[The plasma levels of endothelin in diabetic retinopathy and their changes after treatment with doxium].

The plasma endothelin (ET) concentrations were determined in 45 diabetes mellitus patients. The changes of ET in 20 diabetic retinopathy patients before and after treatment with doxium and 30 normal controls were observed. The level of plasma endothelins was higher in diabetes mellitus patients than that of the controls (P < 0.05), which was even higher in diabetic retinopathy (P < 0.01). After treatment with doxium, the plasma endothelins level decreased significantly (P < 0.01).

Generation of packaging cell lines for pseudotyped retroviral vectors of the G protein of vesicular stomatitis virus by using a modified tetracycline inducible system.

We have previously shown that the G protein of vesicular stomatitis virus (VSV-G) can be incorporated into the virions of retroviruses. Since expression of VSV-G is toxic to most mammalian cells, development of stable VSV-G packaging cell lines requires inducible VSV-G expression. We have modified the tetracycline-inducible system by fusing the ligand binding domain of the estrogen receptor to the carboxy terminus of a tetracycline-regulated transactivator. Using this system, we show that VSV-G expression is tetracycline-dependent and can be modulated by beta-estradiol. Stable packaging cell lines can readily be established and high-titer pseudotyped retroviral vectors can be generated upon induction of VSV-G expression.

Phenotypes of dnaA mutants of Escherichia coli sensitive to phenothiazine derivatives.

The activation of DnaA protein by cardiolipin is inhibited by fluphenazine in vitro. We therefore examined the sensitivity of temperature-sensitive dnaA mutants of Escherichia coli to fluphenazine and other phenothiazine derivatives. Among the eight dnaA mutants tested, dnaA5, dnaA46 dnaA602, and dnaA604, mutants with mutations in the putative ATP binding site of DnaA protein, showed higher sensitivities to phenothiazine derivatives than did the wild-type strain. The dnaA508 and dnaA167 mutants, which have mutations in the N-terminal region of DnaA protein, also showed higher sensitivities to phenothiazine derivatives. On the other hand, the dnaA204 and dnaA205 mutants, with lesions in the C-terminal region of the DnaA protein, showed the same sensitivity to phenothiazine derivatives as the wild-type strain. Complementation analysis with a plasmid containing the wild-type dnaA gene and phage P1-mediated transduction confirmed that dnaA mutations are responsible for these sensitivity phenotypes.

Keratinocyte growth factor is required for hair development but not for wound healing.

Keratinocyte growth factor (KGF), also known as fibroblast growth factor 7 (FGF7), is synthesized by skin fibroblasts. However, its mitogenic activity is on skin keratinocytes, where it is the most potent growth factor identified thus far. To explore KGF's function in vivo, we used embryonic stem cell technology to generate mice lacking KGF. Over time, their fur developed a matted appearance, very similar to that of the rough mouse, whose recessive mutation maps at or near the KGF locus on mouse chromosome 2. In contrast to the recently reported transforming growth factor-alpha (TGF-alpha) and FGF5 knockouts, which showed defects in the follicle outer-root sheath and the hair growth cycle, respectively, the hair defect in the KGF knockout seemed to be restricted to the cells giving rise to the hair shaft. Thus, we have uncovered a third, and at least partially nonoverlapping, growth factor pathway involved in orchestrating hair follicle growth and/or differentiation. Surprisingly, the absence of KGF resulted in no abnormalities in epidermal growth or wound healing. This was true even when we engineered double knockout mice, null for both KGF and TGF-alpha, two factors that are increased dramatically in the normal wound-healing process. Whereas we found no evidence of compensatory changes at the mRNA level of wounded knockout mice, these data imply that the regulation of epidermal growth is complex and involves a number of growth stimulatory factors that go beyond what are thought to be the major paracrine and autocrine growth factors. We suggest that the redundancy in epidermal growth and wound healing is likely to stem from the vitality of these functions to the organism, a feature that is not a consideration for the hair follicle.

Macro-ergonomic risk assessment in nuclear remediation industry.

In this study, a macro-ergonomic risk assessment tool was developed based on criteria of scope, simplicity, practicality, usefulness, reliability and job-specificity. A relative stress index 'RSI' was formulated to take into account multiple parameters, such as frequency, duration, repetition, weight, force, travel distance and horizontal distance. This tool was tested in nuclear remediation industry. The results are presented and discussed.

Projections of the medial and lateral prefrontal cortices to the amygdala: a Phaseolus vulgaris leucoagglutinin study in the rat.

