Structure of N-tert-butoxycarbonyl-L-phenylalanine benzyl ester.

C21H25NO4, Mr = 355.4, monoclinic, P2(1), a = 5.206 (2), b = 17.294 (4), c = 10.972 (2) A, beta = 98.91 (1) degrees, V = 976 (1) A3, Z = 2, Dx = 1.21 Mg m-3, lambda(Cu K alpha) = 1.5418 A, mu = 0.68 mm-1, F(000) = 380, T = 293 K, final R = 0.071 for 1186 observed reflections. The structure is stabilized in the a direction by means of intermolecular hydrogen bonds [N(1) ... O(2i) = 3.01 (1) A, (i) = x + 1, y, z]. The urethane amide bond adopts the trans conformation [H(1)-N(1)-C(5) = O(2) = 169 (3) degrees]. The butoxycarbonyl (Boc) moiety is directed away from both the phenylalanine aromatic ring and the benzyl ester ring, in contrast to the arrangement observed in Bocphenylalanine phenacyl ester [Vlassi, Germain, Matsoukas, Psachoulia, Voliotis & Leban (1987). Acta Cryst. C43, 2173-2175]. The orientation assumed by the Boc group may be the result of steric restrictions imposed by both rings.

Cloning, expression and sequence analysis of an endolysin-encoding gene of Lactobacillus bulgaricus bacteriophage mv1.

The lysA gene specifying an endolysin of Lactobacillus delbrueckii subsp. bulgaricus bacteriophage mv1, was cloned and expressed in Escherichia coli. The 4.05-kb restriction fragment containing this gene was analysed by restriction and deletion mapping, and by subcloning. The nucleotide sequence of a 1150-bp fragment coding for an active lysin was determined. The lysA gene consists of 585 bp and codes for a protein of a deduced Mr of 21,120, which agrees with the size based on in vivo transcription/translation studies. The deduced amino acid sequence of the mv1 lysin (LysA) was compared to that of other known lytic enzymes. Significant homology was observed with the N-terminal portion of the muramidase of the fungus Chalaropsis and that of the muramidase of the Streptococcus pneumoniae phage Cp-1, suggesting that LysA might be a muramidase. In E. coli, the cloned lysA gene was able to complement the muramidase-defective bacteriophage lambda Ram5, proving that the products of these two genes are interchangeable. The lysA gene is preceded by an open reading frame with unknown function and no characteristic prokaryotic promoter sequences could be detected upstream from lysA, suggesting that this gene is part of an operon.

Crystal and molecular structure of BN 52021, a PAF-acether antagonist. Comparison with the conformation of Kadsurenone and related compounds.

BN52021 (Ginkgolide B) monohydrate C20H24O10 X H2O crystallizes in the triclinic space group P1 with two independant molecules in the unit cell of which parameters are a = 7.627(2), b = 11.514(3), c = 12.941(3)A, alpha = 97.05(2), beta = 90.27(2) and gamma = 108.71(2)o, V = 1067.06A3. Calculated density Dx = 1.320g X cm-3. The crystal structure was solved by Patterson methods starting from dreiding model data. The final reliability index R = 0.099 for 3848 observed reflections collected with a four-circle diffractometer. Anti-PAF activities of Kadsurenone, Kadsurin-A, Kadsurin-B and Piperenone, molecular structure of MIRANDIN-A, related compounds and BN52021 suggest that a distance 0-0 of about 6.6A observed in Kadsurenone and in BN52021 could be favourable to anti-PAF activity.

[Molecular modelling of usambarensine, tubulosine and emetine, cytotoxic amebicide alkaloids].

The analyses of the different possible conformations of usambarensine, tubulosine and emetine, which possess interesting cytotoxic and amoebicide properties showed us that they cannot take the planar conformation proposed to explain the activity of this type of molecules on protein synthesis. Nevertheless, a common (non planar) conformation exists where the angles between the two plans containing the aromatic rings are nearly the same for the three compounds. This fact could confirm the hypothesis of a common receptor which would be slightly different in amoebae because of its higher sensitivity to usambarensine.

PAF-receptor. 1. 'Cache-oreilles' effect of selected high-potency platelet-activating factor (PAF) antagonists.

Three-dimensional electrostatic maps were calculated for six potent antagonists of platelet-activating factor (PAF), the antagonists being selected for their apparent structural heterogeneity. The molecules examined were the compact Ginkgolides BN 52020, BN 52021 and BN 52022 (1, 2 and 3), the semi-rigid kadsurenone (4), a flexible synthetic dinor type C furanoid lignan L-652,731 (5a) and the triazolothienobenzodiazepine WEB 2086 (7). Calculation of the electrostatic potential generated around all the above molecules showed the existence of two wells of negative potential or 'cache-oreilles' (ear-muffs), i.e., the isocontours drawn at -10 kcal/mol, located at 180 degrees from each other and separated by a maximum distance of 22-27 A. Except for the synthetic dinor type C furanoid lignan (5a), the molecules also presented a moderate hydrophobic fragment, which constitutes a third point of interaction with the high-affinity binding site in rabbit and human platelets. The findings of the present study allow speculation that this high-affinity acceptor site may be a 'polarized cylinder' with a diameter of 10-12 A.
