Metabolic CRISPR screening identifies RPE as a key regulator of acquired enzalutamide resistance through FKBP5 destabilization in prostate cancer.

Enzalutamide is a cornerstone therapy for castration-resistant prostate cancer (CRPC), yet acquired resistance remains a major clinical challenge. Although metabolic enzymes are increasingly recognized as modulators of therapeutic response, their specific roles-particularly their non-enzymatic functions-in sustaining enzalutamide resistance remain incompletely understood. In this study, we performed an in vivo screen using a custom metabolic CRISPR library in enzalutamide-treated xenografts and identified the pentose phosphate pathway enzyme ribulose-5-phosphate 3-epimerase (RPE) as a critical driver of enzalutamide resistance. Silencing RPE markedly restored enzalutamide sensitivity, enhanced apoptosis in vitro, and significantly suppressed tumor growth in both cell line-derived and patient-derived xenograft models. Mechanistically, RPE promoted resistance independently of its canonical enzymatic activity. Instead, RPE physically interacted with FKBP5 and promoted its ubiquitin-proteasome-mediated degradation. Loss of FKBP5 subsequently hyperactivated AKT signaling, leading to increased p-BAD and BCL-xL levels and suppression of enzalutamide-induced cell death. Conversely, disrupting the RPE-FKBP5 interaction or silencing RPE in vivo using a PSMA-targeted lipid nanoparticle system effectively abrogated these resistance phenotypes. Together, these findings illustrate how CRPC cells hijack the non-enzymatic function of a metabolic enzyme to evade antiandrogen therapy, establishing the RPE-driven degradation of FKBP5 and consequent AKT hyperactivation as a targetable vulnerability for overcoming enzalutamide resistance.

Discovery and Engineering of a Rat Endogenous Retrovirus Reverse Transcriptase for Efficient Prime Editing.

CRISPR-based prime editors (PEs) install precise edits into genomic DNA without generating double-strand breaks. Their editing efficiency is highly dependent on reverse transcriptases (RTs), but efficient RT candidates remain limited. Here, we identified 19 novel active RTs by screening 558 candidates. Among them, RERV-RT, derived from Rattus norvegicus, exhibited the highest activity. Through structure-guided engineering and deep mutational scanning, we developed an optimized variant, enRERV-RT, which outperforms conventional M-MLV-RT-based PE systems by 1.20-fold in mammalian and plant cells, and by 1.88-fold at hard-to-edit loci, while enabling precise multiplex editing of functionally relevant genes. Additionally, we developed a high-throughput platform, TRAP-seq-PE, to systematically evaluate prime editor performance. Across diverse mutation types, we found that PE systems based on enRERV-RT exhibited higher editing efficiencies than those based on M-MLV-RT. Collectively, our work establishes a versatile, high-efficiency PE system, thereby facilitating advances in clinical gene therapy and precise crop breeding.

Development of a strontium-containing hydroxyapatite bone cement.

A new route was developed to synthesis a new type of strontium-containing hydroxyapatite (Sr-HAP) bone cement with precursors of tetracalcium phosphate (TTCP), strontium hydrogen phosphate (DSPA), dicalcium phosphate (DCPA), phosphate acid and water. The processing parameters and fundamental properties including pH value, setting time, compressive strength of final hardened body and the cytotoxicity for serial extracts of each cements were investigated. The result shows that the final product of the cement after setting for 24h is nonstoichiometic Sr-containing hydroxyapatite (Ca(10-m-x)Sr(x) square(m)(HPO4)y(PO4)6-y(OH)2-2m square2m, 0<x<1, nSr-HAP) and no other harmful impurities were detected. The pH value of Sr-containing cement pastes approaches to 7.0-7.6 when they are mixed with a ratio of 1:1 of powder to liquid (P/L) in weight. The setting time of the cement pastes is 4-11 min for the initial one and 10-17 min for the final one when the concentration of diluted phosphate is in a range of 0.5-1.0 mol/l. The compressive strengths of the hardened cements with different molar ratios of Sr/(Sr+Ca) after subjected an immersion in simulated body fluid (SBF) increase uniformly from 1 day to 5 days, where they get maximum values, respectively, but then decrease till to 2 weeks. Especially for the CPC-1, with a Sr/(Sr+Ca) molar ratios of 5% in cement powder composition, the largest compressive strength gained at 5 days is 66.57 MPa and the lowest one gained at 2 weeks is 44.75 MPa, which matches the value of human bones and can be expected to use in clinic application in repairing the nonloading sites on account of the positive result of cytotoxicity test of the extracts of Sr-containing calcium phosphate cement (Sr-CPC).

