[Morphometric analysis of the spleen].

Present possibilities of morphometric analysis of the spleen have been studied. We have compared possibilities provided by stereological calculation and by computer image analysators a) Telemet II (Tesla, Piestany) and b) CUE-2 GALAI (Israel). In both latter cases equidensitometric evaluation and image processing methods were used. Stereological calculations are not technically demanding but time consuming. Computer image processing on the base of equidensitometric measurements is more effective that stereological calculations. The difference between used equipments is based on access to appropriate microscopic and computer technique and software equipment.

[Dermoid cyst of the spleen].

The case report of a 14-year old girl is given. In her spleen of the weight of 1580 g an irregular cyst of glossy inner surface and with striking trabeculation developed. It was filled with clear greenish liquid. The wall was formed by hyalinised collagen tissue which was covered by epidermoid and cuboidal epithelium on the inner surface. Vessel conglomerates were frequent, some bleeding or mononuclear infiltrates, foci of giant cell granulation tissue, lymphoid or fatty tissue were present.

Immunohistochemical markers in uterine tumour diagnostics.

The recent fast growth of data on new markers and their identification in different cellular structures results in the need of narrow selection with regard to their applicability. The aim of the review is description of application possibilities of immunohistochemical methods in the diagnostics of pathological processes in the cervix and body of uterus. (Fig. 5, Ref. 43.)

[Histochemistry of a new Tritrichomonas mobilensis lectin with sialic acid specificity].

BACKGROUND: Histochemistry with lectins highly specific for one sugar represent a sensitive and accurate method for the localization of glycoproteins in tissues. OBJECTIVES: The paper evaluates the possibilities of histochemical applications of the novel lectin from Tritrichomonas mobilensis (TML) with specificity for sialic acid. METHODS: TML purified by affinity chromatography was used for anti-TML monoclonal antibody production and tested with the use of avidin-biotin peroxidase technique. RESULTS: In human biopsy material from different organs and tissues, TML showed strong membrane reactivity with vascular endothelium with the exception of glomerular and pulmonary capillaries and sinuses in the spleen and the bone marrow. Luminal membrane positivity dominated in glandular epithelia, especially in the lining of ducts. In squamous epithelia, the membrane positivity was present in cells of the basal layer, in transitional epithelium also in umbrella cells. Mucus in respiratory tract was positive, in gastrointestinal tract the positivity was irregular. Nerve tissue and endocrine glands were mostly negative. CONCLUSIONS: The lectin from tritrichomonas mobilensis can be considered a perspective reagent for identification of sialylated glycosubstances based on its high specificity for one sugar--the sialic acid and excellent results in histochemistry. (Fig. 7, Ref. 29.)

[Bilateral primary leiomyoma of the ovary].

Primary leiomyoma of the ovary is rare. Most of these tumors are only a few millimeters in diameter and unilateral, generally occurring in women aged 45 to 65 years. We present the exceptional case of a 16 year-old girl with bilateral ovarian leiomyomas, proved by immunohistochemistry, and discuss the feature of the case in the light of the known literature.

[The significance of prostate markers in the orthology of the female prostate].

In addition to knowledge gained in the first half of the 8th decade, the evidence of cross-antigenicity between male prostate and Skene's glands by means of PSA and PSAcP demonstrations in Skene's glands and ducts justifies utilization of the term prostate in both sexes. The authors compared the results of immunohistochemical examination of prostatic markers by means of the PAP method which was used at the beginning of the 8th decade, with that of BSAP technique. Prostatic tissues of 11 females and children at the age ranging from 5 to 71 years were examined. The results gained by means of the BSAP method were identical with those gained by means of the PAP method. Prostatic markers PSA and PSAcP were expressed on the surface and in apical cytoplasm of cells lining the prostatic ducts, and in prostatic glands. The authors proved the expression also in membranes of the stratified cylindrical epithelial cells of the ducts, and in female prostatic fluid in ducts and glands, especially PSAcP. Even though both immunohistochemical methods brought identical results, the authors recommend to prefer the BSAP method to the PAP method due to optically more contrast expression. (Fig. 8, Ref. 36.)

[Trophoblastic disease. I. Use of immunohistochemistry in the diagnosis of complete hydatidiform moles].

