Micelles of poly(oxyethylene)-poly(oxypropylene) block copolymer (pluronic) as a tool for low-molecular compound delivery into a cell: phosphorylation of intracellular proteins with micelle incorporated [gamma-32P]ATP.

Pluronic P85 (poly(oxyethylene)-poly(oxypropylene) block copolymer) was used for in vitro delivery of [gamma-32P]ATP into intact Jurkat cells. Negatively charged ATP molecules are not able to penetrate cell plasma membrane. Hence, exogenous [gamma-32P]ATP added to a cell culture does not participate in phosphorylation of intracellular proteins. The addition to cells of [gamma-32P]ATP solubilized in positively charged (containing dodecylamine) pluronic micelles results in considerable increase of protein phosphorylation. In this case the treatment of intact cells with alkaline phosphatase (resulting in dephosphorylation of external proteins) causes no essential decrease of [32P]-incorporation in cell proteins. That gives an evidence of delivery of solubilized ATP into a cell. Under the experimental conditions used, pluronic micelles neither influence the viability of cells nor permeabilize cell plasma membrane.

Pluronic micelles as a tool for low-molecular compound vector delivery into a cell: effect of Staphylococcus aureus enterotoxin B on cell loading with micelle incorporated fluorescent dye.

Micelles of pluronic P85 (poly(oxyethylene)-poly(oxypropylene) block copolymer) are used as microcontainers for in vitro delivery of fluorescein into Jurkat and MDCK cells. In order to target the fluorescein containing micelles into the cell, Staphylococcus aureus enterotoxin B (SEB) is covalently conjugated with a pluronic molecule and the conjugate is incorporated into the micelle content. The incorporation of SEB capable of receptor-mediated endocytosis results in a drastic enhancement of the efficiency of cell loading with the fluorescent dye. This effect is not observed under the conditions (4 degrees C) when endocytosis is abolished.

Nonrandom chromosomal changes in retinoblastomas.

The analysis of the karyotype in 76 retinoblastomas (24 our cases and 52 described in the literature) has revealed nonrandom changes of Iq, 6p, 13, 16 and the sex chromosomes. Complete or partial trisomy Iq was observed in 44 out of 76 tumours. Tetra-or trisomy 6p was found in 35 and 6 cases respectively. Chromosome 13 monosomy or its long arm deletion was described in 11 tumours. Monosomy 16 and loss of the X or Y--in 18 and 12 cases. The specific feature of retinoblastoma karyotype is presence (along with two normal homologues of the pair 6) of the marker chromosome i (6p). Possible causes of unexpectedly rare abnormalities of chromosome 13 in retinoblastoma cells were discussed in the light of well known data on predisposing role of constitutional deletion 13q14, and recent molecular genetic studies showing the significance of recessive tumour genes in carcinogenesis. The cytological signs of gene amplification (HSRs, DMs) were revealed in few retinoblastomas. However, the recent data on N-myc gene amplification and its elevated expression in several retinoblastomas indicate that amplification of the oncogene(s) might be involved in the genesis of this tumour. Further studies are needed to understand the correlative role of specific chromosome rearrangements, gene(s) amplification and action of recessive rb gene, located at 13q14 in initiation and progression of retinoblastoma.

[Nonrandom karyotype changes in human retinoblastomas].

Cytogenetic analysis of 15 retinoblastomas developed in children having no constitutional chromosome 13 deletion has been carried out. In tumor cells, no deletion or loss of chromosome 13 was revealed. The specific marker chromosome i(6p) described in our previous publications has been found in 9 tumors. Besides, in two cases, trisomy of short arm of chromosome 6 was present. Other non-random changes (trisomy 1q, monosomy 16 and loss of one of the sex chromosomes) were not specific for retinoblastomas, because they were described in literature for some other tumors as well. The possible significance for genesis of retinoblastomas of dose multiplication of the genes located in the 6p is discussed.

[Chromosome disorders in retinoblastoma cells].