[Preparation and properties of hydroxyapatite/epoxy composite].

The hydroxyapatite (HA) powder surface modified with silane coupling agent was used to prepare HA/epoxy composite. It was found that silane has greatly improved the dispersion of HA in epoxy. The composite has good in vitro bioactivity and biocompatibility with 40 wt% HA, and the flexural modulus is close to that of natural bone, but its strength is lower than that of natural bone. So the composite needs further reinforcement in some way or other.

[Application of polyvinyl alcohol in biomedical engineering].

This paper explicated the present application of poly- vinyl alcohol (PVA) in the field of biomedical engineering, such as artificial cartilage, drug delivery systems, microorganism enwrapping, cell micro-capsulation, anti-thrombin materials, and biomedical sponges. And a preliminary study of the good dispersion of PVA as a surfactant on nano-particles of hydroxyapatite was presented.

Photoexcited formation of bone apatite-like coatings on micro-arc oxidized titanium.

A novel method to rapidly deposit bone apatite-like coatings on titanium implants in simulated body fluid (SBF) is proposed in this article. The processing was composed of two steps; for example, micro-arc oxidation (MAO) of titanium to form titania films, and UV-light illumination of the titania-coated titanium in SBF. The morphology, crystalline structure, and bond strength of the MAO films were investigated as a function of the applied voltage (in the range of 250-400 V) by using scanning electron microscopy, transmission electron microscopy, energy-dispersive X-ray spectrometry, Fourier transform infrared spectroscopy, adhesion-tensile test, and scratch test. Results showed that the MAO films were porous and nanocrystalline with pore sizes varying from 1 to 3 microm and grain sizes varying from 10-20 to 70-80 nm; the predominant phase in titania films changed from anatase to rutile, and the bond strength of the films decreased from 43.4 to 32.9 MPa as the applied voltage increased from 250 to 400 V. After UV-light illumination of the films in SBF for 2 h, bone apatite-like coating was deposited on the MAO film formed at 250 V. The bond strength of the apatite/titania bilayer was about 44.2 MPa. However, no apatite was observed on the MAO film formed at 400 V after UV-light illumination.

Surface modification of titanium implant by microarc oxidation and hydrothermal treatment.

Surface modification of titanium implant is processed by microarc oxidation and hydrothermal treatment. A porous surface with a biologically active bone-like apatite layer was formed. The apatite layer consists of very fine crystals and high crystallinity and is integrated with the titanium alloy substrate with a graded structure without a distinct interface. Such a bioactive layer is expected not only to enhance the bony ingrowth into the porous structure, but also to improve the interlocking between implant and bone.

[Prospect of the foveola formation in the bovine trabecular bone under fatigue process].

This study aims at the mechanism of foveola formation in bovine trabecular bone under fatigue process and its relation with biomechanical pathogenesis of senile osteoporosis. The scanning electron microscope equipped with fatigue stage was used to observe fatigue micro injury accumulation of cancellous bone. The massive foveola formation in the laminal bone of vertical trabeculae was found in the tensile fatigue test. There existed the collagen avulsion in the foveola. The massive foveola formation was also observed in the lamina bone of the horizontal trabeculae in the compressive fatigue test. The bone collagen fibers were protracted, debound with hydroxyapatite crystal, and then avulsed under tensile and bending stresses. Finally the retraction of the avulsed collagen fibers brought on the massive formation of foveolae in lamina bone. The mechanical capacity of bone also declined greatly. We infer that the direct mineralization of avulsed collagen and foveola in lamina bone would be one of the main processes of self repair in vivo, which brings on the increase in fragility and stiffness of trabeculae of senile osteoporotic bone along with the agelong accumulation of collagen fatigue injury and foveola formation in the lamina bone.

Orthotopic ileal neobladder similar to original bladder.

