Effect of irradiation on the GTP binding kinetics of chicken embryo brain plasma membranes.

The membrane bound form of the catalytic subunit of adenylate cyclase of chicken embryo brain has been found earlier to be rather radioresistant [28]. The radiation induced changes of G proteins of membrane preparations of 19 day old chicken embryo brains were investigated in this work. The activation of catalytic subunit of adenylate cyclase by G protein dependent activators (Gpp/NH/p and NaF) was found elevated at lower radiation doses (0-400 Gy), while both basal enzyme activity (measured without any activator) and the activity measured in the presence of activators decreased at higher doses (above 800 Gy). Heterogeneity of GTP binding sites measured with 3-H-Gpp/NH/p a GTP-ase resistant GTP analogue was observed in the case of control (with Kd1 = 0.0663 +/- 0.034 mumol/l, Bmax = 0.0079 +/- 0.0022 nmol/ml and Kd2 = 2.038 +/- 0.4779 mumol/l, Bmax2 = 0.0291 +/- 0.0017 nmol/ml). The lower affinity high capacity binding sites seemed to be more radiosensitive, than the higher affinity sites. A marked decrease was observed in the number of low affinity binding sites above 200 Gy and these low affinity binding sites practically disappeared after irradiation with 400 Gy. At high doses (above 1600 Gy) the catalytic subunit was damaged, too. On the basis of the decrease of low affinity binding sites together with an increase in activation of the catalytic subunit via G proteins one can conclude that it is caused by radiation induced damage of Gi protein that can be more radiosensitive, than Gs protein.

Endotoxins and prednisolone alter replication of type 5 adenovirus and its temperature sensitive mutants.

Latency, replication or transformation by adenoviruses require cooperation between their gene products and cellular factors, which are controlled by external stimuli. Clinical observations suggest that bacterial endotoxin (LPS) and steroid hormones have direct effects on the viral permissivity and activation. Therefore, HEp-2 cultures were infected with low multiplicity of wild type (WT) human adenovirus 5 (Ad-5) and its temperature-sensitive mutants ts18 and ts19 damaged in the phosphorylation of structural polypeptides VI and X at 39 degrees C, respectively. Cultures were treated at permissive (32 degrees C) and nonpermissive (39 degrees C) temperatures with native and radio-detoxified (RD) LPS, alpha-tocopherol and prednisolone alone or in combination. Titration of virus yields showed dose-dependent activation and enhanced replication of latent WT and ts mutants by both LPS preparations. alpha-Tocopherol diminished these processes. LPS was likewise unable to augment virus producing capacity of cells. Prednisolone, although activating no latent virus, resulted in augmenting Ad-5 production at 32 degrees C. No compounds activated mutants at 39 degrees C except synergistic effect of LPS and prednisolone resulted in limited but significant replication of mutant ts19. Deliberation of lysosomal enzymes, enhanced cGMP and tumour necrosis factor alpha (TNF-alpha) productions by LPS as well as interaction of Ad early gene expression with prednisolone-utilized cellular transcription factors including AP-1 (c-jun, c-fos) are implicated in these processes. Excluding the role in activation of Ad proteinase processing viral structural polypeptides draws attention to the importance of cellular factors in virus replication.

The effect of X-radiation on reticulo-endothelial system and its treatment with radiodetoxified-endotoxin and trace elements in rats.

A new in vivo method has been developed for the precise observation of RES activity. Both Escherichia coli endotoxin 100 micrograms/100 g i.v. (LPS) and radiodetoxified endotoxin 100 micrograms/100 g body weight i.v. (RD-LPS, TOLERIN) both increased the granulopectic activity of RES. The RD-LPS was more effective. The preparation containing trace elements also increased the activity of RES. The treatment consisting of the use of both trace elements and RD-LPS proved to be the most effective. The activity of RES was inversely proportional to various doses of X-ray irradiation (7, 8, 9 Gy). Trace elements and RD-LPS even improved the immunity system of animals having deteriorated RES.

Effect of radiodetoxified endotoxin and trace elements on reticulo-endothelial system damaged by ethanol in rats.

For the demonstration of RES activity 99-mTc labelled nano-albumon micro-colloid solution (prepared by OSSKI, Budapest, Hungary) was injected into the prepared jugular vein of narcotized rats and the clearance rate was investigated. The effects of various materials have been observed on RES in vivo. Continuous consumption of 20% ethanol solution for 15 weeks decreased the phagocytic activity. One hundred microgram/100 g body weight (i.v.) of the radio-detoxified endotoxin preparation (RD-LPS, TOLERIN) prepared from Escherichia coli endotoxin by 60Co-gamma irradiation (150 KGy) increased the granulopectic activity of RES of intoxicated animals. The preparation containing trace elements also increased the former activity and the treatment consisting of the use of both trace elements and RD-LPD was as effective as the RD-LPD per sc.

Adenylate cyclase-the more membrane associated, the less radiosensitive.