The projections of different subfields of the medial and lateral prefrontal cortices to the amygdala were studied in the rat using the sensitive Phaseolus vulgaris leucoagglutinin anterograde tract tracing technique. Injections into the infralimbic cortex produced anterograde labeling in the lateral capsular subdivision of the central nucleus, superficial (corticomedial) amygdaloid nuclei, lateral and accessory basal nuclei, and the anterior amygdaloid area. Injections into the caudal portion of the infralimbic cortex produced additional labeling in the intermediate subdivision of the central nucleus. The prelimbic cortex had projections to the medial portion of the magnocellular basal nucleus and adjacent portions of the lateral nucleus and lateral capsular subdivision of the central nucleus. The medial precentral cortex had projections to the rostromedial part of the magnocellular basal nucleus and adjacent portions of the lateral capsular subdivision of the central nucleus. Injections into the lateral orbital and ventral agranular insular cortices produced labeled fibers in the rostral part of the superficial amygdala, lateral capsular subdivision of the central nucleus, and the lateral and accessory basal nuclei. The dorsal agranular insular area had projections to several different subdivisions of the central nucleus as well as to the rostrolateral magnocellular basal nucleus; the latter projections were complementary to those originating in the prelimbic area. The present study indicates that each portion of the prefrontal cortex has a distinctive projection to the amygdala. The ventral areas of the lateral and medial prefrontal cortices, which receive olfactory projections, are the only prefrontal cortical areas with projections to the olfactory-related superficial amygdaloid nuclei. The more dorsally situated prefrontal areas, the dorsal agranular insular area and prelimbic cortex, have complementary projections to the basal nucleus, suggesting that they modulate separate prefrontal cortico-striatal-pallid circuits. The specificity of prefrontal cortico-amygdaloid projections is indicative of their involvement in discrete functions.

Coronary vascular and endothelial reactivity changes in transgenic mice overexpressing atrial natriuretic factor.

Recent advances in genetic methods permit the introduction of random and defined mutations into the mouse germ line, producing novel mouse strains, some of which affect the heart and vasculature. A TTR-ANF transgenic strain of mice, which constitutively expresses a fusion gene consisting of the transthyretin promoter and the ANF structural gene, has been shown to result in a lifelong elevation of plasma atrial natriuretic factor (ANF) and a chronically lowered arterial blood pressure. However, no established method for efficient functional analysis of possible alterations in coronary vascular function in mice has been reported. In the present study, we describe an isolated mouse coronary artery preparation that permits an effective and reproducible evaluation of coronary endothelial and vascular function. Both left main and right coronary arteries (resting luminal diam 70-90 microns) were isolated and pressurized, and changes in luminal diameter were determined by videomicroscopy. The coronary pressure-luminal diameter relationship was not significantly different between TTR-ANF transgenic and nontransgenic littermates. Relaxation of coronaries to 0.1-100 nM ANF was significantly reduced in transgenic [maximum effect (Emax) = 43 +/- 10% (mean +/- SE) of 11 vessels] compared with nontransgenic (Emax = 73 +/- 7%, n = 15) mice. Similarly, the relaxant response to an endothelium-dependent dilator, acetylcholine, but not to endothelium-independent dilators sodium nitroprusside and isoproterenol, was significantly decreased in transgenic (Emax = 46 +/- 10%, n = 12) compared with nontransgenic (Emax = 85 +/- 5%, n = 14) mice. In contrast, the dose-dependent vasoconstriction to endothelin-1, KCl and the thromboxane mimetic U-46619 was not significantly different between the two groups. These results indicate that lifelong ANF elevation in mice is associated with a decreased responsiveness of coronary vasorelaxation to ANF, possibly resulting from receptor downregulation and/or desensitization. Endothelium-dependent relaxation was also significantly depressed in transgenic mouse coronary arteries, but smooth muscle-specific dilation and constriction were not affected. The present findings are consistent with previous studies of TTR-ANF transgenic mice showing that ANF regulates arterial blood pressure and vascular function.

A preliminary study on inherited tooth morphology characters of Japanese and Chinese young adults--with special reference to the Carabelli tubercle.

