The detection of chemical impurities by high pressure liquid chromatography and the genetic activity of medical grades of pyrvinium pamoate in Saccharomyces cerevisiae and Salmonella typhimurium.

The genetic activity of several medical grades of the anthelmintic drug pyrvinium pamoate, which is a dipyrvinium salt, was studied in a diploid mitotic recombination and gene conversion assay (strain D5 of Saccharomyces cerevisiae), and in several haploid yeast reversion assays (strains XV185-14C, XY718-1A, and 7854-1A). All of the samples were recombinogenic in strain D5 and mutagenic in the haploid strains, however, the degree of genetic activity varied considerably among the medical grades of pyrvinium pamoate that were tested. Similarly, these samples varied in degree of mutagenicity when they were tested in strains TA97, TA98, TA100, and TA102 of Salmonella typhimurium, but some of the medical grades of pyrvinium pamoate were mutagenic both in the presence and in the absence of the metabolic transformation system, whereas other medical grades of the drug required such activation to be mutagenic. In addition, the medical grades and dosage forms of several brands of pyrvinium pamoate were examined for purity by fluorescence high pressure liquid chromatography (HPLC) using a methanol:water (90:10) solvent system. The HPLC data indicate that monopyrvinium salts are the major contaminants in these pharmaceuticals. In general, there is a correlation between the degree of genetic activity and toxicity, and the number and relative quantity of impurities found in each sample.

Recent applications of electrochemical techniques to the analysis of pharmaceuticals.

The past decade has revealed a remarkable and renewed interest in the application of electrochemical techniques to the analysis of drugs and pharmaceuticals. In addition, several of these techniques are now being employed to elucidate electrode processes by studying the mechanisms involved in organic redox reactions. The renewed interest in electrochemical techniques can be attributed in part to more sophisticated instrumentation and to an increased understanding of the techniques themselves. This paper selectively reviews recent applications of the following electrochemical modes in the analysis of drugs and pharmaceuticals: amperometry, conductometry, coulometry, ion-selective electrodes, potentiometry, stripping voltammetry (anodic and cathodic) and voltammetry.

Semiaqueous potentiometric determinations of apparent pKa1 values for benzothiadiazines and detection of decomposition during solubility variation with pH studies.

Apparent acidity constants obtained for various benzothiadiazine diuretics by semiaqueous potentiometric titrations gave satisfactory agreement with many values obtained by aqueous potentiometry. One suitable method for determining the acidity constants of sparingly soluble drugs, the solubility variation with pH technique, does not take into account instability problems. The preparation of decomposition products and the TLC analysis of sample solutions at various time intervals during solubility studies of methyclothiazide and bendroflumethiazide indicated that decomposition takes place during agitation and equilibration. This decomposition in buffers of pH 8 and higher was confirmed with the acidified p-dimethylaminobenzaldehyde test for primary aromatic amines.

Synthesis of tetracycline ring A analogs.

Studies directed at the synthesis of tetracycline ring A analogs are described. 4-Carbethoxycyclohexane-1,3-dione was converted to the ethyl urethan dispiro[1,3-dioxolane-2,2'-cyclohexane-4',2"(1,3)-dioxolane]-1'-carbamic acid ethyl ester via the dispiro[1,3-dioxolane-2,2'-cyclohexane-4'-2"(1,3)-dioxolane]-1'-carboxylic acid hydrazide. An improved synthesis of another cyclohexenone from methyl vinyl ketone and ethyl nitroacetate is reported. Reaction of N-(3-hydroxy-1-oxo-2-cyclohexen-4-yl)benzamide with alpha-chloroacetyl isocyanate afforded a 4(5H)-oxazolone derivative, as did the identical reaction on 5,5-dimethyl-1,3-cyclohexanedione (dimedone). This reaction provides a novel approach to these oxazolones with potential therapeutic importance. Other ring A analogs were synthesized also.

ac Polarography for tetracycline analysis.

The electrode processes for the reduction of several tetracyclines by ac polarography were examined. In pH 4.0 Walpole acetic acid-acetate buffer, two main waves occurred; the first was quasireversible and the second was reversible. Results showed that the first wave can readily be used for quantitative work. The second wave would also be suitable provided that there was no interference from other electroreducible substances.

Dc Polarographic assay of tetracyclines.

The inhibitory effect of dioctyl sodium sulfosuccinate on hog pepsin activity was investigated over the pH 1.5-3.0 range. The inhibitory effect was studied using a natural substrate, hemoglobin, and a synthetic substrate, N-acetyl-L-diiodotyrosine. The mechanistic studies revealed that a substrate-inhibitor interaction was the major mechanism of inhibition with hemoglobin. However, some direct enzyme inhibition also was involved. With the synthetic substrate, the inhibition las due to a competition between the substrate and the inhibitor molecules for the enzyme. The possible therapeutic significance of the inhibitory effect of the medicinal surfactant is discussed.

Determination of polythiazide in pharmaceutical dosage forms by high-pressure liquid chromatography.

A method is presented for the quantitative analysis of polythiazide tablets by high-pressure liquid chromatography. The powdered tablets are extracted with methanol, containing quinoline as the internal standard, and assayed by comparison of peak heights after liquid chromatography. The chromatographic conditions employed may be adapted for the determination of many other thiazide and nonthiazide diuretics.
