[Study on the interaction between nuclease P1 and CuCl2 (II) by visible absorption spectra].

In this paper, we studied the directly interaction between nuclease Pl and CuCl2 in aqueous solution by visible absorption spectra. As a result, the zinc ion in the active site of nuclease Pl can be replaced by additional Cu(II). At the the same time, Cu(II) entered into the active site of enzyme forming the correspondent derivatives of "Cu(II)-Pl". The variety of pH value influenced the types of derivatives and the enzyme activity.

Application of a three-point method for water-fat MR imaging in children.

BACKGROUND: Effective fat suppression is desirable in clinical magnetic resonance imaging. Conventional frequency selective fat suppression is dependent on accurate prescan shimming and is subject to artifacts due to magnetic field inhomogeneity. Quadrature three-point water-fat imaging with direct phase encoding is an alternative technique for fat suppression that has been previously described in adult volunteers and patients. OBJECTIVE: To evaluate the use of three-point water-fat imaging with direct phase encoding for fat-suppressed MR scans in children. MATERIALS AND METHODS: Sixty-two three-point water-fat imaging studies were performed in 55 children 2 months to 18 years old. T 1-weighted fat-suppressed (water) images from this sequence were compared with frequency selective fat-suppressed images obtained in 15 patients. The reliability and subjective quality of the sequence were assessed in the remaining 47 cases. RESULTS: High-quality fat suppression was achieved in all anatomic sites studied, even where frequency selective fat-suppression failed due to magnetic susceptibility artifact. The three-point water-fat sequence was visually preferred to the frequency selective fat saturation technique in 15/15 cases. CONCLUSION: Three-point water-fat imaging has replaced the conventional frequency selective technique for fat suppression on T 1-weighted MR imaging at our institution.

Infection and AIDS in adult macaques after nontraumatic oral exposure to cell-free SIV.

Unprotected receptive anal intercourse is a well-recognized risk factor for infection with human immunodeficiency virus-type 1 (HIV-1). Isolated human case reports have implicated HIV-1 transmission by oral-genital exposure. Adult macaques exposed nontraumatically to cell-free simian immunodeficiency virus (SIV) through the oral route became infected and developed acquired immunodeficiency syndrome (AIDS). The minimal virus dose needed to achieve systemic infection after oral exposure was 6000 times lower than the minimal dose required to achieve systemic infection after rectal exposure. Thus, unprotected receptive oral intercourse, even in the absence of mucosal lesions, should be added to the list of risk behaviors for HIV-1 transmission.

Epileptiform activity and hippocampal damage produced by intrahippocampal injection of guanidinosuccinic acid in rat.

Guanidinosuccinic acid (GSA) is a guanidino compound found in mammalian central nervous system and physiological fluids. Its level has been found to be greatly increased in serum and cerebrospinal fluid of patients with renal failure, and the compound is suggested to play a role in uremic encephalopathy. In this report we examined the behavioral, electrographic and morphological effects of intrahippocampal GSA injection in unanesthetized rats. Intrahippocampal administration of 2 microliters GSA solution (3.5 nM) was followed by behavior observation, and electrohippocampographic and electrocorticographic recording. GSA-injected animals showed partial clonic seizures leading to generalized clonic seizures, and eventually status epilepticus. These were accompanied by epileptiform electrographic discharges. During generalized clonic seizures, the electrohippocampogram showed arythmic bursting spikes. Epileptiform electric activity persisted even after the generalized clonic convulsions had stopped, and lasted until the animals were killed, 5 days following injection. Microscopic examination of brain slices of these rats revealed severe neural damage in CA1 area of hippocampus. Treatment of rats with the non-competitive NMDA receptor antagonist ketamine prevented both partial and generalized clonic seizures, epileptiform electrographic discharges, and GSA-induced hippocampal damage.

[Seizure activity and lesions of neuronal cells by intrahippocampal injection of guanidinosuccinic acid in rats].

Intrahippocampal injection(ihci) of guanidinosuccinic acid (GSA) to rats, induced typical generarized clonic seizures and epileptiform discharges in electrohippocampogram (EHG) and electrocorticogram (ECoG), degenerative changes of neuronal cells in the injected side hippocampus. The pyramidal cells in CA1 area were found to be more vulnerable to GSA than the granular cells. Phenobarbital and phenytoin are typical antiepiletics, but in no case did they successfully protect against GSA induced convulsions, epileptiform discharges in the EHG and ECoG and neurolysis. Ketamine, a selective noncompetitive NMDA receptor antagonist, was shown to protect against not only seizures, but also neuronal cell damage induced by GSA. All these results indicate that GSA very like the endogenous excitatory amino acid, glutamic acid, it also has such effects mentioned above. Therefore, the NMDA receptor may mediate both effects of GSA.

Water-fat imaging with direct phase encoding.

A new method is introduced for water-fat imaging. With three acquisitions, a general direct phase encoding (DPE) of the chemical shift information is achieved. Pixels containing both water and fat are solved directly. Pixels with only a single component are resolved with local and global orientation filters, which use phase information from neighboring pixels. The fact that a single component is more likely to be water than fat in living tissues is also useful. A second pass solution yields water and fat images with superior signal-to-noise ratio. Unlike other methods, DPE does not rely on the error-prone phase unwrapping; also, it easily handles disconnected tissues. Because the magnetization vectors of water and fat are sampled not only at parallel or antiparallel, they can be not only separated but also identified respectively, which is desirable for routine clinical work. DPE has been implemented on several imagers at various field strengths and has been demonstrated in a large number of clinical cases to be useful and robust in various parts of the body.
