Study on safety and bioavailability of ubiquinol (Kaneka QH) after single and 4-week multiple oral administration to healthy volunteers.

The safety and bioavailability of ubiquinol (the reduced form of coenzyme Q(10)), a naturally occurring lipid-soluble nutrient, were evaluated for the first time in single-blind, placebo-controlled studies with healthy subjects after administration of a single oral dose of 150 or 300 mg and after oral administration of 90, 150, or 300 mg for 4 weeks. No clinically relevant changes in results of standard laboratory tests, physical examination, vital signs, or ECG induced by ubiquinol were observed in any dosage groups. The C(max) and AUC(0-48 h) derived from the mean plasma ubiquinol concentration-time curves increased non-linearly with dose from 1.88 to 3.19 micro g/ml and from 74.61 to 91.76 micro g h/ml, respectively, after single administration. Trough concentrations had nearly plateaued at levels of 2.61 micro g/ml for 90 mg, 3.66 micro g/ml for 150 mg, and 6.53 micro g/ml for 300 mg at day 14, and increased non-linearly with dose in the 4-week study. In conclusion, following single or multiple-doses of ubiquinol in healthy volunteers, significant absorption of ubiquinol from the gastrointestinal tract was observed, and no safety concerns were noted on standard laboratory tests for safety or on assessment of adverse events for doses of up to 300 mg for up to 2 weeks after treatment completion.

Safety assessment of coenzyme Q10 (Kaneka Q10) in healthy subjects: a double-blind, randomized, placebo-controlled trial.

The safety profile of Coenzyme Q10 (Kaneka Q10) at high doses for healthy subjects was assessed in a double-blind, randomized, placebo-controlled study. Kaneka Q10 in capsule form was taken for 4 weeks at doses of 300, 600, and 900 mg/day by a total of eighty-eight adult volunteers. No serious adverse events were observed in any group. Adverse events were reported in 16 volunteers with placebo, in 12 volunteers with the 300 mg dose, in 20 volunteers with the 600 mg, dose and in 16 volunteers with the 900 mg dose. The most commonly reported events included common cold symptoms and gastrointestinal effects such as abdominal pain and soft feces. These events exhibited no dose-dependency and were judged to have no relationship to Kaneka Q10. Changes observed in hematology, blood biochemistry, and urinalysis were not dose-related and were judged not to be clinically significant. The plasma CoQ10 concentration after 8-month withdrawal was almost the same as that before administration. These findings showed that Kaneka Q10 was well-tolerated and safe for healthy adults at intake of up to 900 mg/day.

Reduced coenzyme Q10 supplementation decelerates senescence in SAMP1 mice.

The SAMP1 strain is a mouse model for accelerated senescence and severe senile amyloidosis. We determined whether supplementation with coenzyme Q10 (CoQ10) could decelerate aging in SAMP1 mice and its potential role in aging. Plasma concentrations of CoQ10 and CoQ9 decreased with age in SAMP1 but not in SAMR1 mice. Supplementation with reduced CoQ10 (CoQH2, 250 mg/kg/day) for one week increased plasma CoQ10 concentrations, with an accompanying decrease in plasma CoQ9 concentrations. In two series of experiments, lifelong supplementation with CoQH2 decreased the senescence grading scores from 10 to 14 months, 7 to 15 months, and at 17 months of age. The body weight of female mice increased from 2 to 10 months of age versus controls in the second series of experiments. Lifelong CoQH2 supplementation did not prolong or shorten the lifespan, nor did it alter the murine senile amyloid (AApoAII) deposition rate or cancer incidence. In the second series of experiments, urinary levels of 8-hydroxydeoxyguanosine did not change with age or long-term supplementation with CoQH2. Urinary levels of acrolein (ACR)-lysine adduct increased significantly with age in SAMP1 mice; however, CoQH2 had no effect. Thus, lifelong dietary supplementation with CoQH2 decreased the degree of senescence in middle-aged SAMP1 mice.

A case of cutaneous polyarteritis nodosa manifested by spiking high fever, arthralgia and macular eruption like adult-onset Still's disease.

A 24-year-old Japanese woman was admitted for investigation of recurrent spiking high fever associated with a macular eruption of the upper extremities associated with fever and polyarthralgia. These symptoms were self-limiting but recurrent and seemed to be consistent with a diagnosis of adult-onset Still's disease (AOSD). However, livedo reticularis was detected on the lower extremities, suggesting the presence of vasculitic disease rather than AOSD. Investigation did not reveal any evidence of visceral involvement. Skin biopsy of the affected lower extremity demonstrated a necrotizing vasculitis of small muscular artery and confirmed a diagnosis of cutaneous polyarteritis nodosa (PAN) rather than AOSD. Treatment with 30 mg of prednisolone daily improved the skin lesions and the recurrent spiking high fever and the arthralgia were resolved. PAN and AOSD are clinically similar, and discrimination may be sometimes difficult. The presence of livedo reticularis and the finding of a characteristic skin biopsy appearance may be diagnostically useful to distinguish PAN from AOSD. Indeed, the clinical features of cutaneous PAN may be more similar to AOSD than systemic PAN, and a skin biopsy may be necessary to distinguish cutaneous PAN from AOSD in some cases.

