Action of immobilized xanthine oxidase on purines.

Xanthine oxidase was covalently immobilized on polyacrylamide gel beads, polyamide-11 and dacron. Hypoxanthine (15 ml of 200 microM), prepared in 0.1 M phosphate buffer, pH 8.0, was circulated through a column containing 1.0 g derivatized enzyme at a flow rate of 1.0 ml/min at 28 degrees C. Specific activities of 0.660, 0.072 and 0.016 Units/mg of protein were demonstrable for the polyacrylamide gel beads, dacron and polyamide-11 derivatives, respectively. The action of these water insoluble enzyme derivatives on 6-mercaptopurine (15 ml of 660 microM) was also investigated, under the same experimental conditions, showing specific activities of 0.063 Units/mg, 0.574 muUnits/mg and 0.118 muUnits/mg, respectively. The 6-mercaptopurine oxidative pathway catalyzed by immobilized xanthine oxidase on dacron stopped at the intermediate compound, 6-mercapto-8-hydroxypurine, so that no 6-thiouric acid was produced, whereas the immobilized preparations using polyacrylamide gel beads and polyamide-11 behaved like the soluble enzyme, namely, 6-thiouric acid was the final product. The behavior of dacron-xanthine oxidase compound was similar to that previously described for the derivatives obtained with carboxymethylcellulose and chitosan. The hypoxanthine oxidative pathway catalyzed by xanthine oxidase immobilized on these three supports was similar to the soluble enzyme.(ABSTRACT TRUNCATED AT 250 WORDS)

L-malic acid production by entrapped Saccharomyces cerevisiae into polyacrylamide gel beads.

The yeast Saccharomyces cerevisiae was entrapped within polyacrylamide gel beads by employing a procedure that uses sodium dodecylsulfate as a detergent to improve the spherical configuration of the beads. The resulting preparation showed a rate of fumarate bio-conversion to L-malic acid about 60 times higher than that found for the free cells. Almost all fumarate was converted in 30 min of incubation. The thermal stability of the immobilized cells did not significantly differ from the free cells. An optimal pH of 5.7 was found for the immobilized preparation and no succinic acid was detected as a byproduct in the incubation mixture.

Human T cell leukemia virus type 1 (HTLV-1) antibodies in healthy populations and renal transplanted patients in the north-east of Brazil.

The seroprevalence of human T cell leukemia virus type 1 (HTLV-1) infection was investigated in Brazilians (570): native inhabitants (298) and descendants from Japanese (272) living in Recife and its neighborhoods--North-east of Brazil. Furthermore, polytransfused renal transplanted patients (54) were also examined for the serological status to this virus. The seropositivity to HTLV-1, screened by enzyme-linked immunosorbent assay (ELISA), was low: 1.34% for the local population and 0.73% for the descendants from Japanese. However, the seropositivity for the renal transplanted patients was found to be 11.1%. This higher value suggests that this retrovirus infection seems to be of importance in this clinical condition.

Cytomegalovirus, human herpesvirus 6 and human T cell leukemia virus infection in renal transplanted patients in the northeast of Brazil.

The seroprevalence of antibodies against cytomegalovirus (CMV), human herpesvirus 6 (HHV-6) and human T cell leukemia virus type 1 (HTLV-1), determined by ELISA and by fluorescence in 54 renal transplanted patients, was 96.2%, 88.8% and 11.1%, respectively. These values are relatively high when compared with the results obtained for healthy individuals of the same age groups from Recife, Northeastern Brazil. Active CMV infection was detected by the presence of IgM antibodies and/or virus isolation in 13 (24%) patients. Kidney rejection and renal dysfunction were observed in 11 of these 13 patients, whereas 3 of 6 HTLV-1 antibody-positive individuals presented these complications. All HTLV-1 positive patients were also positive to IgG CMV and HHV-6 antibodies. The importance of the three viruses in this clinical condition is suggested by the high seropositivity rates compared with the healthy population. The group may also represent a potential source of HTLV-1 infection in this non-endemic area.

L-malic acid production using immobilized Saccharomyces cerevisiae.

L-Malate was produced from fumarate by using immobilized Saccharomyces cerevisiae cells entrapped in polyacrylamide. This preparation performed better when pretreated with malonate. Under the experimental conditions described here, succinate was not detected as a by-product of the reaction, as had been reported for other microorganisms.

Immobilization of protein on ferromagnetic Dacron.

