A retrospective analysis of the infectious bovine rhinotracheitis (bovine herpes virus-1) surveillance program in Norway using Monte Carlo simulation models.

Serological surveillance for antibodies against bovine herpes virus type I (BHV-1) which causes infectious bovine rhinotracheitis and infectious pustular vulvovaginitis has been carried out since 1992 in Norway. Since 1993 (when a single infected herd was detected) all bulk-milk and pooled-serum samples have been negative for BHV-1 antibodies. This paper describes the use of Monte Carlo simulation models for the analysis and interpretation of the results of the surveillance and provides support for the contention that the Norwegian cattle population is not infected by BHV-1.

Economic aspects of disease monitoring with special reference to bovine paratuberculosis.

Monte Carlo simulation models were used to evaluate the feasibility and potential results of a proposed national survey of the prevalence of bovine paratuberculosis (PTB) in dairy herds in Norway. The expected herd prevalence was assumed to be 0.2% in the simulations. Infected herds were classified as detected if 1 animal was sero-positive. With a sample size of 6000 herds at least 1 truly infected herd was detected in 99% of the iterations. The low sensitivity of the ELISA test, the assumed low herd prevalence, the typical low within-herd prevalence of PTB and the small herd sizes in Norway all present problems in detection of the disease. The results showed that the ratio between false-positive herds and true positive herds detected had a median of 70:1. At the assumed herd prevalence of 0.2% and a cost/test of 70 NOK the median cost of detecting 1 infected herd was approximately 900,000 NOK. If 2 positive reactors were needed to classify a herd "infected" the median cost of detecting 1 infected herd was 5,055,000 NOK. Our results suggest that a randomized national prevalence survey would not be feasible, due to the low probability of detecting infected herds and because of the high number of false-positive reactions that would be expected.

A simulated surveillance program for bovine paratuberculosis in dairy herds in Norway.

Monte Carlo simulation models were used to evaluate the feasibility and potential results of a proposed national survey of the prevalence of bovine paratuberculosis (PTB) in dairy herds in Norway. The expected herd prevalence was assumed to be 0.2% in the simulations. The low sensitivity of the ELISA test, the assumed low herd prevalence, the typical low within-herd prevalence of PTB and the small herd sizes all present problems in detection of the disease. Simulations with 500, 1000, 2500 and 6000 herds tested were done. Our results suggest that a national survey would not be feasible at present, due to the low probability of detecting infected herds and because of the high number of false-positive reactions that would be expected to occur.

A Monte Carlo simulation model for assessing the risk of introduction of Gyrodactylus salaris to the Tana river, Norway.

The Tana river in northern Norway, the most productive salmon river in Europe, is free of Gyrodactylus salaris. Currently there is one salmon farm in operation on the Tana fjord. Because of the strong association between stocking of rivers with salmon and infestations with G. salaris there is national and international concern that the existing farm might lead to the introduction of the parasite to the Tana river. In response to these concerns a quantitative analysis of the risk of introduction of G. salaris to the Tana river was undertaken. A scenario tree, the Monte Carlo simulation model and results of the simulations including sensitivity analyses are presented and discussed. Results show that the probability of introduction of G. salaris to the Tana river via transfer of smolt to the existing salmon farm is extremely low primarily due to the low probability that the transferred smolt become infested. The total risk was very sensitive to changes in the salinity of the water at the sea site.

Associations between behavioural indicators of "stress" in weaner pigs and respiratory lesions at slaughter.

The aim of the study was to investigate associations on the individual-pig level between eight behavioural indicators of 'stress' (defined in a very general sense), in seven to eight-week old piglets four weeks after weaning and the occurrence of atrophic rhinitis (AR) and chronic pleuritis (CP) at slaughter. The study was designed as a cohort study, including 512 pigs from two commercial farms. With one exception Actinobacillus pleuropneumoniae serotype 2 (App2) infection was not associated with any of the behaviour patterns observed. Using random-effects logistic regression, weak statistical associations, positive as well as negative, were found between the performance of several behaviour patterns and the two pathological lesions. This suggests the existence of biological associations between oral behaviour patterns in young pigs and the health status later in life. However, the results were not consistent between specific oral behaviour patterns or between the two herds indicating that the associations found are either due to chance or reflect the existence of herd-specific confounders. Therefore, results based on studies within only one herd should not be generalized to other herds without care. Behavioural observations on seven- to eight-week-old piglets on the basis of the present investigation cannot be recommended for use as early predictors of AR or CP at slaughter. Further studies are needed in order to elucidate causal relations between behaviour and health.

