Two-component supramolecular helical architectures: creation of tunable dissymmetric cavities for the inclusion and chiral recognition of the third components.

The inclusion and chiral recognition of racemic arylalkanols by supramolecular helical architectures consisting of enantiopure primary amines and achiral carboxylic acids were thoroughly studied. Among the architectures examined, a supramolecular helical architecture composed of the salt of enantiopure erythro-2-amino-1,2-diphenylethanol (1 b) and benzoic acid (2 a) was found to include a wide variety of racemic arylalkanols with recognition of their chirality. The helical architecture gave a dissymmetric 1D groove in the salt crystal, and the arylalkanols were enantioselectively included in the groove. The size and shape of the groove were tunable by proper selection of the achiral carboxylic acid component. The origin of the chiral recognition with the combination 1 b/2 a is discussed on the basis of X-ray crystallographic analyses.

Switching of structural order in a cross-linked polymer triggered by the desorption/adsorption of guest molecules.

A cross-linked polymer, prepared by the in situ polymerization of a thermotropic columnar liquid crystal, was found to work as a host with a flexible framework, which was reminiscent of intercalation hosts, such as clays, graphites, and coordination polymers. The structural order of the cross-linked polymer was reversibly switched by changing the amount or shape of a guest incorporated in the polymer.

Highly regioselective transformation of out/out-equatorial bis(formylmethano)[60]fullerenes: construction of dissymmetric [60]fullerene-centered triads.

Three kinds of out/out-equatorial bis(formylmethano)[60]fullerenes (1b-d) were obtained by the tether-directed bifunctionalization of [60]fullerene with bis(alpha-formylsulfonium ylide)s. The condensation of aromatic amines with 1b-d proceeded with an unexpectedly high regioselectivity to give one of two possible regioisomers of mono-imines as the main products (the ratio of the regioisomers, up to 97:3). By the transformation of the remaining formyl group in the mono-imines thus obtained, the corresponding dissymmetric bis-imines were efficiently synthesized.

Solid-phase synthesis of oligodeoxyribonucleoside boranophosphates by the boranophosphotriester method.

Oligodeoxyribonucleoside boranophosphates (BH3-ODNs), containing four kinds of nucleobases, were synthesized by the solid-phase boranophosphotriester method. The 2'-deoxyribonucleoside 3'-boranophosphate monomers having 2-cyanoethyl (CE) groups as the phosphorus protecting groups were synthesized in good yields. A new condensing reagent, 1,3-dimethyl-2-(3-nitro-1,2,4-triazol-1-yl)-2-pyrrolidin-1-yl-1,3,2-diazaphospholidinium hexafluorophosphate, was found to be highly effective for the condensation reaction on the solid support. We also found that 1,8-bis(N,N-dimethylamino)naphthalene could accelerate the condensation reaction without causing beta-elimination of the CE groups from the boranophosphate triesters. The internucleotidic CE groups were selectively removed by treatment with 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU) under anhydrous conditions. The acetylation of the terminal 5'-hydroxy group was found to be effective to suppress the decomposition of the BH3-ODNs during the DBU treatment on the solid support. Under optimized conditions for the solid-phase synthesis and the deprotection reactions, BH3-ODNs (4mers and 12mers) containing four kinds of nucleobases were synthesized in good yields. The hybridization properties of the BH3-ODN 12mers with the complementary native DNAs and RNAs were determined by the thermal denaturing studies. In contrast to the low thermal melting (Tm) value of the duplex composed of T((PB)T)11 and native dA12 (12.8 degrees C), the duplex consisting of d(C(PB)A(PB)G(PB)T)3 and d(ACTG)3 showed a higher Tm value (44.7 degrees C) under high-salt conditions. Furthermore, d(C(PB)A(PB)G(PB)T)3 formed a more stable duplex with the complementary RNA, r(ACUG)3 with a Tm value of 50.5 degrees C. Thus, we first demonstrated that the binding affinity of BH3-ODN to a complementary DNA or RNA is dramatically increased, owing to the inclusion of the four kinds of nucleobases.

Stereoselective synthesis of dinucleoside boranophosphates by an oxazaphospholidine method.

A stereoselective synthesis of dinucleoside boranophosphates by using nucleoside 3'-oxazaphospholidine derivatives is described. The diastereoselectivity of the internucleotidic bond formation reactions varied with the nucleobase used. (Rp)- and (Sp)-dithymidine boranophosphates were synthesized with excellent diastereoselectivity both in solution and on a solid-support, whereas a loss of diastereopurity was observed for the 2'-deoxycytidine derivative having an unprotected nucleobase amino group. On the other hand, complete chemoselectivity of the 3'-oxazaphospholidine derivatives toward hydroxy groups over amino groups was serendipitously found during the study. This unique chemoselectivity of the 3'-oxazaphospholidine derivatives was investigated by comparing them with the conventional nucleoside 3'-phosphoramidite.

