[Protein import into mitochondria].

This review is focused on the import of processable precursor proteins into the mitochondrial matrix; the import of carrier proteins into the inner mitochondrial membrane is also briefly discussed. Post- and cotranslational theories of the import, specific features of the presequence structures, and effects of some cytosolic factors on the import of precursor proteins are reviewed. The data on the structure of the protein translocases of the outer (TOM complex) and the inner (TIM complex) membranes of mitochondria and the current models of the precursor protein import by these translocases are also summarized.

[Protein composition of venoms from several species of tropical ants and their effect on mitochondrial H+-ATPase].

Protein compositions of venoms of South-American stinging ants, Ectatomma tuberculatum, Paraponera clavata (subfamily Ponerinae), and "tangarana" were analyzed. The venom of E. tuberculatum displayed the most complex protein composition (more than 15 polypeptides). The water-soluble fraction of the venoms of P. clavata and "tangarana" contained acidic proteins (pI < 3.5 to 5.2), whereas the venom of E. tuberculatum contained predominantly basic proteins (pI 8 to > 9.5). N-Terminal residues and N-terminal sequences of a number of polypeptides were determined. High-molecular-mass polypeptides of the P. clavata venom slightly stimulated the ATPase activity of mitochondrial F1-ATPase. Low-molecular-mass nonprotein components of this venom significantly inhibited the ATPase activity of submitochondrial particles and F1-ATPase from bovine heart mitochondria. The venoms of E. tuberculatum and "tangarana" produced no effect on the ATPase activity.

[Effect of regional (wound) revaccination with tetanus anatoxin on the functional activity of immunocompetent cells in the region].

The study made with the use of complex methods established that the local (wound) application of tetanus toxoid rapidly made the manifestation of the lysosomal apparatus more pronounced, increased the oxidizing activity (determined in the nitro blue tetrazolium test) and phagocytic activity of the mononuclear phagocytizing system in the wound and in the regional lymph nodes. The wound application of tetanus toxoid significantly increased blast transformation of T lymphocytes in guinea pigs simultaneously with tetanus wound infection. The study confirmed the pathogenetic expediency of the proposed method for the stimulation of anti-tetanus immunity by the application of tetanus toxoid on the wound which specifically inhibited the primary stage of the infectious process.

[The functional activity of the macrophages in experimental intestinal dysbacteriosis in rats].

The study of the functional activity of macrophages in the peritoneal exudate and spleen of Sprague-Dawley rats was made under the conditions of dysbacteriosis caused by the administration of antibiotics (gentamicin and phenoxymethylpenicillin) and short-term food deprivation. The complex study of the key functions of macrophages, carried out on the same pool of cells, showed that under the conditions of antibiotic therapy the enhanced function of ingestion was accompanied by a decrease in all other investigated functional parameters of peritoneal macrophages: the intensity of protein synthesis, oxidation metabolism, the activity of lysosomal apparatus, 5'-nucleotide activity. In contrast to changes observed in macrophages after the administration of antibiotics, short-term food deprivation induced increased functional activity as shown by most tests used in this investigation (such as tests for the ingestion of sheep red blood cells, 51Cr, nitro blue tetrazolium, acridine orange, 5'-nucleotidase activity), but did not affect the intensity of protein synthesis in macrophages. The comparison of these results with the data obtained by the authors after the administration of an immunomodulating agent made it possible to regard short-term food deprivation, judging by its effect on the functional activity on macrophages, as an immunostimulating action.

[Differences in the receptor apparatus of resident and meat-peptone broth-stimulated macrophage populations detectable under the action of salmozan].

The degree of expression of lectin-like receptors in resident (RPM) and peptone-elicited peritoneal macrophages (PE-PM) using salmozan as polysaccharide was investigated. Obtained results allow to suppose that salmozan screens lectin-like receptors in macrophages and SRBC. The salmozan screening of lectin-like receptors in macrophages and SRBC resulted in decrease of SRBC uptake level in RPM-subpopulation but did not alter it in PE-PM-subpopulation. The same effect was occurred with the intensified erythrocyte loading of this macrophage subpopulations. It is suggested that one of the functional differences of studied macrophage subpopulation consists in the largest expression of lectin-like receptors in RPM-subpopulation.