OBJECTIVE: To report the surgical techniques and results of an 8-year follow-up study of 42 patients with a modified orthotopic ileal neobladder restoring normal anatomical relationship. METHODS: Total cystoprostatectomy was performed extraperitoneally. A 45 - 50 cm segment of the ileal loop was isolated, detubularized, and reconfigured into an "M"-shape to form a pouch. Bilateral ureters were implanted by inserting 1 cm distal segment into the pouch. The bottom of pouch was opened and anastomosed with the urethra. RESULTS: Forty-two patients were followed up for 6 to 96 months,90.5% of whom were continent in the daytime, and 85.7% at night. Two patients had a difficulty in urination. The average volume of the pouch was (361 +/- 48) ml at 12 months postoperation. Urodynamic examination showed the average peak voiding pressure was (86.8 +/- 21.4) cm H(2)O. The average maximum flow rate (Qmax) was (18.4 +/- 6.1) ml/s. No remarkable ureter reflux and obstruction were found. No patient was detected to have urethral carcinoma. CONCLUSIONS: Extraperitoneal cystectomy can avoid the tumor contamination of the abdomen and intestinal interference of the operative field. The ureter-inserting implantation technique is a simple anti-reflux anastomosis method with less ureter stenosis rate. Isolating the neobladder and ureters from the peritoneal cavity can reduce the postoperative complications, such as adhesive ileus, internal hernia, and urine leakage into the peritoneal cavity. The neobladder is similar to the original bladder in position, volume, shape and anti-reflux ureter connection.

Three-dimensional single molecule rotational diffusion in glassy state polymer films.

Measuring three-dimensional orientational motions of many individual molecules within glassy state poly(methyl methacrylate) has enabled nanoscopic probing of bulk-obscured polymer dynamics. Complementing bulk studies, the measured distributions of nanoscale barriers to rotational motion afforded by our single molecule orientational methods directly probe the spatial heterogeneity and nanoscopic alpha-relaxation dynamics deep within the glassy state.

Evaluation of nanostructured carbonated hydroxyapatite coatings formed by a hybrid process of plasma spraying and hydrothermal synthesis.

Carbonated hydroxyapatite (CHA) coatings on a titanium alloy were prepared by hydrothermal synthesis of precursors plasma-sprayed with brushite as a raw powder. The structures, residual stresses, and bond strengths of the precursors and CHA coatings were investigated. The results showed that the sprayed precursors consisted of beta-Ca(2)P(2)O(7), alpha-Ca(3)(PO(4))(2), and CaHPO(4), whereas the CHA coatings exhibited a unique phase construction, nanostructured and needle-like crystals, and a fairly low tensile residual stress. The bond strength of a CHA coating 200 microm thick was 15 MPa, equivalent to that of a plasma-sprayed hydroxyapatite (HA) coating. The evaluation of the CHA coatings was performed together with that of plasma-sprayed HA coatings immersed in distilled water. The dissolution and bond-strength degradation of the CHA coatings were much lower than those of the plasma-sprayed HA coatings.

In vitro and in vivo tests of hydrothermally synthesised hydroxyapatite coating.

High pure and crystalline Hydroxyapatite (HA) coatings on titanium alloy were prepared by hydrothermal synthesis (HS) of plasma-sprayed (PS) precursors from brushite powders (HS-HA). In vitro and in vivo tests were done to evaluate its biological property. The HS-HA coating was compared with the current PS-HA coating. Cultures of the primary osteoblasts on these two HA coatings showed similar cell attachment, proliferation and alkaline phosphatase (ALP) expression. The cell morphology on the coatings was demonstrated by scanning electron microscopy (SEM). The cell spread well at 1 day after seeding culture and the extracellular matrix was secreted after 14 days culture. Histomorphometric analysis was conducted on samples implanted in femoral bone of four dogs for 1 and 3 months, and bone-implant contact percentage was evaluated by light microscopy. The calcium and phosphate distribution on the interface of bone-implant was analysed by SEM and electron dispersive X-ray (EDX) analysis. The results show the osteoconduction of HS-HA coated implants.

[Microencapsulation of immortalized mandibular condylar chondrocytes].

To explore the possibility of microencapsulation of chondrocytes in cartilage tissue engineering, immortalized manibular condylar chondrocytes (IMCCs) were microencapsuled by Alginate-polylysine-alginate (APA) method, according to air pressure shearing model. Phase contrast microscopy, trypan blue staining exclusion, cell number counting, HE staining and immunohistochemistry method were used to observe the morphology of the microencapsules, the growth character of cells, cartilage characteristics, and so on. The results showed that IMCC could survive and grow in microencapsule, and the viability rate of cells is more than 80 per cent. The diameter of microcapsule is 779 microns in average. The number of cell increased with time, and cells went into platform in about 20 days. Cells grew in clusters and cartilage specific proteoglycans and type II collagen were highly expressed. It was concluded that IMCC could form cartilage-like tissue within microencapsulation, implying that microencapsule technique might be applicable to cartilage tissue engineering.