Synthesis and reactions of 2,2-[60]fullerenoalkanoyl chlorides.

2,2-[60]Fullerenoalkanoyl chlorides (1a-d) were easily and securely prepared from the corresponding 2,2-[60]fullerenoalkanoic acids (2a-d) by the reaction with thionyl chloride in an unusual mixed solvent, CH2Cl2/dioxane. The characterization of 1a-d by 1H and 13C NMR, FT-IR, and MALDI-TOF-MASS was conducted for the first time. The 2,2-[60]fullerenoalkanoyl chlorides thus obtained were readily converted to the corresponding amides and esters in moderate to excellent yields by the condensation with amines and alcohols, respectively. Upon applying the condensation, [60]fullerene-biomolecule hybrids were easily prepared.

Enantiopure O-substituted phenylphosphonothioic acids: chiral recognition ability during salt crystallization and chiral recognition mechanism.

[structure: see text] The chiral recognition ability of enantiopure O-methyl, O-ethyl, O-propyl, and O-phenyl phenylphosphonothioic acids (1a-d) for various kinds of racemic amines during salt crystallization and the chiral recognition mechanism were thoroughly investigated. The chiral recognition abilities of enantiopure 1a-d for a wide variety of racemic amines varied in a range of 6 to >99% enantiomeric selectivity. Deposited less-soluble diastereomeric salts were classified into two categories, prism- and needle-type crystals; the prism-type crystals were composed of a globular molecular cluster, while there existed a 2(1) column in the needle-type crystals. In contrast to a general observation of a similar 2(1) column in the less-soluble diastereomeric salt crystals of chiral primary amines with chiral carboxylic acids, the globular molecular cluster is a very unique hydrogen-bonding motif that has never been constructed in diastereomeric salt crystals. Excellent chiral recognition was always achieved when the less-soluble diastereomeric salts were prism-type crystals. Significant correlations were found between the degree of the chiral recognition with 1a-d, the crystal shape of the less-soluble diastereomeric salts, and the hydrogen-bonding motif (molecular cluster/2(1) column). The chiral recognition mechanisms via the molecular cluster and the 2(1) column formations are discussed in detail on the basis of X-ray crystallographic analyses.

The first synthesis of a methano[60]fullerene with an electron-donating group at the methano-bridge carbon: synthesis and reaction of aminomethano[60]fullerene.

[reaction: see text] Aminomethano[60]fullerene was synthesized for the first time as a trifluoromethanesulfonic acid salt by applying the Curtius rearrangement of azidocarbonylmethano[60]fullerene as the key reaction. Aminomethano[60]fullerene thus obtained was found to be able to react with various acyl chlorides to afford the corresponding amides.

Periodic ab initio approach for the cooperative effect of CH/pi interaction in crystals: relative energy of CH/pi and hydrogen-bonding interactions.

The bonding property of the CH/pi interaction in organic crystals has been investigated by the means of a periodic ab initio method. The energy of the CH(sp(2))/pi interaction in crystals, estimated with periodic RHF/6-21G*, showed a reasonable attractive CH(sp(2))/pi interaction owing to a cooperative effect, whereas the results calculated with RHF/cc-pVDZ indicate a negligibly small or repulsive interaction. The relative contribution of the CH(sp(2))/pi interaction to the column packing energy was found to be roughly half of the energy of a conventional hydrogen bond. The calculation of the charge distributions on the aromatic rings participating in the CH(sp(2))/pi interaction in crystals revealed that the atoms were more ionic than those in the gas phase. These theoretical calculations suggest a hydrogen-bonding characteristic for the CH(sp(2))/pi interaction in crystals, which does not occur in solution nor gas phase. We present computational evidence of the existence of the cooperative effect of CH(sp(2))/pi interaction in crystals.

Synthesis, absolute configuration, and application of enantiopure trans-1-aminobenz[f]indan-2-ol.

Both novel enantiopure trans-1-aminobenz[f]indan-2-ols (4) were obtained from the racemate by the diastereomeric salt formation with (+)- and (-)-dibenzoyltartaric acids (8), respectively, and the absolute configuration of the enantiomer 4 in the less-soluble diastereomeric salt of racemic 4 with (+)-8 was determined to be (1S,2S) by an X-ray crystallographic analysis. The chiral recognition ability of the enantiopure amino alcohol was examined for the enantioseparation of racemic 2-arylalkanoic acids by the diastereomeric salt formation. The role of the naphthylene group of the amino alcohol was found to be closely associated with the stabilization of the crystal by CH/pi interactions on the basis of an X-ray crystallographic study.

A novel method for the synthesis of DNA using 2'-deoxyribonucleoside 5'-phosphites as monomer units.

