[Expression of matrix metalloproteinase and tissue inhibitor of metalloproteinase in adenomyosis].

OBJECTIVE: To investigate the expressions of matrix metalloproteinase -2, -3 (MMP-2,-3) and tissue inhibitor of metalloproteinase-2 (TIMP-2) in internal uterine endometriosis tissue and endometrium from women with and without endometriosis throughout the menstrual cycle. METHODS: Immunohistochemical staining of tissues was performed to study the expressions and locations of MMP-2, MMP-3, TIMP-2 between women with and without endometriosis. The real-time PCR technique was applied to detected the mRNA expressions of MMP-2, MMP-3 and TIMP-2. RESULTS: We found that ectopic endometrium from patients with endometriosis expressed higher levels of MMP-2 and MMP-3 and lower levers of TIMP-2 than normal controls did (P < 0.05). MMP-2 and MMP-3 were detected strongly in both stromal and epithelial cells of ectopic endometrium from patients with endometriosis, when mostly detected in the epithelial cells in the control group. All eutopic and ectopic endometrium samples from women with and without endometriosis throughout the menstrual cycle showed similar expressions of MMP-2, MMP-3 and TIMP-2. Quantitative expressions of MMP-2 mRNA, MMP-3 mRNA and TIMP-2 mRNA were significantly lower in eutopic endometrium from controls compared with ectopic endometrium from patients with endometriosis (P < 0.05). Eutopic endometrium from controls in the proliferative phase showed significantly increased expressions of MMP-2 mRNA compared with that in the secretory phase (P < 0.05). CONCLUSION: The results suggest that ectopic endometrium with adenomyosis has more invasiveness and is prone to peritoneal implantation maybe to involved in the high expressions of MMP-2 and MMP-3 and the less expressions of TIMP-2 than endometrium from women without adenomyosis does.

[P53 codon 72 polymorphism in cervical cancers and its correlation with HPV16,18E6].

OBJECTIVES: To examine p53 codon 72 polymorphism in cervical cancers and its correlation with HPV16/18 E6. METHODS: Cervical specimens were taken from 81 patients with cervical squamous cancer, 18 patients with cervical adenocarcinoma, 88 patients with CIN II - III and 60 patients without cancers. PCR was used to examine the p53 genotypes and the expression of HPV16 and 18 E6. RESULTS: The frequencies of p53 Arg homozygosity in cervical squamous cancer, cervical adenocarcinoma and CIN (II - III) were 58.020%, 55.55% and 59.09% respectively, greater than those of p53 Arg/Pro heterozygosity (30.86%, 27.78%, 21.59%) and those of p53 Pro homozygosity (11.12%, 16.67%, 19.32%). The normal cervical samples also showed less frequency of p53 Arg homozygosity (23.33%) than cervical squamous cancer. There were no significant differences in the frequencies of p53 Arg homozygosity, p53 Arg/Pro heterozygosity and p53 Pro homozygosity (23.33%, 40.00% and 36.67% respectively). The frequency of HPV16,18 E6-positive in cervical cancer and CIN was much higher than that in control group (81.84%, 50.00% and 53.41%) for the normal cervical samples. The expression of HPV16 and 18 E6 in cervical squamous cancers was more frequent than in CIN. The frequency of p53 Arg homozygosity in HRHPV E6-positive cervical squamous cancers (64.06%) was greater than in HRHPV E6-negative cervical squamous cancers (35.29%) and in HRHPV E6-positive normal samples (33.33%). The p53 codon 72 polymorphism showed no differences in samples with different FIGO staging and grades. CONCLUSION: p53 Arg homozygosity could serve as a risk indicator for the tumorigenesis of cervix. In combination with HRHPV E6, it might be able to predict the progression of cervical lesions. p53 codon 72 polymorphism is not associated with FIGO staging and grades of cervical cancers.

[VCAM-1 and E-selectin expression in extravillous cytotrophoblasts of severe preeclampsia].

OBJECTIVE: To investigate the expression pattern of vascular cell adhesion molecule-1(VCAM-1) and endothelial selectin (E-selectin) in the placental bed extravillous cytotrophoblast (EVCT) as well as its relationship with preeclampsia. METHODS: Placentas from severe preeclampsia (n=20) and gestional age-matched normal pregnancies (n=20) were studied. The staining intensity and the Mean Optical Density (MOD) of VCAM-1 and E-selectin in EVCT of placental bed deciduas were measured by immunohistochemistry. RESULTS: The staining intensity and MOD of VCAM-1 and E-selectin on placental bed EVCT in severe preeclampsia were significantly lower than those in the control group respectively (P<0.05, P<0.01). CONCLUSION: The decreased EVCT expression of vascular adhesion phenotype VCAM-1 and E-selectin may account for the shallow trophoblast invasion in preeclampsia, which is the major pathological change of this disease.

[Effects of nonsteroidal anti-inflammatory drug celecoxib on expression of cyclooxygenase-2 (COX-2) in ovarian carcinoma cell].