A case of polyarteritis nodosa with lesions of the superior mesenteric artery illustrating the diagnostic usefulness of three-dimensional computed tomographic angiography.

A 40-year-old Japanese man who developed upper abdominal pain, weight loss, and hypertension was diagnosed as suffering with polyarteritis nodosa (PAN) with lesions of the superior mesenteric artery (SMA). Three-dimensional computed tomographic angiography (3D-CTA) and conventional angiography revealed smooth segmental luminal narrowing of the branches of the SMA. In addition, an enhanced abdominal CT scan demonstrated diffuse thickening of the wall of the affected SMA. Treatment with high-dose corticosteroids resulted in immediate improvement of the abdominal manifestations and normalized the serum C-reactive protein levels. Furthermore, the subsequent enhanced CT and 3D-CTA revealed improvement of the wall thickening and luminal narrowing of the SMA and its branches during the treatment period. In addition to possessing the diagnostic usefulness of conventional angiography, 3D-CTA is also less invasive and facilitates the prompt and accurate diagnosis of PAN. Furthermore, thickening of the wall of medium-sized arteries evidenced by enhanced CT scan may also support a diagnosis of PAN.

Spatial and temporal variations of atmospheric 85Kr observed during 1995-2001 in Japan: estimation of atmospheric 85Kr inventory in the Northern Hemisphere.

Atmospheric 85Kr concentrations have been continuously monitored since 1995 at the Meteorological Research Institute (MRI) in Tsukuba, Japan. They have also been observed once a year at several stations over the Japanese islands since 1995. The annual growth rate of the background atmospheric 85Kr concentrations in Tsukuba was 0.03 Bq x m(-3) x yr(-1) during 1996-2001. The atmospheric 85Kr concentrations at several stations over Japan were within the range of the annual variations in Tsukuba. However, higher and lower 85Kr concentrations in early winter, compared with those in Tsukuba (36.1 degrees N, 140.1 degrees E), occurred in Sapporo (43.1 degrees N, 141.3 degrees E) and Ishigaki (24.3 degrees N, 124.2 degrees E), respectively. The reason for this is that Sapporo is covered by a continental air mass, some from European sources, whereas Ishigaki is still covered by a subtropical air mass. The Northern Hemispheric background 85Kr concentrations from 1994 to 2001 was calculated from the 85Kr inventory and the release rate of 85Kr from the nuclear fuel reprocessing plants in Europe. Calculated 85Kr concentrations in surface air were in good agreement with annual average observed values at the MRI, Tsukuba. The global atmospheric inventory of 85Kr in December 2001 was also estimated to be approximately 5 EBq by using observed data in Tsukuba.

[Report on the 89th Scientific Assembly and Annual Meeting of the Radiological Society of North America--evaluation of spatial resolution and contrast resolution for MPR diagnosis].

PURPOSE: The advent of the helical scanner and the multi-slice helical scanner has encouraged image diagnosis using MPR display. In this study we investigated methods of evaluating spatial and contrast resolution when using MPR image display. METHOD AND MATERIALS: Using a three-dimensional ladder phantom to evaluate spatial resolution, we placed ladders along the MPR sections and measured the responses in the CT values. For evaluating contrast resolution, we used a phantom involving subtle differences in the CT values. We then found the CNR of those CT image. RESULTS: The results of the experiment showed that when the FOV value, collimation and helical pitch were set at 240 mm, 0.5 mm, 3 and 320 mm, 1.0 mm, 3 respectively, good MPR display were achieved with respect to both spatial resolution and contrast resolution. CONCLUSION: Image diagnosis using MPR display is an extremely useful method of diagnosis from an anatomical perspective. We were able to get better understanding through this investigation of the scanning parameter that optimizes time resolution and contrast resolution when using MPR display. Textural properties corresponding to visual perception based on the correlation mechanism in the visual system.

We present a set of texture parameters that correspond to perceptual properties of visual texture. For machine vision or a computer interface, it is important that the computational measurements of texture correspond well to the perceptual properties. To understand the mechanism of our visual system, it is important to know how we extract or characterize information for texture perception. In this study, we show that the autocorrelation function (ACF) analysis provides useful measures for representing three salient perceptual properties of texture: contrast, coarseness, and regularity. The validity of the ACF analysis was examined by comparing the calculated factors to the subjective scores collected for various kinds of natural textures. The effectiveness of the analysis depends on the structure of the estimated ACF. When a texture has a harmonic structure, the estimated ACF has periodical peaks corresponding to the periods of the texture. Both perceived coarseness and regularity are strongly related to these peaks in the ACF. However, the estimated ACF does not have a periodical structure when the texture is random. In this case, the texture coarseness and regularity are represented by the decay rate of the ACF.