We propose a new method for the synthesis of DNA analogs completely different from the conventional methods (Scheme 1). Base-unprotected 2'-deoxyribo-nucleoside 5'-phosphites were employed as monomer units in this method. The 3'-OH of the monomer unit is condensed with the H-phosphonate monoester group at the 5'-terminus of the oligomer. The removal of the phosphite protecting groups gave the corresponding H-phosphonate monoester, which was allowed to condense with the 3'-OH of the monomer unit again. The synthesis of the monomer units by a simple one-step reaction from base-unprotected 2'-deoxyribo-nucleosides, and mild reaction conditions for the quantitative and rapid conversion of phosphites into H-phosphonate monoesters are described.

A novel method for the synthesis of DNA and its analogs by the use of BH3 as a protecting group for phosphonic acid.

Recently, we have developed a novel reaction for the transformation of boranophosphate diesters to the corresponding H-phosphonate diesters in the presence of trityl cation under acidic conditions. In this study, DNA and backbone-modified DNA analogs were synthesized in good yields upon applying this reaction. We report the transformation of boranophosphate DNAs, fully protected with 2-azidomethylbenzoyl groups, to various backbone-modified DNA analogs via the H-phosphonate intermediates in solution and on a solid support.

A new hydrogen-bonding motif for chiral recognition in the diastereomeric salts of racemic 1-phenylethylamine derivatives with enantiopure O-ethyl phenylphosphonothioic acid.

An enantiopure phosphonothioic acid showed a unique and superior chiral recognition ability, arising from its P-stereogenicity, for racemic 1-phenylethylamine derivatives through diastereomeric crystallization. Spherical molecular clusters, associated by hydrogen bonds and CH/pi interactions, aggregated with high symmetry in the less-soluble diastereomeric salts.

Rational design of CH/pi interaction sites in a basic resolving agent.

A novel synthetic basic resolving agent, cis-1-aminobenz[f]indan-2-ol (ABI), was rationally designed by introducing effective CH/pi interaction sites to cis-1-aminoindan-2-ol (AI), whose chiral recognition ability has been reported from our laboratory. ABI was applicable to a wide variety of racemic arylalkanoic acids and showed moderate to excellent chiral recognition ability, which was obviously higher than that of AI. The fundamental and important role of CH/pi interactions, such as tunable CH(sp(2))/pi and CH(sp(3))/pi interactions, in the chiral recognition by ABI was revealed by X-ray crystallographic study.

Supramolecular architecture consisting of an enantiopure amine and an achiral carboxylic acid: application to the enantioseparation of racemic alcohols.

Three-dimensionally dissymmetric cavities are created from an enantiopure amine and an achiral carboxylic acid, which can incorporate the third components to realize the formation of closely packed crystals. The combination demonstrates enantioselective inclusion for 20 kinds of racemic alcohols. The inclusion phenomenon occurs not only during crystallization but also in the solid state.

A novel method for the synthesis of dinucleoside boranophosphates by a boranophosphotriester method.

2'-Deoxyribonucleoside-3'-boranophosphates (nucleotide monomers), including four kinds of nucleobases, were synthesized in good yields by the use of new boranophosphorylating reagents. We have explored various kinds of condensing reagents as well as nucleophilic catalysts for the boranophosphorylation reaction with nucleosides. In the synthesis of dinucleoside boranophosphates, undesirable side reactions occurred at the O-4 of thymine and the O-6 of N2-phenylacetylguanine bases. To avoid these side reactions, additional protecting groups, benzoyl (Bz) and diphenylcarbamoyl (Dpc) groups, were introduced to thymine and guanine bases, respectively. As a result, the condensation reactions proceeded smoothly without any side reactions, and the dimers including four kinds of nucleobases were obtained in excellent yields. In the deprotection of the 5'-DMTr group, Et3SiH was found to be effective as a scavenger for the DMTr cation which caused a P-B bond cleavage. After removal of the other protecting groups by the conventional procedure, four kinds of dinucleoside boranophosphates were obtained in good yields.

Reaction of [2-(SiH3)C6H4]2SiH2 with Ni(Et2PCH2CH2PEt2)(PEt3)2: characterization of eta2-(Si-H)Ni and NiIV-H complexes.

The reaction of (2-SiH3C6H4)2SiH2 with Ni(Et2PCH2CH2PEt2)(PEt3)2 afforded a new silylnickel complex, which, in the solid state, was determined to be a bis(silyl)eta2-(Si-H)nickel complex, the first example of eta2-(Si-H)nickel complex by single-crystal X-ray analysis. Variable-temperature NMR spectroscopy revealed fluxional behavior of the complex in solution; at room temperature, exchange of five hydrogens (two SiH2 and one hydrogen bound to nickel) was observed, while at -80 degrees C the exchange of hydrogens appeared frozen to adopt a tris(silyl)(hydrido)nickel(IV) structure, which was confirmed by theoretical calculation. The latter complex is the first example of hydridonickel(IV) complex.