The karyotype of 7 unilateral retinoblastomas was studied. Loss or a gain of chromosomes and structural chromosomal changes were observed in all tumors. Modal chromosome number was pseudodiploid in four and near-diploid in three tumors. There were no sex chromosomes (X or Y) in 3 tumors. Four tumors were characterized by the presence of an identical marker chromosome--i(6p). Two other markers--lp+ and 17q+ (probably, not quite identical)--were present in 4 and 3 tumors, respectively. The cells of two retinoblastomas contained both i (6p) and 17q+ markers.

[Cytogenetic characteristics of transplantable mouse tumors].

The cytogenetic control of 17 mouse tumor cell strains from of the collection of the bank of the Centre for Oncology Research of the AMS USSR was made: 4 leukemias (L-5178Y, L-1210, L-1210 resistant to 6-mercaptopurine, P-388), 2 sarcomas (S-180, S-298), 8 carcinomas (Ehrlich ascitic carcinoma, carcinoma 755,6-mercaptopurine resistant carcinoma 755, lung cancer LC-67, cervical cancer CC-2, cervical cancer CC-5, stomach cancer GC-5), 2 melanomas (B16, S91) and 1 plasmocytoma (MOPC 21). A comparison of their cytogenetic features allowed a conclusion to be drawn on the absence of any contamination among 14 strains of this collection. Carcinoma 755, sarcoma 298 and leukemia L-5178Y need some further examination for such inference to be valid.

[Radiation protection using a gaseous hypoxic mixture in oncological practice].

Studies on volunteers have shown that the gas hypoxic mixture containing 10% of oxygen and 90% of nitrogen (GHM-10) renders a protective action on the genetic apparatus of human skin cells but provides no protection of the peripheral blood leucocytes, which show the identical character of metabolic processes as neoplastic cells. Under clinically performed distant x-ray therapy for breast cancer the inhaling of GHM-10 was found to render the antiradiation protective action on different normal tissues (skin, subcellular connective tissue, muscle tissue, mammary gland tissue), but it fails to protect the tumor tissue and regional lymph nodes involved. The clinical observations were supported by pathomorphological examination of the operation material.

Body temperature and tumor virus infection. I. Tumorogenicity of Rous sarcoma virus for reptiles.

Rous sarcoma virus (RSV) was oncogenic for the following nine species of reptiles representing 6 families from Chelonia and Squamata orders: family of Testudinidae: 1. Testudo horsfieldi, family Agamidae: 2. Agama sanguinolenta; 3. Agama erythrogastra, family Lacertidae: 4. Eremias persica; 5. Eremias velox; 6. Eremias grammica, family Scincidae: 7. Eumeces taeniolatus, family Boidea: 8. Erix tataricus, 9. Ancistrodom blomhoffi. RSV did not induce tumors in 13 studied species of reptiles. Histologically 26 reptile tumors studied were polymorphous sarcomas with spindle-shaped (fibroblast-like), round and polygonal macrophage-like cells and sometimes peculiar giant polynuclear cells. Chromosomal analysis showed that reptile tumors arose out of reptile cells. RSV was pathogenic for adult reptiles. Reptile tumors did not contain a mature infectious virus. The tumors of 2 snakes were virogenic. The effect of increased temperature at the body level on the transformation of a symptomless viral infection into a viral disease is discussed in the evolutionary aspect.

[Effect of methylcobalamin and fluoralkylcobalamins on E. coli 113/3 cell growth and on a primary human embryonic fibroblast culture].

Comparative analysis of the functional activity of several fluoralkylcobalamines was carried out using E. coli 113/3 strain deficient in vitamin B12 and methionine. Difluoro chlor methylcobalamine (CF2Cl-Cbl) exhibited the most distinct inhibitory effect on growth of bacterial cells in the medium with cobalamine. Effect of methylcobalamine and CF2Cl-Cbl on the proliferative activity of human embryonal fibroblasts was studied in media of various composition. The proliferative activity of fibroblasts was distinctly increased in the medium with methylcobalamine at various periods of cultivation; the fraction of 3H-thimidine labelled cells and the mitotic index were increased. The distinct decrease in amount of cells, synthesizing DNA, and in their mitotic activity was observed in medium with CF2Cl-Cbl. The data obtained suggest that difluorochlor methylcobalamine affects the cell proliferation as the antagonist of methylcobalamine in experiments with bacterial cells and the primary culture of human embryonal fibroblasts.