OBJECTIVE: To explore the effects of nonsteroidal anti-inflammatory drug Celecoxib on the expression of cyclooxygenase-2 (COX-2) in the SKOV3 cell line and the xenografted nude mice of ovarian carcinoma. METHODS: The expression of COX-2 in the SKOV3 cell was determined by reverse transcription polymerase chain reaction (RT-PCR), flow cytometry (FCM), and Western blot analysis. The expression of COX-2 in tumor cells was measured with Immunocytochemistry. RESULTS: RT-PCR showed that the expression COX-2 mRNA was strongly down-regulated in SKOV3 cells after treatment with Celecoxib or Aspirin. FCM and Western blot analysis showed that the protein product of COX-2 was strongly decreased by Celecoxib or Aspirin. The Celecoxib was more potential effects than Aspirin. The immunocytochemistry result showed that the expression of COX-2 in 10, 25, 50 mg/kg x d of Celecoxib were lower obviously than it in the control group in Xenografted nude mice. CONCLUSION: The anticarcinogenic effects of Celecoxib is probably related to the down-regulation of COX-2, and can be explained to both COX-2-dependent and -independent mechanisms.

[Relation between the expression of hypoxia inducible factor-1alpha and angiogenesis in ovarian cancer using tissue microarray].

OBJECTIVE: To study the relation between the expression of hypoxia inducible factor-1alpha (HIF-1alpha)and angiogenesis in ovarian cancer. METHODS: The expressions of HIF-1alpha mRNA, vascular endothelial growth factor (VEGF), and CD(34) in 295 patients with epithelial ovarian tumors were analyzed by tissue microarray technology, in situ hybridization and immunohistochemistry, and compared with those of 13 normal ovarian tissue samples. RESULTS: The expressions of HIF-1alpha mRNA were observed in 0, 13.2%, 42.1% and 81.9% of normal ovarian tissue, benign, borderline and malignant ovarian tumors respectively. Expression rates of HIF-1alpha mRNA in borderline and invasive tumors were significantly higher than those in normal ovarian tissues and benign tumors (P < 0.01). Statistical analysis revealed that the expression of HIF-1alpha mRNA was not related to surgical stages and histological subtypes. Close positive relation was observed between the expression of HIF-1alpha mRNA and tumor histological grade (r = 0.246, P < 0.01). There were significant correlations between the expression of HIF-1alpha mRNA and VEGF (r = 0.206, P = 0.01) and microvessel density (MVD) level (r = 0.451, P < 0.01). CONCLUSION: HIF-1alpha may play a role in angiogenesis of ovarian carcinoma, and may promote the development of the carcinoma.

[The expression of TRAIL, TRAIL-R (DR4, DcR1) and Fas-L in patients with ovarian cancer].

OBJECTIVE: To investigate the apoptosis induced by TRAIL, TRAIL-R and Fas-L in patients METHODS: The expression of TRAIL, TRAIL-R (DR4,DcR1) and Fas-L in ovarian cancer was detected with immunohistochemistry. RESULTS: The levels of DR4 and DcR1 expression in ovarian carcinoma with ovarian carcinoma were lower than that of normal tissues. The positive rate of DR4 in well-differentiated ovarian carcinoma was significantly higher than that in poorly differentiated ovarian carcinoma (92.31% vs 54.55%, P < 0.05). The expression of TRAIL was significantly correlated with that of Fas-L (r = 0.777). CONCLUSION: The expressions of DR4 and DcR1 are lower than that of normal tissues, and the expression of DR4 might be a prognostic factor in ovarian cancer.

[Relationship between the reduction of FHIT expression and the development of cervical carcinoma].

OBJECTIVE: To explore the relationship between reduction of FHIT expression of fragile histidine triad (FHIT) and the development of cervical carcinoma. METHODS: Expression of the FHIT product was detected by immunohistochemistry in 22 normal cervices and 35 cervical intra-epithelial neoplasias (CINs) as well as 60 primary invasive cervical carcinomas. RESULTS: The rates for loss or reduction of expression of FHIT protein in the squamous epithelium of normal cervices, CIN I-II, CIN III and noninvasive carcinoma and invasive cervical carcinoma were 0% (0/22), 20% (4/20), 53.3% (8/15), 81.7% (49/60) respectively (P<0.05). Among the well differentiated, intermediately differentiated and poorly differentiated invasive cervical carcinoma, the rates for loss or reduction of expression of FHIT protein were 60.0% (6/10), 70.0% (14/20), and 96.7% (29/30) respectively (P<0.05). The rate of the impaired FHIT protein expression in the invasive cervical carcinoma with lymph node metastasis (90.9%) was higher than that without lymph node metastasis (79.6%). CONCLUSION: The impaired FHIT protein expression might be a useful indicator in identifying the possibility of the progression of advanced CINs into invasive cervical carcinoma. FHIT protein expression might indicate the clinical characteristic of cervical carcinoma cells and the prognosis of cervical carcinoma.