A dose dependent but not parallel decreases were observed both in SH content and catalytic activity of "free" catalytic subunit after irradiation (0-3200 Gy), while SH groups of membrane-associated adenylate cyclase were insensitive (under 3200 Gy). An initial "radioactivation" of membrane-associated enzyme was found under 800 Gy, then an inhibition above 1600 Gy. The SH alkylating agent, N-ethylmaleimide resulted in a complete inactivation, both of membrane associated form of adenylate cyclase and "free" catalytic subunit with similar inactivation profiles. These data indicate that in the radiosensitivity or "radioprotection" of adenylate cyclase, its membrane association/integration might play a more important role than the SH groups themselves.

Modifying effects of the parent and radio-detoxified endotoxins on cell-mediated cytotoxicity and cytokine release.

The cytotoxic effects of the parent and radio-detoxified lipopolysaccharides (LPS and RD-LPS, respectively) were studied at various concentrations (0-50 micrograms/ml) upon cultured human lung carcinoma target cells. There was no significant difference between the effects of the two endotoxins. LPS and RD-LPS at the same concentrations, however, modified the cytolytic activity of human leukocytes (effector cells) and their mediators in different extent. The most remarkable difference was found at 10 micrograms/ml. A much higher cytotoxic activity of effector cells was observed in the case of RD-LPS as compared to the parent LPS. This concentration used for the treatment of X-irradiated and unexposed effector cells at 24 h incubation resulted in elevated release of some cytokines as measured by ELISA. For increasing the natural defence, RD-LPS as immunomodulator may be of practical value.

Lipopolysaccharide is able to bypass corticotrophin-releasing factor in affecting plasma ACTH and corticosterone levels: evidence from rats with lesions of the paraventricular nucleus.

Stimulation of the immune system or experimental conditions (bacterial lipopolysaccharide (LPS) treatment) provoke a broad spectrum of physiological responses. It was recently shown that one of them is the activation of the hypothalamic-pituitary-adrenal (HPA) axis. The mechanism and the site or sites through which LPS stimulates the HPA axis are not well understood. To establish whether the effect of bacterial LPS is related in vivo to the presence of hypothalamic hypophysiotrophic peptides (corticotrophin-releasing factor-41, arginine vasopressin, etc.), plasma ACTH and corticosterone levels were monitored in intact and sham-operated rats, and in rats with paraventricular nucleus lesions in order to remove the main source of these neuropeptides. Evidence was obtained that 4 h after treatment, LPS was able to activate the hypophysial-adrenal system in the absence of hypophysiotrophic neuropeptides of paraventricular origin. It is suggested that, in vivo, LPS could have a direct effect on the pituitary gland or that it acts through an extrapituitary, non-paraventricular pathway to activate the HPA axis.

CRF-dependent and CRF-independent mechanisms involved in hypophysial-adrenal system activation by bacterial endotoxin.

The immune system and the hypothalamic-pituitary-adrenal (HPA) axis play important role in the overall inflammatory response. The mechanism through which lipopolysaccharide (LPS, endotoxin) stimulates the HPA axis is not well understood. In order to clarify the role of hypophysiotropic peptides of paraventricular origin in the effect of LPS on ACTH and corticosterone secretion, the effect of LPS was studied on rats with lesions of hypothalamic paraventricular nucleus (PVN). It was shown that 90 min after 2 mg/kg LPS i.p. the ACTH, but not the corticosterone response was effectively blunted in PVN-lesioned rats, as compared to sham operated animals. However, in PVN-lesioned rats 240 min after treatment with LPS a significantly higher plasma ACTH and corticosterone level was monitored. It is, therefore, suggested that in response to LPS activation of HPA both CRF(s)-dependent and CRF(s)-independent mechanisms are involved, even a direct effect of the adrenal cortex should be taken into account.

Effect of bacterial endotoxin on placentation of rats.

The effect of bacterial endotoxin on placentation in rats was studied on 160 CFY pregnant rats. Based on this experiment, it was concluded that (i) the endotoxin (1 mg/animal i.p.) inhibited placentation (in 90% of animal). (ii) The endotoxin-induced fetopathy almost exclusively resulted in abortion. (iii) The fetuses reacted to endotoxin with relatively the same degrees of susceptibility. (iv) The growth of surviving fetuses seemed to be undisturbed. (v) Endotoxin-induced damages in mothers first of all depend on the individual susceptibility of these pregnant animals and (vi) the endotoxin tolerance induced by radio-detoxified endotoxin (TOLERIN) significantly protects both the mothers and the fetuses against endotoxin challenge.

Influence of experimentally induced endotoxemias on the thyroid function of rats.

