Rapid induction of neurotrophin mRNAs in rat glial cell cultures by Semax, an adrenocorticotropic hormone analog.

The proliferation, differentiation and survival of neuronal and glial cells are affected by a number of neurotrophic factors, such as nerve growth factor (NGF), brain-derived neurotrophic factor (BDNF) and others. In a previous study, we observed the effects of 'Semax' (Met-Glu-His-Phe-Pro-Gly-Pro), the physiologically active analogue of adrenocorticotropic hormone(4--10), on neuronal cell survival in vitro. We hypothesized that these effects may be mediated by the regulation of expression of some neurotrophic factors. To test this hypothesis we analyzed NGF and BDNF gene expression in glial cells obtained from the basal forebrain of newborn rats, following in vitro treatment with 'Semax'. We observed changes in mRNA levels for both the NGF and BDNF genes. The greatest increase in expression was found after 30 min of 'Semax' administration. At this time, BDNF mRNA level was increased eight-fold in comparison with control, and NGF mRNA level was increased five-fold.

[Superhigh-dosage chemotherapy with the transplantation of autologous hemopoietic precursor cells in patients with a prognostically unfavorable relapse and resistant course of lymphogranulomatosis].

Eighteen patients with relapsed or refractory Hodgkin's disease (HD) have been treated with high-dose chemotherapy (BEAM regimen) followed by autologous peripheral stem cells and/or bone marrow rescue. There were no treatment-related deaths. Overall response rate was 82%. With a median follow-up of 10 months (3-24 months) overall survival and freedom from progression were 100 and 94% (95% confidence interval 58-97%), respectively. The use of peripheral stem cells in addition to bone marrow resulted in a significant shortening of the time to engraftment (p < 0.01). The BEAM regimen is an effective conditioning schedule which is well tolerated.

Use of tetrabutylammonium salts of amino acids in peptide synthesis.

Tetrabutylammonium (TBA) salts of amino acids and peptides have increased solubility, as compared with that of alkali metals salts, in organic solvents. We have compared the reaction rates for tripeptide formation in methylene chloride from Boc-Gly-Phe activated with various phenols, N-oxysuccinimide and azide, and TBA-salt of tryptophan, as well as Trp-OCH3. H-Trp-O-.TBA+ as an amino component significantly accelerates the rate of reaction. Although a significant degree of racemization has been found, the use of TBA-salt of amino acids and peptides is justified in many cases due to high conversion rates.

[Classification of respiratory insufficiency in pulmonary tuberculosis].

605 patients with different forms of pulmonary tuberculosis were examined. Blood gases and acid-base condition (ABC) were studied. In 22.2 per cent of the patients, hypoxemia was lacking with normal ABC and no respiratory failure (RF); in 36.6 per cent, hypoxemia was not accompanied by ABC changes and hence was caused by circulatory disturbances; in 15.6 per cent, it was combined with respiratory alcoholism; and in 6.8 per cent, with respiratory acidosis. The rest of the cases found to have metabolic disturbances of the ABC. Thus, hypoxemia is observed both with chronic forms of tuberculosis and with infiltrative/focal one, i.e. it follows a subacute course. In case of pulmonary tuberculosis, it is recommended to isolate subacute RF with PaO2 of 68.3 +/- 0.56 mm Hg, chronic RF of the 1-st phase with PaO2 of 71.8 +/- 0.7 mm Hg and chronic RF of the 2-nd phase with PaO2 of 64.2 +/- 0.88 mm Hg and accompanied by chronic cor pulmonale. In this case it seems necessary to find the basic mechanism of RF which is important for the treatment purposes.

[The effect of nitroglycerin on the external respiration function and central hemodynamics in patients with tuberculosis of the lungs].

To estimate the effect of nitroglycerin (NTG) on the external respiration function and central hemodynamics, 124 patients with various forms of pulmonary tuberculosis and chronic nonspecific diseases of the lungs were examined. The indices of the external respiration function, central hemodynamics, pulmonary artery systolic pressure (PASP), general pulmonary resistance (GPR) and blood gaseous composition were determined before and after NTG sublingual use in a dose of 1 mg. It was noted that improvement of bronchial patency after the use of NTG was not significant and subsequently its indication as an agent arresting bronchospasm is not expedient. A statistically significant decrease in PASP and the indices of central hemodynamics, as well as a decrease in GPR might be considered as a basis for the use of nitrates in prophylaxis and treatment of chronic pulmonary heart in patients with pulmonary tuberculosis.

[The effect of stressfully loud sound on motor asymmetry in rats].

In 43 female Wistar rats, a loud sound did not alter the asymmetry in behavioral tests even if is was loud enough to induce seizure. Under apomorphine test, the sound tended to decelerate the rotation, the asymmetry being preserved both in sound-activated and in sound-resistant subgroups. A shortening of the rotation onset latency occurred in sound-activated subgroup which may be associated with an increased permeability of the hemato-encephalic barrier following the sound-induced seizure.

[Chemico-enzymatic synthesis of an oligonucleotide template for an analog of ACTH (4-10) fragment].

Six oligodeoxyribonucleotides ranging from 9-mer to 13-mer were synthesized in solution by the phosphotriester approach and enzymatically joined by T4 DNA ligase. The obtained double-stranded DNA (32 b.p.) with protruding 5'-ends corresponding to the recognition sites for restrictases EcoRI and BamHI represents an oligonucleotide template coding for the modified amino acid sequence 4-10 of the adrenocorticotropic hormone, [Pro8,Gly9,Pro10]ACTH-(4-10).

[Clinical picture and treatment of epilepsy and epileptic syndromes in children (according to the results of an epidemiologic study)].

On the basis of clinical-epidemiological studies, the authors present data on the prevalence of the convulsive syndrome in children and on the characteristics of major clinical manifestations in patients with epileptic attacks: the premorbid state, age at the onset of the first paroxysms, pathomorphosis of epilepsies, type of the process development and therapeutic results. A high risk group of patients requiring appropriate preventive measures was identified. It was established that in most cases, epileptic attacks occurred in the presence of normal psychic and physical development. In 1/3 of all cases the disease first manifested itself in children under 3 years. Attention is given to the fact that the last decade has seen a decline in malignant forms with a serial course of disease. The authors discuss the results of therapeutic work and the questions of social adaptation in children suffering from epilepsy in Moscow. It is demonstrated that definite successes achieved by child psychiatric services of the city in treating epilepsy and epileptic syndromes of variable nature are consequent on modern diagnosis, proper organization of the therapeutic process as well as on the prophylaxis of exacerbation and recurrences of the disease.
