Effects of 5 alpha-reductase inhibitors on intraprostatic androgens in the rat.

FCE 27837 is a novel inhibitor of 5 alpha-reductase, the enzyme responsible for the conversion of testosterone (T) to 5 alpha-dihydrotestosterone (DHT). The compound caused inhibition of human and rat prostatic enzymes, with IC50 values of 51 and 60 nM, respectively. The in vivo effect of FCE 27837 on 5 alpha-reductase was evaluated in adult male rats, treated orally at 10 mg/kg/day for 10 days. The compound caused 33 and 42% reductions in ventral prostate and seminal vesicle weights, respectively. The prostatic content of DHT, measured 6 h after the 10th dose of FCE 27837, was reduced by 75%, whereas T content increased by 442%. Similar effects were observed with 10 mg/kg/day of finasteride, whereas epristeride, tested at the same oral dose, was found to be the least effective compound, decreasing prostate weight by 22% and DHT content by 46%. Castration caused > 90% reductions in prostatic weight and prostatic DHT.

[Determination of plasma concentration of amiodarone and desethylamiodarone with high pressure liquid chromatography. Preliminary experience].

A simple specific and sensitive high pressure liquid chromatography method (HPLC) to measure amiodarone and desethylamiodarone is described. The plasma sample is deproteinized with acetonitrile and a 50 ul aliquote run in the HPLC system with reverse phase column (C-18). Methanol-ammonium hydroxide is used to separate and spectrophotometric measurements are read at 254 nm.95% of the drug present in plasma is extracted with this method, with a sensitivity of 0.03 ug/ml and a day to day variation under 3%. Preliminary measurements were analyzed in 14 subjects receiving amiodarone for treatment of supraventricular arrhythmia. With a mean dose of 267 +/- 116 (SD) mg/day, plasma levels of 1.3 +/- 0.6 ug.ml for amiodarone and 0.9 +/- 0.5 mg/ml for desethylamiodarone were obtained.

Previous experience facilitates preservation of spatial memory in the senescent rat.

In an attempt to evaluate whether previous training antagonizes age-related memory deficits, aged rats with previous training experience were compared with aged and young naive rats in the Morris water maze test. The findings of our study indicate that experience seems to facilitate the preservation of the spatial reference memory for 12 months in the aged rat, whereas senescent naive animals showed the usual age-related memory deficits. However, experience seems task-specific since the same aged rats fail to acquire a new different task.

[A form for soliciting the measurement of drug plasma level and its use in a university hospital].

The correct clinical use of drug plasma levels is frequently jeopardized by neglecting to consider pharmacokinetic parameters at the time of sample drawing. A form to be filled at this time revealed that inadequate information concerning the drug was present in 28% and sampling time in 50% of cases. Accordingly, a new form is proposed to help avoid this information inadequacies and thus lead to better clinical use of drug plasma levels.

[Carbamazepine: the bioavailability of 4 oral pharmaceutical products].

The bioavailability of carbamazepine from 4 commercially available products was evaluated in 12 healthy volunteers. A crossover design was used and each patients received 400 mg of each product. Plasma levels of the drug were determined periodically for 72 h using a gas-liquid chromatographic method. An open model of one compartment for first-order absorption was assumed to derive pharmacokinetic parameters. Dissolution kinetics was also evaluated in each product. Significant differences in bioavailability were shown for one product. Results correlated with the in vitro dissolution findings.

[The bioavailability of 4 sustained-release theophylline preparations from the Chilean market].

We evaluated pharmacokinetic parameters and bioavailability of 4 sustained release theophylline preparations. A crossover design was used in 12 healthy males aged 22 to 27 years old. Each individual received 250 mg iv followed by 250 mg orally of each preparation, and then 400 mg rapid acting aminophylline. A 7 day period was allowed between drug courses. HPLC was used to determine plasma levels of theophylline at regular intervals up to 48 hr following drug administration. Significant (p < 0.05) differences in pharmacokinetic parameters were found among preparations, 2 of them having larger integrals of plasma levels and one of them different times to peak plasma level and peak plasma concentration, compared to both remaining preparations.

Age-related ability of rats kept on a life-long hypocaloric diet in a spatial memory test. Longitudinal observations.

Cognitive behavior was assessed in a test specific for spatial memory, according to a longitudinal experimental model, in populations of 12-, 24- and 30-month-old rats which were fed ad lib either a standard (ST) or a hypocaloric (HY) diet, that had been shown to reduce age-related pathologies and to increase survival. Already at 12 months of age, some cognitive deficits were observed in ST but not in HY rats. When retested 12 and 18 months later, the animals performed better at the beginning of the test than when tested for the first time, indicating that some aspects of previous experience lead to a preservation of spatial memory. Deficits which had been previously observed in 2-year-old groups in a cross-sectional experimental model were not evident this time. Some of the differences between the two diet groups observed 12 months before disappeared. When testing was repeated for the third time at 30 months of age, ST fed rats presented very marked deficits in learning and in memory, which were not seen in the HY group. It thus appears that a dietary regimen in which part of the calorie-rich components, such as lipids and carbohydrates, are replaced with vegetable fibers, retards some of the age-related deteriorations of brain functions.
