Membrane cofactor protein (MCP or gp45-70) is a recently described regulatory glycoprotein of the complement system which binds iC3 or C3b and is present on human platelets, T cells, B cells, monocytes, and mononuclear-derived cell lines. On sodium dodecyl sulfate-polyacrylamide gel electrophoresis, MCP migrates as a doublet with an Mr of the upper band of 63,000 and the lower band of 58,000. The same pattern was found on all cell populations in a given individual and was stable over time. In order to further characterize the two band pattern on sodium dodecyl sulfate-polyacrylamide gel electrophoresis, MCP was isolated by affinity chromatography or immunoprecipitation from 72 healthy unrelated donors. All individuals expressed both bands and, based on the densitometric scanning of gels, three patterns were noted: upper band predominant in 65%, approximately equal distribution of upper and lower bands in 29%, and lower band predominant in 6%. These observed phenotypic frequencies fit with expectations based on Hardy-Weinberg equilibrium for a two-allele system. Family studies also support this model as none of the 26 offspring had a phenotype that deviated from the expected, assuming an autosomal codominant model of inheritance. These results are consistent with a simple two-allele system that controls the expression of the two bands of MCP.
A primary genetic map for human chromosome 12 has been constructed from data on 23 restriction fragment length polymorphic systems collected in 38 normal families with large sibships. Linkage analysis of the genotypic data has ordered 16 loci into a continuous genetic map of 111 cM in males and 258 cM in females. Although most of the genetic map reflects a higher rate of recombination in females relative to males, significantly more frequent recombination was observed in males than in females in intervals between loci on the distal portion of the short arm of the chromosome. The mapping data shown here will serve as a first step toward a high-resolution genetic map for human chromosome 12.
Ninety infertility patients with moderate endometriosis were randomized between laparoscopic electrocautery and 6 months of danazol therapy and studied for 7 months after treatment. Twenty of the 45 patients undergoing electrocautery conceived (44%) during the follow-up interval. Forty-one of the 45 patients in the danazol group completed the course of therapy, and 16 of those conceived (39%) during the same follow-up interval. There was no statistical difference between the two groups (P less than 0.53). No patients in the electrocautery group experienced complications. This demonstrates that electrocautery is safe and effective in the treatment of moderate endometriosis.
Utilizing affinity chromatography, a C3-specific binding protein was isolated from 125I surface-labeled human platelets. Analysis by sodium dodecyl sulfate-polyacrylamide gel electrophoresis demonstrated two bands with mean Mr of 64,000 and 53,000, characteristic variability in the relative density of the two bands in a given individual, and the presence of N-linked complex oligosaccharides as well as sialic acid residues not associated with N-linked sugars. These characteristics are similar to those of a human leukocyte iC3- and C3b-binding glycoprotein, termed gp45-70. Further analysis showed that leukocyte gp45-70 and the platelet C3-binding glycoprotein have identical Mr and other similar structural features. Functional characterization of solubilized platelet preparations indicated that gp45-70 has cofactor activity. This membrane glycoprotein is structurally and antigenically distinct from decay accelerating factor (DAF), a complement regulatory protein previously identified on human platelet membranes. DAF and gp45-70 have complementary activity profiles inasmuch as DAF can prevent assembly of and dissociate the C3 convertases but has no cofactor activity, whereas gp45-70 has cofactor activity but no decay accelerating activity. We suggest that these two proteins function conjointly to prevent autologous complement activation.
