Concise synthesis of arnottin I and (-)-arnottin II.

Application of the Buchwald protocol to the coupling of o-bromobenzoates and 1-tetralones directly affords benzodihydronaphthopyrones with a fused tetracyclic system. Aromatization of the 7,8-dimethoxy-2,3-methylenedioxy derivative yielded arnottin I, whereas oxidation with dioxirane afforded dihydroarnottin II composed of a spiro phthalide-tetralone system. Sharpless asymmetric dihydroxylation using AD-mix yielded optically active dihydroarnottin II with good enantioselectivity. The absolute stereochemistry of the stereogenic center in the (+)-spiro product was determined to be R by X-ray crystallographic analysis of the dibromo derivative. (+)-Dihydroarnottin II was subjected to successive bromination and dehydrobromination to prepare (-)-arnottin II. The R-configuration of natural (-)-arnottin II, previously assigned by application of the exciton chirality method to the Cotton effects observed in the CD spectrum, was confirmed by asymmetric synthesis.

Facile synthesis of highly congested 1,2-diphosphinobenzenes from bis(phosphine)boronium salts.

[Structure: see text] Bis(phosphine)boronium salts 3a-c were designed and prepared as key building blocks for the synthesis of highly congested diphosphinobenzenes. The preparation of sterically hindered ortho-phenylene-bridged diphosphines 1a-c was achieved by the reaction of the bis(phosphine)boronium salts 3a-c with difluorobenzenechromium complex 2 and subsequent removal of the BH2 group. The steric nature of diphosphine 1a was revealed in single-crystal X-ray analysis of its Rh complex.

Spontaneous resolution of aromatic sulfonamides: effective screening method and discrimination of absolute structure.

An effective screening method combining parallel synthesis and solid-state CD measurements was established to identify achiral aromatic sulfonamides that show spontaneous resolution with rapidity. We found that 4 of the 12 achiral sulfonamides crystallized as chiral crystals through this method. The chirality of each sulfonamide was discriminated by solid-state CD spectra and Flack parameter in an X-ray analysis. Correspondence between the observed Cotton effect and the absolute configuration could be confirmed by time-dependent DFT calculations. [structure: see text]

Naphthalene- and anthracene-based aromatic foldamers with iminodicarbonyl linkers: their stabilities and application to a chiral photochromic system using retro [4 + 4] cycloaddition.

A circular dichroism (CD) spectral study on chiral aromatic chain imides possessing anthracene and naphthalene moieties with bulky N substituents showed that their helical chirality based on folding remained for a reasonably long time without racemization in solution. Racemization due to conformational equilibration occurred very slowly, requiring over 1 week at ambient temperature. Their CD spectra both in solution and in the solid state gave similar CD signals, suggesting retention of helicity observed in the solid state even after dissolving. As an application of this novel chiral folding of aromatic chain imides, a chiral photochromic system was investigated based on the photo [4 + 4] cycloaddition and its thermal cycloreversion of an anthracene-naphthalene system. The foldamer possessing an anthracene moiety in the center connected with two naphthalene moieties below and above it by iminodicarbonyl linkers was prepared for this purpose. Induced CD was observed for the foldamer with (S)-1-(1-naphthyl)ethyl substituents at the imide nitrogen atoms. Chiral photochromic cycles were monitored by CD spectral measurement.

Guanidinium ylide mediated aziridination: identification of a spiro imidazolidine-oxazolidine intermediate.

We successfully isolated a spiro imidazolidine-oxazolidine intermediate in the reaction of guanidinium ylide mediated aziridination using alpha-bromocinnamaldehyde. X-ray crystallographic analysis unambiguously revealed that the stereogenic centers of the spiro intermediate were in a trans configuration. The role of the spiro compound as an intermediate in the aziridination reaction was confirmed by observation of its smooth chemical conversion into aziridine products.

Highly stereoselective asymmetric Pummerer reactions that incorporate intermolecular and intramolecular nonbonded S...O interactions.