The general membrane-damaging effect of endotoxins (LPS) may be also demonstrated on the follicular cells of thyroid gland. Serum T4 level significantly decreased and the response of thyroid gland to exogenous THS was markedly inhibited in experimental endotoxin and other so-called enteroendotoxemic shocks (e.g. intestinal ischemia, tourniquet shock, intestinal syndrome of radiation disease). A single subtoxic dose of LPS given to newborn rats decreased the T4 level, the response of thyroid to TSH in adulthood and caused a somatic retardation. The radio-detoxified endotoxin (TOLERIN) did not inhibit the thyroid response to TSH. TOLERIN pretreatment protected the rats against LPS and other enteroendotoxemic shocks.

Study of the endotoxin sensitivity of pregnant rats and their fetuses.

The endotoxin sensitivity of pregnant rats and their fetuses was studied and the following conclusions were drawn: 1. In the third trimester the fetus-damaging effect of endotoxin also involves considerable damage to the mother which often ends with the mother's death (in this experiment in 40% of the cases). 2. The endotoxin sensitivity of the fetuses of one mother is relatively the same. 3. Within the race-specific range the endotoxin sensitivity of a given population of pregnant rats largely varies according to the individual sensitivity. 4. There is no "threshold dose" which would kill each fetus and would not kill any of the mothers. Based also on literary data, it is concluded that the effect of endotoxin causing fetal death is mediated by humoral factors released from the mother's organism, and fetal death is primarily due to anoxia.

Potential role of SH groups in the radiosensitivity of adenylate cyclase.

The role of oxidation of SH groups in the activity of adenylate cyclase and in radiosensitivity of the enzyme was investigated. Adenylate cyclase activity was measured in purified membrane preparation of 19 day old chicken embryo brains. N-ethyl-maleimide (NEM) and lead-acetate were used as SH inhibitors. Gamma irradiation was carried out with 60-Co source. NEM inhibition of adenylate cyclase was dose dependent and 50 per cent inhibition was observed at 40-50 microM NEM. Activity of adenylate cyclase was elevated at lower concentrations of lead-acetate (10 nM-100 microM) and was inhibited at higher concentrations (above 100 microM). The presence of 40 microM NEM did not alter the shape of lead acetate saturation curve of adenylate cyclase. Gamma irradiation in the dose range of 100-800 Gy elevated the adenylate cyclase activity measured in the presence of 5 mM NaF but did not alter the basal activity. Gamma irradiation did not have significant effect on NEM saturation of adenylate cyclase, while it altered slightly the lead acetate saturation curve.

A possible mechanism of the inhibitory effect of endotoxin on adenylate cyclase.

The possible mechanism of the inhibitory effect of endotoxin on adenylate cyclase was investigated. Plasma membranes were isolated from porcine thyroid gland and rat liver. The effects of endotoxin on adenylate cyclase activity was determined. The adenylate cyclase activity followed in the presence of various concentrations of guanosine-imidodiphosphate (5 x 10(-8)-10(-6) M) and NaF (0.1-10 mM) was markedly inhibited by endotoxin. Moreover, the basal activity of adenylate cyclase was inhibited similarly. The inhibition was concentration dependent. At 400 micrograms/ml of endotoxin a 60% inhibition of adenylate cyclase both without and in the presence of activators was detected. The inhibitory effects of endotoxin and its radiodetoxified derivative were similar. Neither of the two investigated endotoxin preparations tested had any effect on the 3H-guanosine-imidodiphosphate binding activity of adenylate cyclase (neither on the binding capacity, nor on the Kd value). Forskolin activated adenylate cyclase was also inhibited markedly by both endotoxin and radiodetoxified endotoxin. These results suggest that beyond the other membrane effects there is a probability of direct inhibitory effect of endotoxin on the catalytic subunit of adenylate cyclase.

Alterations in the chemical composition and antigenicity of Escherichia coli O89 lipopolysaccharide and lipid A after 60Co gamma irradiation.

The chemical composition and the antigenicity of Escherichia coli lipopolysaccharide (LPS) and lipid A were investigated after irradiation with 150 kGy 60Co-gamma ray. Compared to the original preparations, the irradiated LPS showed a significant reduction in glucosamine, glucose and galactose and a decrease in the phosphate content. The fatty acid components were reduced to a smaller degree. Irradiation induced a reduction in the antigenicity of the polysaccharide part. The amount of glucosamine and phosphate decreased in the irradiated lipid A. The fatty acid content was significantly reduced. The alteration in the chemical composition was not paralleled by changes in the antigenicity of irradiated lipid A.

Age dependent effect of toxic and detoxified endotoxins on the natural anti-DNA antibody level in rats.

Toxic endotoxin (LPS) and its irradiated detoxified form (RD-LPS) induced only small changes in the DNA level in the sera of 4, 12 and 18 months old rats. Changes in the anti-DNA titres were age-dependent. In 4 months old rats one or two injections of the LPS preparations failed to increase the circulating anti-DNA level. In 12 and 18 months old rats both toxic and detoxified LPS preparations raised the anti-DNA titres. The second LPS injection evoked a smaller response in 18 months old rats than the first one.