An epidemiologic and economic study of respiratory diseases in two conventional Danish swine herds. I: Prevalence of respiratory lesions at slaughter and their effects on growth.

A total of 578 slaughter pigs from 2 Danish conventional farrow-to-finish operations (Herds A and B) were followed from an age of 14 days to slaughter. Pigs were weighted at 3 weeks intervals and at slaughter and an extended post mortem examination of the plucks was done. Comparison of growth rates in pigs with and without specific types of lesions by the t-test and those with multiple lesions with regression models demonstrated that Mycoplasma-like pneumonia, complicated pneumonia, anterio-ventral pleuritis, fissures and atrophic rhinitis significantly reduced mean daily gain and increased the time required to reach slaughter weight. The total impact of the lesions in Herd A was an estimated reduction in mean daily gain of 27 grams and a 2 day increase in the interval from 14 days of age until slaughter (MDG14). Decreases in MDG14 in Herd B were more substantial, 98 grams and 16.7 days. Reductions in mean daily gains during the interval from the fourth weighing until slaughter were 31 grams in Herd A and 137 grams in Herd B. Chronic dorso-caudal and parietal pleuritis, without other lesions present, had no significant adverse effects on growth rates in either herd. Interactions between lesions did not significantly alter the estimates. The R2 values obtained for the regression models showed that the presence, absence or extent of lesions at slaughter explained only 13-27% of the variations in growth rates in the 2 herds.

An epidemiologic and economic study of respiratory diseases in two conventional Danish swine herds. II: Associations between lesions present at slaughter and mean daily gains during specific intervals of the growth period.

A total of 578 slaughter pigs from 2 Danish conventional farrow-to-finish operations (Herds a and B) were followed from an age of 14 days to slaughter. Pigs were weighted at 3 weeks intervals and at slaughter and extended post mortem examination of the plucks was done. Regression models with second and third order interaction terms demonstrated that Mycoplasma-like pneumonia, complicated pneumonia, anterior-ventral pleuritis, pericarditis, fissures and atrophic rhinitis, separately and through interactions with other lesions, significantly reduced mean daily gains during specific intervals of the growth period in 2 conventional swine herds. It is likely that the periods of reduced growth reflect the times when the diseases were in the acute and early recovery stages. Maximum estimated reductions in daily gains associated with the combined lesions were 82 grams and 283 grams during the interval 120-140 days in Herds A and B, respectively. Although dorso-caudal pleuritis and parietal pleuritis had minor negative effects during 2 intervals neither had a significant adverse effect on total growth rate in either herd (Paisley et al. 1993). R2 values for the regression models were less than 0.27 showing that the lesions present at slaughter explained less than 27% of the variation in herd mean daily gains during any interval.

Survey of the prevalence and types of infertility in beef cows and heifers.

Thirty to 180 days after natural mating to bulls, 8,184 beef cows and heifers were palpated rectally for pregnancy determination. Of these, 7,396 (90%) were pregnant and 815 (10%) were not pregnant. Regarding the nonpregnant cattle, 315 (38.7%) had a normal uterus and a functional corpus luteum, suggesting that they were cycling; the rest had palpable pathologic findings: 455 (55.8%) had ovarian inactivity and uterine atrophy; 16 (2%) had pyometra; 12 (1.5%) had a mummified fetus; 7 (0.9%) had chronic metritis; 6 (0.8%) had an involuting uterus; and 4 (0.5%) had adhesions of the uterus, ovaries, or a combination of both. For the purpose of comparison, the cattle were grouped according to age: group 1-1,035 nulliparous heifers 18 to 24 months of age; group 2-1,137 primiparous heifers 30 to 36 months of age; group 3-5,548 multiparous cows between 4 and 9 years of age; and group 4-464 cows 9 years of age or older. The pregnancy rates were 79.1, 72.2, 96.8, and 78.2, respectively, for these groups. The pregnancy rate for 4- to 9-year-old cows was significantly greater (P less than 0.05) than that for the other age groups.

Mechanisms and therapy for retained fetal membranes and uterine infections of cows: A review.

