Fate of clonal lineages during neoplasia and metastasis studied with an incorporated genetic marker.

The fate of clonal lineages in tumor formation and metastasis has been studied by genotypic marking of cells from three separate tumor lines of different malignant potential. Marking was accomplished by random incorporation of the neomycin resistance gene and visualized by Southern blot analysis of integration sites. Primary tumors formed by polyclonal cell suspensions of all three cell lines injected s.c. usually remained polyclonal even at late stages of tumor growth and metastatic spread. Lung metastases were often clonal, but it was not unusual to find ones of polyclonal origin. Lymph node metastases were almost always polyclonal and remained so, as they grew large. Sometimes clones present in the original inoculum were absent in the primary. Other times clones visible in the metastases were undetectable in the corresponding primary tumor. Occasionally a single clone became dominant in the primary, and others were eliminated, but this was not a necessary prelude to the onset of invasive or metastatic behavior. It is concluded that there is considerable variation in the results obtained with various cell lines in different circumstances. Even clones which are underrepresented in the original inoculum or the primary tumor can acquire metastatic capability. Hence, progression of malignancy is not uniformly dependent on prior or concurrent extinction of other non- or less metastatic clones in the neoplasm, and the underlying mechanisms of invasion and metastasis can be separated from those which sometimes confer growth supremacy on a clone of tumor cells. The frequent continuing genetic heterogeneity of cells in a neoplasm has substantial implications for clinical treatment protocols.

Mapping of genes for the human C5a receptor (C5AR), human FMLP receptor (FPR), and two FMLP receptor homologue orphan receptors (FPRH1, FPRH2) to chromosome 19.

The genes encoding receptors for the chemotactic ligands C5a (C5AR) and FMLP (FPR) were mapped using a panel of somatic cell hybrids to chromosome 19. Because the hybridization pattern on Southern analysis suggested an intron structure or related genes in the case of FPR, genomic clones were characterized. Two structural homologues of the FMLP receptor, clones 81 (FPRH1) and 82 (FPRH2), were identified, which similarly map to chromosome 19. The structural homologues do not recognize the ligand FMLP, but are likely chemotactic receptors.

[The detection and analysis of leptospiral DNA in patients' serum of early leptospirosis by polymerase chain reaction and DNA hybridization with digoxigenin-AMPPD].

We have detected and analysed the leptospiral DNA in serum of patients with early Leptospirosis from the epidemic area of China by PCR and DNA hybridization with Digoxigenin (Dig)-3-(2'-Spiroadamantane)-4-methoxy-4-(3"-phosphoryloxy)- phenyl-1,2-dioxetane (AMPPD) to develop a sensitive, specific and reliable technique for the early diagnosis of leptospirosis, and full satisfactory results have obtained. Fourteen serum specimens from patients with leptospirosis proven by blood culture and serological test were prepared according to Boom's methods for PCR test, and oligonucleotide primers, named G1 G2, were obtained from a genomic library of leptospira interrogans. PCR amplification with serum specimens was performed. Each cycle of amplification consisted of denaturation at 94 degrees C for 1 min, annealing at 55 degrees C for 1 min and elongation at 72 degrees C for 2 min. Each sample was subjected to 32 cycles. The amplified DNA were separated by electrophoresis with 2% agarose gel and hybridized with the homologous DNA probe labelling with Dig-AMPPD by means of Southern blotting. All of 14 samples revealed the presence of leptospira and the strong signals were visualized with homologous DNA probe hybridization by Southern blotting. Negative and positive controls appeared correctly. The DNA fragment generated from PCR amplification homologically hybridized with the DNA of 16 strains which are from Yasudas' genomic species and represent the different genomic groups of leptospires. The single recognized band (about 400 bps) from 6 out of the 16 strains has come out which are representative of the principal strains in Sichuan, China.(ABSTRACT TRUNCATED AT 250 WORDS)

Obstinate cases of stromal keratitis treated by combined traditional Chinese and Western medicine.

Twenty-two cases 23 eyes with obstinate stromal keratitis treated by combination of traditional Chinese and Western medicines are reported in this paper. According to clinical manifestations, they are consistent with the clinical diagnosis of stromal herpes simplex keratitis. Since no specific Western medicine is known to be effective in treating this disorder, it is proposed to apply systemic and topical TCM and, if necessary, add beta-ray irradiation to shorten the clinical course of disease, decrease or prevent recurrence and maintain useful visual acuity, avoiding frequent relapses leading to blindness.

[Detection of the microquantity DNA of Leptospira interrogans by polymerase chain reaction].

Leptospirosis is a severe zoonosis in the world. The methods for detecting leptospira are not sensitive and specific so far. The problems in early diagnosis and epidemiological identification of Leptospirosis remain unsolved. Two recombinant DNA fragments of serogroup Icterohaemorrhagiae, Leptospira were selected by repeated molecular cloning and screening in this study firstly. One of them can hybridize with the DNA of various serogroups of Leptospira interrogans; the other can only hybridize with the DNA of serogroup Icterohaemorrhagiae. After the nucleotides sequence analysis, from these 2 recombinant DNA fragments, 2 pairs of polymerase chain reaction (PCR) oligonucleotide primers were synthesized, named primer B 1, 2 and primer B 3, 4 PCRs were carried out with these 2 primers for detecting the microquantity (0.1 ng) of various serovars, serogroup of leptospires. All the DNA of Leptospira interrogans can be amplified by primer B 1, 2, and only the DNA of serogroup icterohaemorrhagiae, Leptospira reacted specially with primer B 3, 4. The DNA of non-pathogenetic Leptospira and some other microbes, however, had no amplification at all. This study is first reported at home and abroad. The results demonstrate that PCR is a very sensitive and specific technique of DNA amplification, which can be used as a powerful tool in the early diagnosis and epidemiological identification of leptospirosis.