Single nucleotide polymorphisms in the human complement C6 and C7 genes.

We analyzed the single nucleotide polymorphisms (SNPs) in the sixth (C6) and the seventh (C7) component genes of the complement system in a sample of the Japanese population, using polymerase chain reaction (PCR)-based methods and PCR direct sequencing. SNPs in the C6 gene studied here are as follows: A413C in exon 3, T1674C in exon 10, T7145A in exon 13, G[357+32]A in intron 2, and G[503-78]A in intron 3. We confirmed that nt413A and nt413C were associated with C6A and C6B, respectively. The result of the nt2145 typing showed that two subtypes exist in the C6B allotype. The SNP of G[357+32]A in intron 2 could be analyzed by using the PCR-RFLP method with HinfI. Allele frequencies in the Japanese population were found to be *G=0.920 and *A=0.080. SNPs in the C7 gene are as follows: T382C in exon 4, G1166C and A1258C in exon 9, and G[+10]A in intron 13. Nt382C and nt1258C would be responsible for C7-5 (=C7-3) and C7-4 allotypes, respectively.

Short tandem repeat typing in exon 1 of the androgen receptor gene.

Exon 1 of the androgen receptor (AR) gene on the X chromosome contains a polymorphic CAG trinucleotide short tandem repeat. We describe here the rapid and reliable method of typing CAG repeats using electrophoresis, with denaturing polyacrylamide gel and an allelic ladder marker. Twenty-one alleles (the repeat number ranges from 13 to 35) were found using CAG repeat typing in normal Japanese individuals (83 males and 82 females). The allelic diversity (h) calculated was h=0.889, illustrating that CAG repeats at the AR locus is a highly polymorphic system.

A-elute alleles of the ABO blood group system in Japanese.

The ABO blood group system is important in forensic genetics, as well as transfusion medicine. Since the elucidation of the molecular basis of ABO gene regulation, nucleotides of variant alleles or suballeles have been analyzed by polymerase chain reaction (PCR)-based methods and sequencing. Ael (A-elute) is one of the subgroups of A in the ABO system. By analyzing the suballeles responsible for Ael phenotype by PCR-RFLP and PCR direct sequencing, we found seven types of Ael allele. The allele frequency of ABO*Ael in a Japanese population was calculated to be 0.0049.

[A case of pulmonary infiltration with eosinophilia (PIE) syndrome induced by bucillamine treatment of rheumatoid arthritis].

Bucillamine is used mainly in treating rheumatoid arthritis (RA). We report a case of bucillamine-induced pulmonary infiltration with eosinophilia (PIE) syndrome in a 51-year-old woman. When RA was diagnosed, she was treated with bucillamine from December 2000. In April 2001, she was admitted to our hospital because of fever and skin eruptions. Chest radiography and CT revealed both diffuse ground-glass opacity and fine nodular shadows. Laboratory data showed a normal white cell count with eosinophilia. Bronchoalveolar lavage (BAL) studies showed that total cell counts and the proportion of eosinophils were increased, and that the CD4/CD8 ratio of the T-cell subsets was decreased to 0.93. The patch test to bucillamine was positive. After bucillamine was withdrawn, the fever and the abnormal chest shadows improved. We concluded from the patient's clinical course, laboratory data and BAL findings that this was a case of bucillamine-induced PIE syndrome. Since most cases of bucillamine-induced interstitial pneumonitis are lymphocytic alveolitis, we consider that PIE syndrome in such a case is a very rare condition. We concluded that bucillamine should be added to the list of drugs capable of producing PIE syndrome.

Intracellular signal-responsive artificial gene regulation for novel gene delivery.

We describe two types of artificial gene-regulation systems responding to cyclic AMP-dependent protein kinase (PKA) or caspase-3. These molecular systems use newly synthesized cationic polymers, PAK and PAC. The PAK polymer includes substrate oligopeptide for PKA, ARRASLG, as receptor of PKA signal, while the PAC polymer possesses oligopeptide that is comprised of a substrate sequence of caspase-3, DEVD, and a cationic oligolysine, KKKKKK. These polymers formed stable complexes with DNA to totally suppress the gene expression. However, PKA or caspase-3 signal disintegrates the PAK-DNA or the PAC-DNA complex, respectively. This liberates the DNA and activated the gene expression. These systems are the first concept of an intracellular signal-responsive gene-regulation system using artificial polymer. We expect that these systems can be applied to the novel highly cell specific gene delivery strategy that is involved in our previously proposed new drug delivery concept, the drug delivery system based on responses to cellular signals.