Literature pertaining to retained fetal membranes and postpartum uterine infections of cows was reviewed. Many therapeutic failures are explained by the effects of the uterine environment on antibacterials, the effects of therapy on the uterine defense mechanism, bacterial resistance patterns and the failure to recognize normal physiological processes in the postpartum period. Prostaglandin F(2)alpha and its analogues provide effective alternatives to antibiotic and antibacterial therapy for most postpartum disorders.

Prevalence of postservice pyometra in a herd of beef cows infected with trichomoniasis: A case report.

Trichomoniasis was detected in a herd of beef cows during a routine yearly reproductive examination. The examination revealed that 12.6% of the cows (75 597 ) were not pregnant. Of these, 65.3% (49 75 ) had a normal uterus and a functional corpus luteum, suggesting that they were cyclic; 21.3% (16 75 ) had pyometra; 6.7% had inactive ovaries and uterine atrophy and were not cyclic; 4% (3 75 ) had an involuting uterus, suggestive of a recent abortion; and 2.7% (2 75 ) had a mummified fetus. The herd had been on a reproductive herd health program for the past 10 years and had averaged 92-99% pregnant cows during this time. Pyometra was diagnosed by rectal palpation, and trichomonads were detected in the pyometra exudate.

Experimental abortion and the systemic immune response to "Haemophilus somnus" in cattle.

"Haemophilus somnus" has been identified in the etiology of bovine abortion on the basis of the isolation of the organism from aborted fetal and placental tissues. To investigate the role of hematogenous dissemination of "H. somnus" in the pathogenesis of abortion and to monitor the humoral immune response to infection, 19 pregnant cows (gestation ages, 1.4 to 7 months) were challenged intravenously (11 cows) or intrabronchially (8 cows). Five cows challenged intravenously aborted, and one cow challenged intrabronchially resorbed her fetus. "H. somnus" was isolated in large numbers from aborted tissues, and placental lesions were similar to those reported in a field case of "H. somnus" abortion. Antibody titers in serum were measured by the microagglutination test (MAT) and by enzyme-linked immunosorbent assay (ELISA). A response to challenge was measured by MAT; it was also measured by ELISA within the immunoglobulin G1 (IgG1), IgG2, and IgM isotypes. On comparison of pre- and postchallenge antibody titers, the greatest and most persistent response was detected within the IgG2 isotype. Prechallenge antibody titers (measured by MAT and by IgG2 ELISA) were lower in animals that aborted than in normal calving animals, indicating that IgG2 antibody may have a role in limiting hematogenous dissemination of "H. somnus."

Seasonal variations in scrotal circumference, sperm quality, and sexual ability in rams.

Seasonal variations in scrotal circumference, spermatozoal motility, morphologically normal spermatozoa, libido, and serving capacity were studied in Suffolk, Lincoln, Columbia, and Polypay rams (5 of each breed). There were marked seasonal variations in the scrotal circumference, morphologically normal spermatozoa, libido, and serving capacity for each of the breed types. Mean scrotal circumference was highest from August through October and lowest in February. Spermatozoa that were morphologically normal were highest in proportion in September (82.3%) and lowest in proportion in February (57.8%). Libido and serving capacity scores were highest from September through November and lowest in March.

The relationship of libido and serving capacity test scores in rams on conception rates and lambing percentage in the ewe.

A procedure for assessing sexual activity (libido) and serving capacity (number of ejaculations) in individual rams is described. Libido and serving capacity was measured by this procedure in 10 Lincoln, 10 Suffolk, 2 Columbia, and 2 Polypay rams prior to the breeding season. Semen was evaluated, and the scrotal circumference measured. The rams were single-sire mated to 10-12 whiteface, crossbred ewes during a 35-day breeding season (September 26 - October 29). First-service conception rates, total conception rate, lambing percentage per ewe lambing, and lambing percentage per ewe exposed were calculated for each ram. The mean libido score for the 24 rams was 8.95 and the mean serving capacity score was 3.16. The 24 rams were classified as low serving capacity (<2 ejaculation), medium serving capacity (2-3 ejaculations), or high serving capacity (>3 ejaculations). There were no significant differences in either conception rate or lambing percentages between the 3 groups of rams. The first-service conception rate, total conception rate, lambing percentage per ewe lambing, and lambing percentage per ewe exposed were 74.8, 93.8, 148.6, and 139.4 percent, respectively, for the 24 rams. Libido and serving capacity tests in the rams, as used in this experiment, prior to the breeding season, were not successful in predicting fertility or prolificacy in ewes.