[Immunomodulating properties of salmozan and its effect on the functional activity of macrophages].

The effect of salmozan on the resistance of mice to Listeria monocytogenes infection, the formation of delayed hypersensitivity (DH) to sheep red blood cells in the animals, as well as changes in some functional activity characteristics of macrophages have been studied. The study has revealed that salmozan enhances anti-infectious resistance, suppresses the dermal manifestations of DH, and decreases the level of 5'-nucleotidase in peritoneal macrophages, stimulating their phagocytic activity. The intensity of the drug action depends on the time of its administration. The most pronounced immunomodulating action and maximal changes in the function of macrophages have been registered simultaneously after the treatment of the animals with salmozan.

[Regulation of cell function in the macrophagal phagocytic system with salmozan].

Salmozan has been shown to bring about changes in the phagocytic function of peritoneal macrophages, to produce a labilizing effect on the state of lysosomal membranes and to enhance the intensity of protein synthesis in peritoneal macrophages. The presence of relationship between the functional activity of the cells of the macrophagal phagocytic system, the resistance of the body to infections and the capacity for response to an antigenic stimulus has been revealed. The character of changes observed in this investigation has been found to depend on the interval between the injection of salmozan and the injection of sheep red blood cells or challenge with Listeria. The differences in the functional activity of the cells of the macrophagal phagocytic system, appearing under the influence of salmozan, can be attriduted to the action of this immunomodulating agent on various populations of peritoneal macrophages.

[Enhancement of the phagocytic activity of cells of the mononuclear phagocytosing system by the peroral administration of lactobacilli].

The phagocytic function of the peritoneal exudate cells after oral and intraperitoneal administration of Lactobacillus casei subsp. casei, L. plantarum and L. fermentum was studied on mice C57Bl and inbred rats CDF/F344/Crl. The phagocytic function was estimated by the capacity for absorbing and excreting 14C-labeled typhoid vaccine. Intraperitoneal administration of a mixture of various Lactobacillus strains resulted in two-fold increase of the bacteria absorption and digestion when the Lactobacillus strains were administered 4 days before administration of the test antigen. On their simultaneous administration the phagocytic function of the peritoneal exudate cells lowered. Oral administration of the Lactobacillus strains for 6 and 18 days resulted, respectively, in 1.5- and 3-fold increase of absorption and digestion of the labeled typhoid vaccine. This route of Lactobacillus administration provided more rapid increase of the level of the antibodies to the typhoid antigen. Possible immunomodulating action of Lactobacillus administered orally is discussed.

[Mechanism of action of levamisole on cells of the mononuclear phagocytizing system].

The levamisole effect on the phagocytic capacity of macrophages and their membrane permeability was studied in A and CBA mice. It was shown that both in vivo and in vitro, levamisole treatment of peritoneal and splenic macrophages resulted in the diminution of antigen uptake and slowing down of its clearance as compared with control animals. The exposure to levamisole led to the enhanced membrane permeability and the decreased total cathepsin activity in the peritoneal cells and in their lysosomes.

[Changes in the functional activity of cells of the mononuclear phagocytizing system in mice of different ages].

The capacity to phagocytize and the lysosomal cathepsin activity of mononuclear phagocytic system (MPS) cells in mice of different age was studied beginning from the first day of life. The lowest cathepsin activity in peritoneal macrophage cells was revealed in one day-old mice. The activity rose by the third day, approaching the level of adult mice on day 21. The SRBC uptake by macrophages of different origin was not identical in different age groups. The highest uptake was demonstrated by peritoneal macrophages of one-day-old mice. The level of phagocytizing activity in 4-5-day-old mouse macrophages was still high, decreased in 14-day-old mice and reached the control (adult) level in 28-day-old mice. Conversely, spleen macrophages of 1-4-day-old mice had little phagocytizing activity. This activity rose in 2-week-old mice and reached 100% in 2-month old mice. The experiments have shown that lysosomal function and phagocytizing capacity of macrophages obtained from different mouse organs are age-associated. It is suggested that one of the mechanisms of immunologic incompetence lies in the immaturity of MPS cell lysosomes.

