) response to influenza virus. Previously, interleukin-12 (IL-12) was shown to partially restore in vitro influenza virus-specific CD8(+) CTL activity and interferon-gamma (IFN-gamma) production in aged mice. The present study investigated IL-18 production and its ability to collaborate with IL-12 to enhance these responses to the levels of young mice. IL-18 protein production and mRNA expression in influenza virus-specific CTL from aged mice were higher than from young mice. In contrast, IL-18 receptor (IL-18R) mRNA expression was significantly reduced in CD8(+) CTL from aged mice. Generation of CTL in the presence of IL-12 alone caused a significant increase in IFN-gamma production in both old and young mice. IL-18 treatment alone significantly increased IFN-gamma in CTL from young but not old mice. However, a combination of IL-18 and IL-12 significantly increased IFN-gamma in both old and young mice. IL-18 and IL-12, either alone or in combination, stimulated significant influenza virus-specific cytotoxicity in both old and young mice, but no significant synergistic effect was observed. These results represent an initial demonstration of downregulated IL-18R expression in aging mice and are consistent with age-related cytotoxic T lymphocyte 1 (Tc1) deficiency. Potentially, IL-18 and IL-12 can augment IFN-gamma production and reverse CD8(+) CTL deficiency in aging, independently or synergistically.

Effects of a modified sandblasting surface treatment on topographic and chemical properties of titanium surface.

A modified sandblasting surface treatment (a noncoating, roughening surface modification of dental implants) has been developed that will overcome the defects of conventional coating techniques. To verify the feasibility and reliability of this method at the chemical and topographic levels, scanning electron microscopy, x-ray diffraction, and a titanium ion releasing test were used; the topography of titanium surface, the embedding of sandblasting particles, nonpollution of heteroelements, and anticorrosiveness of titanium were criteria. Results showed that the rough surface created by sandblast was rather irregular, full of sharp tips and many embedded sandblast particles, and its corrosive rate was increased. These characteristics were modified by oxalic acid attack; the contour of the rough surface became more regular and round, the embedded particles and the heteroelement pollution were thoroughly removed, and the Ti corrosive rate decreased dramatically. Oxalic acid attack modification also created numerous secondary micropores (2.0-micron diameter) on the basis of sandblasted surface macrotexture. This modified sandblasting surface treatment is feasible and reliable to apply to dental implants and does not decrease the biocompatibility of titanium.

DNA-binding activity of amino-terminal domains of the Bacillus subtilis AbrB protein.

Two truncated variants of AbrB, comprising either its first 53 (AbrBN53) or first 55 (AbrBN55) amino acid residues, were constructed and purified. Noncovalently linked homodimers of the truncated variants exhibited very weak DNA-binding activity. Cross-linking AbrBN55 dimers into tetramers and higher-order multimers (via disulfide bonding between penultimate cysteine residues) resulted in proteins having DNA-binding affinity comparable to and DNA-binding specificity identical to those of intact, wild-type AbrB. These results indicate that the DNA recognition and specificity determinants of AbrB binding lie solely within its N-terminal amino acid sequence.

DNA melting analysis for detection of single nucleotide polymorphisms.

BACKGROUND: Several methods for detection of single nucleotide polymorphisms (SNPs; e.g., denaturing gradient gel electrophoresis and denaturing HPLC) are indirectly based on the principle of differential melting of heteroduplex DNA. We present a method for detecting SNPs that is directly based on this principle. METHODS: We used a double-stranded DNA-specific fluorescent dye, SYBR Green I (SYBR) in an efficient system (PE 7700 Sequence Detector) in which DNA melting was controlled and monitored in a 96-well plate format. We measured the decrease in fluorescence intensity that accompanied DNA duplex denaturation, evaluating the effects of fragment length, dye concentration, DNA concentration, and sequence context using four naturally occurring polymorphisms (three SNPs and a single-base deletion/insertion). RESULTS: DNA melting analysis (DM) was used successfully for variant detection, and we also discovered two previously unknown SNPs by this approach. Concentrations of DNA amplicons were readily monitored by SYBR fluorescence, and DNA amplicon concentrations were highly reproducible, with a CV of 2.6%. We readily detected differences in the melting temperature between homoduplex and heteroduplex fragments 15-167 bp in length and differing by only a single nucleotide substitution. CONCLUSIONS: The efficiency and sensitivity of DMA make it highly suitable for the large-scale detection of sequence variants.

[Association analysis of attempted suicide and 5-HT2A receptor gene].