Ferromagnetic Dacron (polyethyleneterephthalate) is proposed as a matrix to immobilize proteins covalently. Dacron in powder was magnetized by reacting ferrous (Fe+2) and ferric (Fe+3) ions with its hydrazide form at pH 8.3. Ferromagnetic hydrazide Dacron was converted to ferromagnetic azide Dacron and amyloglucosidase (E.C. 3.2.1.3) was covalently bound through the latter group. The catalytic property of the enzyme was preserved (8% of the specific activity estimated for the soluble enzyme) and all the magnetic amyloglucosidase Dacron derivative was recovered by using a magnetic field. No activity was detected in the supernatant.

IMP synthesis using immobilized adenosine (phosphate) deaminase.

Inosinic acid (IMP) has been prepared by the deamination of adenosine monophosphate (AMP) with an immobilized adenosine (phosphate) deaminase extracted from the snail Biomphalaria glabrata. The enzyme has been immobilized in polyacrylamide beads. The preparation and characterization of this system are described.

Activity of immobilized alpha-amylase.

1. alpha-Amylase immobilized on polyamide 11 showed higher specific activity and retention of activity than the derivatives employing polyacrylamide and polyethyleneterephthalate as supports. 2. Polyamide 11 and polyethyleneterephthalate alpha-amylase derivatives exhibited a higher extent of multiple attacks on starch than the water-soluble enzyme whereas the polyacrylamide derivative presented less. 3. The polyamide 11 alpha-amylase derivative acted on amylose-azure in the same way as the water-soluble alpha-amylase.

Prevalence and pathogenicity of Entamoeba histolytica in three different regions of Pernambuco, northeast Brazil.

Parasitological examinations were carried out on 663 individuals of three different cities of Pernambuco State, Northeastern Brazil: Recife, Palmares and Bodocó. The population from a drought area of Pernambuco State, Bodocó, was investigated for amoebiasis and compared with Recife, metropolitan city (about 1.3 million of inhabitants) and another inland community, Palmares, located inside of the sugar-cane plantation region of the State. No evidence of invasive strains of E. histolytica were found in these inhabitants, provided that the isolated zymodemes I, III, IV, VIII, IX, X, XVII and XVIII are recognized as nonpathogenic strains of E. histolytica. Furthermore, the prevalence of intestinal helminths and other protozoan infections showed that these individuals are infected by other agents responsible for diarrhoeal diseases.

Standardization of the dot enzyme-linked immunosorbent assay (dot-ELISA) for experimental plague.

A dot enzyme linked immunosorbent assay (dot-ELISA) was previously developed to detect specific antibodies in rabbits sera immunized against F1A protein obtained from Yersinia pestis. This antigen was covalently linked onto the surface of dacron (polyethyleneterephthalate). Here, standard conditions are described for the optimization of this procedure: an amount of 20 ng of F1A protein was fixed onto dacron; anti-rabbit IgG peroxidase conjugate diluted 1:8,000 and 30% non-fat instant milk as blocking substance were used throughout the method. This procedure was compared with that employing nitrocellulose as solid-phase which showed to be more sensitive. However, the method based on dacron did not show false positive reactions against non-immunized rabbits sera at low antigen amount and diluted anti-IgG peroxidase conjugate.

The use of ferromagnetic Dacron as solid-phase in enzyme immunoassays.

Ferromagnetic dacron is proposed as an alternative solid-phase for magnetic enzyme immunoassays. Human serum albumin (HSA) was covalently immobilized onto ferromagnetic dacron and an enzyme immunoassay was developed using anti-HSA rabbit sera. Peroxidase, o-phenylenediamine (OPD) and hydrogen peroxide were used as the enzymatic label and substrates, respectively. Best results were observed when particles of 63-100 microns (diameter) and 10 micrograms of immobilized antigen were used. Positive reactions were detected until dilutions of 1:51200 of immune sera. Its reproducibility was similar to standard ELISA. Disruption of the immunocomplexes formed and recuperation of the immobilized antigen in other immunoassays also proved to be reliable.

The use of polyvinyl alcohol glutaraldehyde as solid-phase in ELISA for plague.

Discs of polyvinyl alcohol cross-linked with glutaraldehyde were synthesized under acid catalysis (H2SO4). Then, the antigen F1 purified from Yersinia pestis was covalently linked to this modified polymer. Afterwards, an enzyme-linked immunosorbent assay (ELISA) was established for the diagnosis of plague in rabbit and human. The best conditions for the method were achieved by using 1.3 micrograms of F1 prepared in 0.067 M phosphate buffer, pH 7.2, containing 1 M NaCl (PBS); anti-IgG peroxidase conjugate diluted 6,000 times and as a blocking agent 3% w/v skim milk in PBS. The titration of positive rabbit serum according to this procedure detected antibody concentrations up to 1:12,800 times. The present method, the conventional ELISA and passive haemagglutination assay are compared.