[Limits of the phagocytizing capability of macrophages].

The capacity of macrophage peritoneal cells to ingest both different doses of the antigen (sheep red blood cells--SRBC) and the antigens differing in their physico-chemical properties (SRBC, rat red blood cells, typhoid vaccine) was studied. A manifold increase in the SRBC dose failed to influence the amount of the antigen ingestion in the course of a definite time period. When mice macrophage cells were obtained at short periods after the injection of red blood cells of different animal species, there was a decrease of the capacity of these cells to the red blood cells ingestion in vitro. Preincubation of macrophages in vivo or in vitro with all the antigens tested increased sharply their capacity to phagocytize the typhoid vaccine.

[Mitochondrial H+-ATPase: identification of subunits of the F0 subcomplex contacting with membrane lipids].

The subunits of the F0 membrane sector of bovine heart mitochondrial H(+)-ATPase that contact the lipids of the mitochondrial inner membrane were identified with the use of specially synthesized proteoliposomes that contained active mitochondrial H(+)-ATPase and a photoreactive lipid, which was 1-acyl-2-[12-(diazocyclopentadiene-2-carbonylamino)-[12-14C]dodecanoyl]-sn- glycero-3-phosphocholine, 1-acyl-2-[11-([125I]diazoiodocyclopentadiene-2-carbonyloxy)undecanoyl]-sn- glycero-3-phosphocholine, or 1-acyl-2-[12-(diazocyclopentadiene-2-carbonylamino)dodecanoyl]-sn-glycero- 3-phosphocholine, where acyl is a mixture of the residues of palmitic (70%) and stearic (30%) acids. An analysis of the cross-linked products obtained upon the UV-irradiation of these proteoliposomes indicated that subunits c and a of the F0 membrane sector contact the lipids. The cross-linked products were identified by SDS-PAGE and MALDI mass spectrometry.

[The correction action of Phosprenyl and Gamavit on the functional activity of mouse peritoneal macrophages in response to high doses of Staphylococcus aureus alpha-toxin].

The study of the functional activity of peritoneal macrophages of BALB/c mice at different stages of the toxic action caused by S. aureus alpha-toxin (ST) was carried out. The analysis of the dynamics of toxic reaction revealed the main critical points of triggering necrotic processes: the first hour and day 2. One hour after the injection of large doses of ST a sharp increase in the process of antigen binding with its subsequent sharp decrease. Simultaneously, a decrease in the activity of the lysosomal enzymes cathepsin D and acidic phosphatase was established, which was indicative of the destabilization of both lysosomal and cellular macrophage membranes. The increase of oxygen metabolism on day 2, together with the release of lysosomal proteases into the extracellular area, correlated with the maximum death rate of mice and served as the main index of the development of necrosis. The prophylactic and therapeutic use of the preparations Gamavit and Phosprenyl revealed their antitoxic activity and capacityfor stimulating the level of natural body resistance.

[The Amben regulation of the intestinal microflora and macrophage functional activity in an experiment].

The possibility of the correction of intestinal microflora disorders and the functional activity of macrophages in dysbiosis, caused by the intragastric administration of ampiox, with the use of amben (PAMBA), an inhibitor of proteolytic enzymes, was studied. Quantitative and qualitative changes in the main representatives of automicroflora, the functional activity of macrophages in the phagocytosis of 51Cr-labeled sheep red blood cells, the intensity of protein synthesis, the content of cathepsin D, acidic phosphatase and nitro blue tetrazolium activity were determined. The combined administration of ampiox and amben normalized quantitative and qualitative ratios of the main representatives of intestinal microflora, as well as the characteristics of macrophage functional activity, studied in this investigation. The administration of amben to intact animals was found to stimulate bifido- and lactoflora.