OBJECTIVE: To examine the relationship of attempted suicide with A-1438G and T102C polymorphisms of 5-HT2A receptor gene. METHODS: Genotypes and allele frequencies of A-1438G and T102C polymorphisms of 5-HT2A receptor gene in 149 attempted suicides and 190 normal controls were examined by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP). RESULTS: No differences in genotypes and allele frequencies of A-1438G and T102C polymorphisms of 5-HT2A receptor gene were observed between attempted suicides and the controls. However, an association between male attempted suicides and genotype(GG) of A-1438G polymorphism was found. CONCLUSION: These results suggested that liability to suicide behavior might be associated with A-1438G polymorpyhism of 5-HT2A receptor gene in male subjects.

[Wet synthesis and characterization of fluoride-substituted hydroxyapatite].

Hydroxyapatite and fluoride-substituted hydroxyapatite powders are synthesized by wet method, compacted into pellets and subjected to heat treatment at 300, 600, 900 degrees C respectively. Analysis of the fluoride-substituted hydroxyapatites and HA by FTIR spectroscopy indicated that with the increasing of sintering temperatures, the phase-pure fluoride-substituted hydroxyapatite and stoichiometric hydroxyapatite could be got. According to the X-ray diffraction analysis, the fluoride-substituted hydroxyapatite is less stable for decomposing into beta-Ca3(PO4)2 at the temperature of 900 degrees C. The in vitro bioactivity of the fluoride-substituted hydroxyapatites and hydroxyapatite heat treated at 900 degrees C was assessed by immersion testing in simulated body fluid (SBF). Both scanning electron microscopy (SEM) analysis and ionic solution changes (Ca2+) demonstrated that the low fluoride content apatite could induce a surface bonelike apatite layer formation.

[A new briquetting method for biomass coal and its influencing factors].

Regarding to the limitation and low economical efficiency of conventional biomass coal briquetter determined by its briquetting method, a new briquetting mechanism using the linkage function of biomass fiber was established. The effect of different coal, briquetting pressure, biomass shape and content, water content on new brequtting mechanism was studied preliminarily. The results showed that the new mechanism was suitable for varied kinds coal and biomass. The briquette mechanical performance was improved as biomass content increased with a reasonable low limitation of about 15%. Pure biomass also could be briquetted. The briquetting pressure which could consolidate the linkage of biomass fiber was no more than 120 MPa. There would be a negative effect at too high pressure. Increasing biomass content and decreasing pressure at certain degree could offset the negative effect of water content significantly, with a permissive limitation over 10% for Datong coal.

[Study on graft-versus-leukemia effects of donor lymphocyte infusion after nonmyeloablative allogeneic bone marrow transplantation].

OBJECTIVE: To explore the graft-versus-leukemia effects of donor lymphocyte infusion(DLI) after nonmyeloablative allogeneic bone marrow transplantation while attenuating treatment-associated morbidity, mortality and graft-versus-host disease. METHODS: 615(H-2k) mice were loaded with L615 cells and 3 days later received total body irradiation (TBI) 5 Gy (60 Co gamma-ray) followed by allogeneic bone marrow transplantation (allo-BMT). The allo-grafts consisted of 3 x 10(7) bone marrow cells and 1 x 10(7) spleen cells from BALB/c (H-2d) donor mice. Two days after allo-BMT, the recipient mice were given 200 mg/kg of CTX. Afterwards these recipient mice were infused either donor spleen cells (2 x 10(7)) on d14 and d21 or hydrocortison (HC) and cyclosporine A (CsA) treated donor spleen cells (5 x 10(7)) on d14 post-BMT. RESULTS: Survival time of mice received DLI on d21 and pretreated DLI on d14 post-BMT was longer than that of control group and d14 DLI group(P < 0.01) and without severe graft-versus-host disease. CONCLUSION: Donor lymphocyte infusion after nonmyeloablative bone marrow transplantation could reduce transplantation-associated morbidity and mortality while strengthening graft-versus-leukemia effects.

[Long-term morphological changes in mucosa of intestinal urinary reservoir].

OBJECTIVE: To investigate the relationship between long-term morphological changes in the mucosa of the intestinal urinary reservoir (IUR) and postoperative complications. METHODS: A total of 25 patients undergoing the replacement of bladder with IUR were followed up. Of these 25 patients, 15 had colonic pouch and 10 ileal pouch. Tissues of IUR were biopsied examined with microscope and transmission electron microscope. RESULTS: In patients with colonic pouch, the glandular depth was (0.17 +/- 0.04) mm and microvillous height (0.35 +/- 0.05) micron, which were significantly lower than those (0.42 +/- 0.04) micron and (0.69 +/- 0.08) mm in the control group. In those with ileal pouch, the microvillous height was (0.70 +/- 0.14) mm, (0.54 +/- 0.10) mm and (1.99 +/- 0.14) mm in the short-term group, long-term group and control group, respectively. As compared with the control group, mucous atrophy in these patients was more severe. Tight junctions of the epithelium in both kinds of pouches were well preserved even in the long-term cases. No remarkable malignant changes were found in the IUR mucosa in all the patients. Meanwhile, no special morphological changes were seen in the patients with metabolic acidosis or pouch calculus. CONCLUSIONS: IUR mucosa has some morphological "adaptive" changes after the operation. Intestinal segments are ideal bladder substitutions.