Detection of early pregnancy factor in swine: A need for dialogue.

The rosette inhibition test, which has been used to detect early pregnancy factor (EPF) and other immunosuppressive factors in the serum of pregnant mice, women, and sheep, was adapted for use in swine. Since the initial methods described for use of the assay were cumbersome and time consuming, our first efforts were to simplify the technique. Substitution of microtiter plates for test tubes and treatment of sheep erythrocytes with AET to increase numbers and stability of the rosettes were successful. Although consistently higher rosette inhibition titers were obtained after incubation of lymphocytes in the serum of pregnant sows compared to controls, the variability in numbers of rosettes obtained on a day to day basis and the cumbersome nature of the test have impeded progress in firmly establishing the presence of EPF in pregnant swine and demonstrating the homology between it and the EPF reported in other species.

The effect of season on the scrotal circumference and sperm motility and morphology in rams.

Seasonal variations in scrotal circumference, sperm motility, morphology, and body weight were studied in ten Suffolk and ten Lincoln yearling rams. There were marked seasonal variations in the scrotal circumference and sperm morphology for each of the breed types. Mean scrotal circumferences were highest in October, 36 cm and 37 cm for Suffolk and Lincoln, respectively. Sperms which were morphologically normal were highest in October, 92.8% for both breeds, and lowest in February, 56.1% and 58.8% for Suffolk and Lincoln, respectively. There was a subsequent rise in percent of normal sperm which was seen in April with a decline again during the summer months. Mean body weight was 251.5 and 192.5 pounds in October and 250.77 and 200.63 pounds in February for Suffolk and Lincolns, respectively.

The effect of scrotal circumference, sperm motility and morphology in the ram on conception rates and lambing percentage in the ewe.

Ten Suffolk and ten Lincoln yearling rams were examined for breeding soundness. Semen was evaluated, and the scrotal circumference measured. They were single-sire mated to ten to twelve, whiteface ewe lambs during a 45-day breeding season (October 15-December 1). First-service conception rates, total conception rate, lambing percentage and lambing percentage per ewe exposed were calculated for each ram and breed. The mean scrotal circumference was 36.5 cm (range 31-41.5 cm). The breeding data was then compared for eleven rams with a scrotal circumference of less than 36.5 cm and nine rams greater than 36.5 cm. No direct relationship was demonstrated between scrotal circumference, sperm motility or morphology with fertility in the rams used in this experiment. However, there was a tendency for rams with a high first-service conception rate to have sired more lambs per ewe exposed. The mean first-service conception rate was 63.2% (range 44.4-80.0%). When rams with greater than 63.2% first-service conception rate were compared to those with less than 63.2%. the lambs born per ewe exposed were 1.04 and .89, respectively, a significant difference (P<.05). There were no significant differences (P<.05) in performance between the two groups when rams with a scrotal circumference less than 36.5 cm and greater than 36.5 cm were compared, nor were there any differences between scrotal circumference, sperm motility, and morphology when rams with total conception rates greater than 80% were compared to those with conception rates less than 80%.

Continuous collection and analysis of bovine oviduct fluid: preliminary results.

Oviduct fluid was collected by cannulating the oviducts of nine cows. The fluid was analyzed for sodium (Na), potassium (K), chlorine (Cl), calcium (Ca), inorganic phosphorus (Pi), magnesium (Mg) concentration and osmolarity (Osm). The mean concentrations +/- the standard error of the means of the constituents were: Na (140.7 +/- 0.37 mEq/L), K (5.12 +/- 0.08 mEq/L), Cl (101.8 +/- 1.54 mEq/L), Ca (1.88 +/- 0.08 mEq/L), Pi (1.97 +/- 0.07 mEq/L), and Mg (1.00 +/- 0.03 mEq/L). Osmolarity was 281.0 +/- 2.56 m Osmols. Significantly lower concentrations of Na (117.4 +/- 2.58 mEq/L) were found in the fluid collected from the oviduct ipsilateral to the ovulating ovary on the day of ovulation. This decrease in Na concentration did not occur in fluid from the contralateral oviduct. The lowest concentrations of Ca were found during days 18 through 21, while the highest were found during days 2 through 6. No significant cyclic changes in the other constituents were observed although the concentrations tended to be highest during days 18 through 21 and lowest on day 1. The concentrations of many of the constituents analyzed were different than those previously reported for bovine oviduct fluid (1).