New chiral sulfoxides (R(S),S)-3, (S(S),S)-3, (R(S),S)-4, and (S(S),S)-4 and known chiral sulfoxides (R(S))-5, (R(S))-6, and (R(S))-7 were synthesized, and the stereochemistry of the new sulfoxides (R(S),S)-3 and (R(S),S)-4 was determined by X-ray crystallographic analysis. In their crystallographic structures, the intramolecular nonbonded S...O close contacts were recognized. Analyses of several sulfoxide complexes including rac-11 with N,N-dimethylacetamide (DMAC) or N-methyl-2-pyrrolidone (NMP) in a MeOH solution utilizing cold-spray ionization mass spectrometry provided, for the first time, direct information for intermolecular nonbonded S...O interactions between sulfoxides and amide (or lactam) in a solution. Highly diastereoselective and enantioselective Pummerer reactions based on the concept of intermolecular and intramolecular nonbonded S...O interactions were performed by treatment of several chiral sulfoxides (R(S), S)-3, (S(S), S)-3, (R(S), S)-4, (S(S), S)-4, (R(S))-5, (R(S))-6, and (R(S))-7 with acetic anhydride and trimethylsilyl triflate (TMSOTf) in DMAC, NMP, N,N-dimethylformamide, and N-formylpiperidine. Mechanistic studies on these facile stereoselective Pummerer reactions revealed the necessity for the amide/TMSOTf complex, such as 26 or 27, to be an efficient activation reagent for Ac(2)O and a trapping reagent for the released acetate ion, and that DMAC and NMP had a positive effect on this highly stereoselective chiral transfer reaction.

Structure and reaction pathway of TMP-Zincate: amido base or alkyl base?

The novel directed ortho metalation (DoM) reagents for functionalized aromatic rings, TMP-Zn-ates (R2Zn(TMP)Li (R = Me, 1; tBu, 2)), have been reported to be synthetically useful for the chemo- and regioselective construction of multi-functionalized aromatic compounds. Here, we present the first comprehensive structural and mechanistic investigation by means of X-ray, NMR, and DFT studies on the DoM reaction employing our original TMP-Zn-ate base. The structures of TMP-Zn-ates in solution and in the solid state were determined. The DFT study strongly suggested that the deprotonation involving the TMP ligand on the TMP-Zn-ate is kinetically more favorable than that involving the alkyl ligand, and this view was supported by monitoring of the 13C NMR spectrum of the reaction mixture.

Liquid crystal and crystal structure of octahomotetraoxacalix[4]arenes.

Octahomotetraoxacalix[4]arenes bearing long alkyl chains on their lower rim were prepared. Ester 4a existed in a 1,2-alternate conformation in its crystal structure, which was examined by single-crystal X-ray diffraction analysis. To prepare liquid crystalline materials possessing calixarene moieties by self-assembling, carboxylic acid derivatives 5 were synthesized. Among them, 5c, the octadecyloxy derivative, showed smectic liquid crystal phase. Homooxacalixarenes 5 also formed liquid crystal phases with longer layer distances when two equivalent moles of 1,2-ethylenediamine were added as a linker. These phases were investigated with X-ray diffraction, differential scanning calorimetry, and polarized optical microscopy.

Assembly state of catalytic modules as chiral switches in asymmetric Strecker amino acid synthesis.

Self-assembled chiral polymetallic complexes often demonstrate novel properties as asymmetric catalysts. We report the three-dimensional structures of two such asymmetric catalysts (crystals A and B) for Strecker alpha,alpha-disubstituted amino acid synthesis. These complexes are constructed via assembly of the same chiral modules derived from d-glucose, but their assembly modes differ. The enantioselectivity in the Strecker reaction was dramatically switched, depending on which assembly mode was used: the catalyst generated in situ whose structure is represented by crystal B, or by crystal A. These findings provide insight into the functional importance of higher-order structures of an artificial catalyst.

Synthesis and cofacial pi-stacked packing arrangement of 6,13-bis(alkylthio)pentacene.