[Diagnosis and treatment of primary trigeminal neurilemmoma].

OBJECTIVE: A series of 51 patients with trigeminal neurillemmoma were analyzed with respect to the clinical manifestations, radiologic features, operative approaches and factors influencing tumor total resection. METHODS: Three different approaches were used to remove the tumors according to the site of tumor growth. Tumors in middle fossa were removed via extended middle fossa approach or extended transpterion approach. Dumbbel tumors on petrosal bone were resected by presigmoid approach. Retrosigmoid approach was used to excise tumors in posterior fossa. RESULTS: Tumors removed completely in 80.0% of patients, 75.0% of the patients had a normal cranial nerve function after operation. Tumors recurred in two patients 2 years after operation. CONCLUSION: The new operative approach increases the complete tumor resection rate.

Application of ultrasonic scalpel in gynecologic operative laparoscopy.

OBJECTIVE: To investigate the applied value of the ultrasonic scalpel in gynecologic operative laparoscopy. METHODS: Gynecologic operations were performed using the ultrasonic scalpel under laparoscopy. Operative bleeding and time, perioperative body temperature and hemogram, and tissue damage were observed. RESULTS: Forty-two cases of benign gynecologic diseases were treated with the ultrasonic scalpel under laparoscopy. Among them, there were 4 hysterectomies, 9 ovarian cystectomies, 18 salpingotomies, 4 ectopic pregnancies, 3 myomectomies, 3 adhesiolysis and 1 adnexectomy. The amount of operative bleeding, operating time and the tissue injury were related to the type of operation. The minimual operative bleeding amount, operating time, depth of tissue necrosis, tissue injury score and fibrin deposition score were 24.77 +/- 4.71 ml, 23.39 +/- 3.01 min, 0.22 +/- 0.10 mm, 0.98 +/- 0.21 and 0.38 +/- 0.26, respectively. The maximal operative bleeding amount, operating time, depth of tissue necrosis, tissue injury score and fibrin deposition score were 166.7 +/- 47.18 ml, 127.2 +/- 16.99 min, 0.35 +/- 0.20 mm, 1.25 +/- 0.20 and 0.81 +/- 0.29, respectively. The levels of body temperature, white blood cells and hemoglobin before and after the operations were not statistically different (P > 0.05) except for hysterectomy (P < 0.05). CONCLUSION: The ultrasonic scalpel can be safely applied for gynecologic operative laparoscopy and should be used widely.

[Detection of genital tract cytomegalovirus and Chlamydia trachomatis infection in patients with tubal pregnancy].

OBJECTIVE: To investigate the influence of genital tract cytomegalovirus and chlamydia infection on the incidence of tubal pregnancy. METHODS: Ninety-five women with tubal pregnancy (study group) and 42 women with ovarian cysts (control group) were selected for this study. Serum cytomegalovirus (CMV) IgM was identified by enzyme-link-immunosorbent-assay (ELISA). Cervical secretions, endometrium, and salpinx tissue were tested for CMV gH gene and chlamydia heat shock protein (HSP) gene by nest polymerase chain reaction (PCR) and PCR respectively. RESULTS: CMV IgM was positive in 14 (15%) women with tubal pregnancy, and 1 (2%) in the control group. CMV gH gene was detected in 18 women (19%) and chlamydia HSP gene in 25 (26%) of the tubal pregnancy group, and 2 (5%) and 2 (5%) in the control group respectively. There were significant differences between the study and the control group (P < 0.05) with respect to detection of CMV IgM, CMV gH gene and chlamydia HSP gene. CONCLUSIONS: CMVand chlamydia trachomatis genital tract infection were more common in women with tubal pregnancy than that in the control group. Genital CMV and chlamydia trachomatis infection may associate with an increased incidence of tubal pregnancy.

Inborn errors of metabolism discovered in Asian department of pediatrics and mental retardation research center.

To heighten the effectiveness of chemical diagnosis for inborn errors of metabolism (IEM) using urease pretreatment and GC-MS analysis, a sample collection and transportation method was contrived. The resulting "filter paper set" allows simple urine collection and transportation, and enables anyone from anywhere to receive the GC-MS analysis without the limitations of place or time. Using filter paper sets, high-risk screening of undiagnosed children or mentally retarded children with unknown cause was conducted in cooperation with hospitals and universities in several Asian countries. During 8 months 203 patients from China and India were analyzed and 20 cases of IEM were chemically diagnosed. These diagnoses greatly contributed to the treatment of children with intractable diseases who lived in Asian countries where analytical techniques and facilities for IEM were not sufficient.