[DNA hybridization and identification of Leptospira serogroup icterohaemorrhagiae with Leptospira recombinant DNA probes].

Two recombinant DNA probes-PLIpso1 (15kb) and PLIEc34 (4kb), derived from Leptospira serovaricterohaemorrhagiae genomic libraries, were applied for the hybridization and identification of 13 strains of Leptospira in serogroup icterohaemorrhagiae. Difference in hybridization signal in combination with the banding pattern provide a good way for identification of serovars and strains. The recombinant DNA, specific to L. serogroup icterohaemorrhagiae, hybridized with a limited number of DNA fragments which had been digested by several restriction endonucleases. The less complex banding pattern and higher sensitivity facilitate characterization of various serovars and strains in serogroup icterohaemorrhagiae. In general the DNA patterns recognized by both probes have extensive genomic homology in same serovar (but still can distinguish strains in same serovar by some unique bands) and apparent difference in various serovars (especially serovars naam, nanxi and honghe). The results indicated that Southern blotting with recombinant DNA probe might provide tools for identification, characterization and analysis of leptospira.

[Effects of tiao-zhi-tang on lipoprotein and apolipoprotein in hyperlipemic patients].

In this research, 68 patients suffering from hyperlipemia were divided into observation group taking Tiao-Zhi-Tang (TZT) and control group taking inositol and Mai Tong at random and matched-pair, and the serum levels of HDL-c, HDL2-c, LDL-c, Apo A-I and Apo B were measured respectively by the methods of polyethylene glycol precipitation separation and single radial immunodiffusion. The results showed that there were significant difference in the levels of lipoprotein, apolipoprotein between the patient groups and the healthy group before taking medicines. After taking medicines for four weeks, the serum levels of total cholesterol, triglyceride (TG), LDL-c and Apo B were lowered and the serum levels of HDL-c, HDL2-c and Apo A-I were elevated in the observation group. The serum level of TG was lowered and the serum levels of HDL2-c, Apo A-I were elevated in the control group. The effects of TZT, with the serum levels of LDL-c and Apo B being lowered and the serum level of HDL-c being elevated, were more beneficial than inositol and Mai Tong. These results suggested that TZT had a good effect on regulating the lipoprotein metabolism.

[Endocytosis and cytotoxin effect induced by the attachment of the glycolipoprotein of leptospires to vascular endothelial cell in culture].

The Glycolipoprotein (GLP) of various virulent leptospires was extracted, purified and identified. The GLP was combined with fluorescence rhodamine B and the colloidal gold as the probe. Meanwhile, the bovine aortic endothelial cell (EN) was isolated and cultured in the medium 199 "Nissui". The GLP-rhodamine B and GLP-colloidal gold probe were added to cultured EN and incubated at 4 degrees C for 2 h then at 37 degrees C for 2 h. The interaction between GLP-rhodamine B, GLP-CG probe and EN were observed and photography by fluorescence microscopy (Leitz) and scanning electronmicroscopy. The releasing rate of lactic acid dehydrogenase (LDH), acid phosphatase (AcP) and protein from the EN were detected. The results indicated that the GLP of Leptospira interrogans serovar Lai, strain 017 attached to EN and a endocytosis was happened subsequently. The LDH and AcP releasing rate were significantly higher than control and the cell protein was much less than control. The results showed that the GLP of L. interrogans strain 017 had the cytotoxin effect through a specific attachment, endocytosis in EN. The endocytosis probably is a receptor-mediated endocytosis.

RhoC GTPase overexpression modulates induction of angiogenic factors in breast cells.

Inflammatory breast cancer (IBC) is a distinct and aggressive form of locally advanced breast cancer. IBC is highly angiogenic, invasive, and metastatic at its inception. Previously, we identified specific genetic alterations of IBC that contribute to this highly invasive phenotype. RhoC GTPase was overexpressed in 90% of archival IBC tumor samples, but not in stage-matched, non-IBC tumors. To study the role of RhoC GTPase in contributing to an IBC-like phenotype, we generated stable transfectants of human mammary epithelial cells overexpressing the RhoC gene, and studied the effect of RhoC GTPase overexpression on the modulation of angiogenesis in IBC. Levels of vascular endothelial growth factor (VEGF), basic fibroblast growth factor (bFGF), interleukin-6 (IL-6), and interleukin-8 (IL-8) were significantly higher in the conditioned media of the HME-RhoC transfectants than in the untransfected HME and HME-beta-galactosidase control media, similar to the SUM149 IBC cell line. Inhibition of RhoC function by introduction of C3 exotransferase decreased production of angiogenic factors by the HME-RhoC transfectants and the SUM149 IBC cell line, but did not affect the control cells. These data support the conclusion that overexpression of RhoC GTPase is specifically and directly implicated in the control of the production of angiogenic factors by IBC cells.