Immunohistochemical study of 15 complete moles detected expression of human chorion-gonadotropin (hCG), human placental lactogen (hPL) and placental alkaline phosphatase (PLAP). Syncytiotrophoblast showed the most conspicuous expression of hCG, hPL positivity was mild, PLAP positivity poor. Intermediate trophoblast had stronger hPL positivity There was not any positivity in cytotrophoblastic cells.

[Trophoblastic disease. II. Immunohistochemical and cytogenetic parameters of partial hydatidiform moles].

A group of 30 cases of partial hydatid mole were studied by cytogenetics and immunohistochemistry. Trophoblastic cells were always triploid. Syncytiotrophoblastic cells and intermediate trophoblast showed the strongest expression of PLAP, hPL positivity was mild. Expression of hCG was only focal. There was not found any detectable immunopositivity in trophoblastic cells.

[Comparison of susceptibility to Legionnaires' disease in small laboratory animals].

The authors tested under laboratory conditions the susceptibility of five species of small laboratory animals (guinea pigs, rats, syrian and chinese hamsters and mice) to legionella infection after intraperitoneal inoculation with 10(8) cells of nine strains of legionellae of the Legionella pneumophila species. The suitability of the animal model was evaluated on the basis of clinical manifestations of the infection, pathological macroscopic changes and detection of legionellae from animal organs on CYE (Charcoal-Yeast Extract) media. The results of the analysis of experimental animal infection revealed that guinea pigs are the most suitable experimental animals for investigations and the diagnosis of legionellosis.

[Monoclonal antibodies in the biopsy diagnosis of tumors].

Immunohistochemistry has made rapid advances over the last decades. The 80s have been the era of monoclonal and polyclonal antibodies which have made identification of different antigens more exact and more simple. Demonstration of individual markers of tumor cells has greatly contributed to the determination of histogenesis and to classification of tumors. The initial euphoria, however, has been on the wane as facts have accumulated demonstrating that the expression of many markers is disappearing particularly in less differentiated tumors or that new markers appear which were considered to be specific for a different type of tissue. Some of the problems involved are being dealt with in the short overview presented.

[Immunohistochemical analysis of the liver using monoclonal antibodies against intermediate filaments].

The expression of some types of cytokeratin and vimentin in the liver was studied by using monoclonal antibodies to these types of intermediary filaments. Necrotic samples from livers without pathological finding were examined. A panel of 5 monoclonal antibodies, prepared in Czechoslovakia, to cytokeratins Nos. 7,8,18, and 19, as well as to vimentin was used. The findings confirmed the characteristic expression of cytokeratins 8 and 18 in hepatocytes and of cytokeratins 7, 8, 18, and 19 in epithelial cells of intrahepatic bile canaliculi, that monoclonal antibodies prepared in Czechoslovakia (LAMO, Brno and VUKEO, Brno) can be successfully used in immunochemical studies of intermediary filaments in the liver.

[Cardiac complication of mucoviscidosis].

Over the years 1971 to 1988 autopsies of 33 children with the clinical diagnosis of mucoviscidosis were performed at the Institute of Pathology, Medical Faculty, Comenius University, Bratislava. In three cases pronounced changes were observed on the myocardium, predominantly on the left ventricle. Multifocal necrosis and fibrosis of the myocardium represent rare but severe changes occurring as a complication of mucoviscidosis in infancy.

[Development of adrenergic innervation of the heart in man].

The presence of postganglionic adrenergic nerve fibres in the myocardium of human fetuses ranging in gestational age from 9 weeks to 8 months was studied by the method of glyoxilic acid induced fluorescence (GIF). Heart samples were obtained after legal induced interruption of pregnancy, indicated in part of the cases for genetic defects. Identification of adrenergic nerve fibers in the human heart proved possible only in the second half of gestation. This finding supports the assumption of humoral rather than nervous regulation of heart activity in the first half of intrauterine development of the fetus.

[Myocardial damage as a rare complication of mucoviscidosis].

A postmortem examination was performed on a male child who had lived to twelve months of age, at the Institute of Pathology of the Medical Faculty of Comenius University in Bratislava, Czechoslovakia. The clinical diagnosis had been mucoviscidosis, bronchopneumonia, and dystrophy. Findings characteristic of cystic pancreas fibrosis and bronchopneumonia of the right lower lung lobe were macroscopically recorded and were microscopically established. Massive fibrotic and necrotic lesions were primarily recorded from the left cardiac ventricle and were considered to be unusual findings. Damage to the myocardium is a very rare complication of mucoviscidosis. Less than 50 cases have so far been described, according to Benesová et al. (1983).
