Complete remission in prolymphocytic leukemia with 4-demethoxydaunorubicin and arabinosyl cytosine.

Prolymphocytic leukemia (PL) is a morphologically distinct disease generally characterized by unsatisfactory therapeutic response and brief survival. Aggressive chemotherapy protocols including doxorubicin (DX) have been successfully used as alternatives to treatments usually effective in chronic lymphocytic leukemia (CLL). A patient with typical PL, resistant to regimens containing DX, was treated with 4-demethoxydaunorubicin (4-dm DNR), a new anthracycline analog derived from daunorubicin (DNR). The therapeutic response was rapid and impressive; furthermore 4-dm DNR combined with arabinosyl cytosine (Ara-C) produced a complete remission which is unusual in PL.

Effect of polyethylene glycol 400 on adriamycin toxicity in mice.

The effect of a widely used organic solvent, polyethylene glycol 400 (PEG 400), on the toxic action of an acute or chronic treatment with adriamycin (ADR) was evaluated in mice. PEG 400 impressively decreased both acute high-dose and chronic low-dose-ADR-associated lethality. Light microscopic analysis showed a significant protection against ADR-induced cardiac morphological alterations. Such treatment did not diminish the ADR antitumor activity in L1210 leukemia and in Ehrlich ascites tumor.

Enhancement of paracetamol induced hepatotoxicity by prior treatment with carboxymethylcellulose.

This study was undertaken to determine if interactions of toxicological significance might occur during treatment of mice with paracetamol (P) and carboxymethylcellulose (CMC). P (300 mg/kg) administration induced only slight hepatic morphologic changes and lowered hepatic reduced glutathione (GSH) to 40% of the level in controls, when measured 1 hour after treatment. Intraperitoneal pretreatment with CMC (1% in NaC1, 20 ml/kg), significantly decreased GSH concentration (to 19% of controls) and severely enhanced hepatic injury. Such an interaction is of considerable significance since both CMC and P are widely used substances in experimental pharmacology and clinical practice.

Clinical stage and immunological findings in chronic lymphocytic leukemia.

The total and T-lymphocyte counts, different types of lymphocytic surface immunoglobulins and levels of serum immunoglobulins were studied in 112 consecutive patients with chronic lymphocytic leukemia in order to investigate possible correlations between the immunological picture and the different stages according to the staging criteria recently proposed by Binet et al. Patients were also subdivided according to the form of their disease, "indolent" or "active", and correlations with the immunological picture investigated. The total lymphocyte count showed a significantly increasing trend through the three different stage (p less than 0.01), and it was higher in the "active" than in the "indolent" disease (p less than 0.01). Also the number of T-lymphocytes increased in the "active" forms (p less than 0.01). No statistically significant correlations were observed between the different surface immunoglobulin subclasses and the clinical pattern. As regard the various classes of serum immunoglobulins, the number of patients, with a reduction of at least one of the classes tended to increase with worsening of the clinical stage.

Enhancement of adriamycin toxicity by carboxymethylcellulose in mice.

Carboxymethylcellulose (CMC) (1% in 0.9% NaCl, 0.2 ml/10 g ip) a common suspending agent, enhanced adriamycin (ADR) (15 mg/kg ip) toxicity when administered to mice 5 hr before the antibiotic. Compared with ADR alone, this combination treatment produced, after 7 days, an increase in lethality from 15 to 80%. The pathologic analysis of hearts, livers, kidneys, and small bowels was performed, revealing an increase in the incidence and severity of hepatic damage in mice receiving ADR + CMC. Furthermore, reduced glutathione (GSH) was measured in livers of all mice; the animals treated with CMC and ADR + CMC showed a significant (p less than 0.01) reduction of hepatic GSH in comparison with controls and ADR-alone-treated animals. These data further confirm a crucial protective role for GSH in ADR toxicity and prove that CMC exerts an important biochemical effect on hepatic GSH.

Effects of ICRF 159 on adriamycin-induced cardiomyopathy in rats.

The effect of ICRF 159 on adriamycin (ADR) cardiotoxicity and on total myocardial calcium content was examined in rats. ICRF 159 did not increase the survival time of ADR-treated animals; however the histological findings showed a significant prevention of ADR-induced cardiomyopathy (CMP) by ICRF. The total myocardial calcium content of animals treated with ADR was significantly higher, while no significant difference was seen in animals pretreated with ICRF 159 as compared with controls. As these findings suggested a role of calcium in ADR CMP and in the pharmacological action of ICRF in this disease, we also tested a closely related chelating agent, EDTA. This molecule decreased myocardial calcium levels in ADR-treated animals almost to normal values; however the histological cardiac alterations were not prevented.

Antitumor activity of N-diazoacetyl derivatives of glycine and phenylalanine against P388 leukemia and B16 melanoma in mice.

N-Diazoacetyl derivatives of glycine and phenylalanine show antitumor activity in mice bearing P388 leukemia or B16 melanoma. The presented data indicate that antitumor activity is shown by diazomethylamide derivatives of glycine and phenylalanine, in addition to that already established for the amino acid derivatives having an O-diazoacetyl group (azaserine) or a diazoketone structure (DON and azotomycin) and for 1,2-bis-diazoacetyl ethane.

Presence of circulating immune complexes in patients with chronic lymphocytic leukaemia. Correlations between clinical and immunological features.

The presence of circulating immune complexes (CIC) was evaluated in 60 untreated patients with chronic lymphocytic leukaemia with the polyethylene-glycol precipitation assay. The mean values of optical density (OD) in patients' sera was 0.161 +/- 0.121 compared with control values of 0.082 +/- 0.045 (P less than 0.001). Sera of 16 patients gave OD values higher than 0.172 (mean of controls plus 2 SD) and were considered positive for the presence of CIC. Using the new staging proposed by Binet for CLL we did not find any correlation between presence of CIC and stage of the disease. On the contrary, it was possible to correlate the presence of CIC with the stability of the disease: CIC were found in only 2 of the 22 patients (9.1%) with progressive disease but in 10 of the 30 patients (33.3%) with stable disease (P less than 0.05). CIC were present in 12 of the 38 patients in whom the only surface immunoglobulins present were IgM, but in only 1 of the 14 patients in whom both IgM and IgD were contemporaneously present on the lymphocyte surface. No correlations were found between levels of serum immunoglobulins and presence of CIC.

[Some parameters of cellular immunity in renal transplantation after more than three years].

This paper evaluates the cellular immune system of 16 cadaver kidney transplant recipients more than three years after transplantation. The number and percent of active E rosette forming cells, of E rosette forming cells and of IgG and IgM bearing lymphocytes were determined; the blastogenic response of peripheral blood lymphocytes to PHA and PWM was studied in homologous and autologous serum. Significant alterations of the tests performed were found in patients who had undergone kidney transplant, suggesting a pathogenetic role of chronic immunosuppressive therapy but not excluding other possible factors.