[reaction: see text] 6,13-Bis(alkylthio)pentacenes directed toward organic field-effect transistors (OFETs) were synthesized by the ZnI(2)-mediated reaction of trans-6,13-dihydroxy-6,13-dihydropentacene with alkylthiols, followed by the dehydrogenative aromatization of the resulting trans-6,13-bis(alkylthio)-6,13-dihydropentacenes with p-chloranil. The X-ray crystallographic analysis of 6,13-bis(methylthio)pentacene reveals that this compound is arranged as a result of cofacial pi-stacking with S-S and S-pi interactions.

Epoxysilane rearrangement induced by a carbanion generated by conjugate addition of enolates of chloroacetate and alpha-chloroacetamides: formation of functionalized cyclopropane derivatives.

[reaction: see text] Reaction of an enoate bearing an epoxysilane moiety at the alpha-position with lithium enolate of 2-chloroacetamide afforded highly functionalized cyclopropane derivatives via a tandem process that involves Michael addition, ring opening of the epoxide, Brook rearrangement, and intramolecular alkylation.

DNA and RNA as new binding targets of green tea catechins.

The significance of catechins, the main constituent of green tea, is being increasingly recognized with regard to cancer prevention. Catechins have been studied for interactions with various proteins, but the mechanisms of the various catechins are not yet elucidated. Based on our previous observation that nucleic acids extracted from catechin-treated cells are colored, we studied whether catechins directly interact with nucleic acids using surface plasmon resonance assay (Biacore) and cold spray ionization-mass spectrometry. These two methods clearly showed that (-)-epigallocatechin gallate (EGCG) binds to both DNA and RNA molecules: the Biacore assay indicated that four catechins bound to DNA oligomers, and cold spray ionization-mass spectrometry analysis showed one to three EGCG molecules bound to single strand 18 mers of DNA and RNA. Moreover, one or two molecules of EGCG bound to double-stranded (AG-CT) oligomers of various nucleotide lengths. These results suggest that multiple binding sites of EGCG are present in DNA and RNA oligomers. Double-stranded DNA (dsDNA) oligomers were detected only as EGCG-bound forms at high temperature, whereas at low temperature both the free and bound forms were detected, suggesting that EGCG protects dsDNA oligomers from dsDNA melting to single-stranded DNA. Because both galloyl and catechol groups of EGCG are essential for DNA binding, both groups seem to hold strands of DNA via their branching structure. These findings reveal for the first time the link between catechins and polynucleotides and will intensify our understanding of the effects of catechins on DNA in terms of cancer prevention.

Selective formation of a self-assembling homo or hetero cavitand cage via metal coordination based on thermodynamic or kinetic control.

The selective formation of a homo or hetero cavitand cage composed of two molecules of tetra(4-pyridyl)-cavitand (1), tetrakis(4-cyanophenyl)-cavitand (2), or tetrakis(4-pyridylethynyl)-cavitand (3), and four molecules of Pd(dppp)(OTf)(2) (4) or Pt(dppp)(OTf)(2) (5) has been studied. A 1:1:4 mixture of 1 with more steric restriction, 2 with less coordination ability, and 4 or 5 specifically self-assembled into a hetero cavitand cage 6 or 7, respectively. In contrast, a 1:1:4 mixture of 2, 3, and 4 in CDCl(3) at room temperature assembled into the most labile homo cyanophenyl cavitand cage 8 and the most stable homo pyridylethynyl cavitand cage 9 in a 1:1 ratio. Upon heating at 50 degrees C, the thermodynamic equilibrium was shifted to a 1:1:1 mixture of 8, 9, and a hetero cavitand cage 10. When 1 equiv of 3 was added to 8 at room temperature, 8, 9, and 10 were formed initially in a 1:1:3 ratio and finally shifted to a 1:1:1 ratio. In the Pt-system, upon addition of 1 equiv of 3 to homo cyanophenyl cavitand cage 11 in CDCl(3) at room temperature, the ratio of hetero to homo cavitand cage (13/12) initially attained was 8.7 and remained above 5.6 at room temperature. Upon heating at 50 degrees C, 13 was finally converted to 11 and 12. Thus, the selectivity for the self-assembly of the homo or hetero cavitand cage is controlled by the balance between kinetic and thermodynamic stabilities of cages based on a combination of factors such as coordination ability and steric demand of the cavitands.