A comparative study on inherited tooth morphology characters, in particular the incidence of Carabelli tubercles in the maxillary dentition, was conducted on standardized stone casts obtained from 240 Japanese (124 males (male) and 116 females (females)) and 160 Chinese (74 male, 86 female) young adults. The following results were obtained: 1) No significant differences in the incidence of Carabelli tubercles according to sex or body height were found between the two groups. 2) However, significant differences in the incidence of Carabelli tubercles according to sex were found in both groups. The tubercles occurred more frequently in males. 3) Significant differences between the bilateral and unilateral occurrence of Carabelli tubercles were observed in both the Japanese (p < 0.05) and Chinese (p < 0.01) groups; Carabelli tubercles on upper first molars were always bilateral. 4) The highest incidence of Carabelli tubercles was found in individuals with a body height of 166 approximately 175 cm in both groups, and a significant difference (p < 0.05) between the 156 approximately 165 cm and 166 approximately 175 cm groups was particularly evident in the Japanese adults. The authors postulate that bilateral occurrence of Carabelli tubercles was originally an inherited character in the molar region, but that the character was inhibited during the process of evolution of the masticatory system and regression of the molar dentition.

[Preliminary exploration on Zhang Jingyue's thinking of Chinese psychiatry].

As a great medical scholar of the Ming Dynasty with great achievements, Zhang Jingyue made great contributions to the course of TCM, both clinical and theoretical. This paper explores his achievements in psychiatry from the following points: 1. Expounding of etiology and pathogenesis of depressive psychoses; 2. Recognition on etiology and pathogenesis of Mania; 3. Recognition of some special psychoses; 4. Recognition of epilepsy; 5. Recognition of dementia; 6. Recognition of depression; 7. Diagnosis of feign disorders.

[Expression of epidermal growth factor receptor (EGFR) in endometrioid carcinoma of the ovary].

OBJECTIVE: To determine the expression of EGFR expression in endometrioid carcinoma of the ovary and its relationship to clinicopathologic features and prognosis. METHODS: Expression of EGFR oncoprotein was studied by immunohistochemistry in paraffin-embedded tumor tissues from 28 patients with endometrioid carcinoma of the ovary. RESULTS: Tissues from 13 patients (46.4%) were stained positively with EGFR polyclonal antibody (1005). The staining was membranous and/or cytoplasmic. There was no relationship between EGFR expression and age at diagnosis, preoperative CA125 titer, clinical stage, histologic grade, and lymph node metastases. A higher expression rate was observed in patients with residual tumors (56.3% versus 33.3%). The mortality rate was higher in patients with EGFR expression than in those without EGFR expression (46.2% versus 14.3%), but the difference was not statistically significant. CONCLUSION: EGFR expression was not related to the clinicopathologic features in endometrioid carcinoma of the ovary, but was related to the poor prognosis of this disease.

[Uterine papillary serous carcinoma--one kind of special type of uterine endometrial carcinoma].

OBJECTIVE: To study the clinico-pathologic features of uterine papillary serous carcinoma and point out the complete difference between the two kinds of endometrial carcinoma. METHODS: One hundred and ten cases of endometrial carcinoma who received treatment in the our hospital from May 1, 1982 to April 30, 1994 were analyzed retrospectively. RESULTS: Of them, 10 cases (9.1%) of uterine papillary serous carcinoma (UPSC) were found. The age in average was 67.2 years old, which is nearly 10 years older than those with ordinary endometrial cancer (P < 0.01). The occurrence of UPSC was revealed no relationship with the relative factors of occurrence of endometrial carcinoma. The study shows that tumor has very marked tendency of lymph node metastasis, much worse grade, much deeper myometrial invasion and much earlier extrauterine disease. CONCLUSIONS: UPSC is a special pathologic type of endometrial carcinoma with completely different clinico-pathologic behavior. The surgical staging is necessary for the patients with UPSC to get comprehensive stage because the clinical stage is usually not correct enough.

[Clinicopathological evaluation on ovarian serous borderline tumours and their implants].

OBJECTIVE: To investigate the clinicopathologic features and the expressions of proliferation cell nuclear antigen (PCNA) in ovarian serous borderline tumours (OSBT). METHODS: The primary ovarian tumours, implants and recurrent specimens of 48 cases of OSBT were studied. Expression of PCNA was determined by avidin-biotin-peroxidase complex (ABC) method. Clinical data for all patients were abstracted from hospital records. RESULTS: While the primary ovarian tumours have the characteristic histological pictures of borderline tumour, three different types of histologic appearance including benign, noninvasive and invasive were shown by their peritoneal implants. All benign and most noninvasive implants were PCNA negative or weakly positive, and the 25 patients were free from tumour 5-28 years after their operations. Some of the noninvasive implants (9/22) were recurrent, and a few (3/22) underwent malignant transformation 10-23 years after surgery. The only one invasive implant was PCNA strongly positive and the patient died 3.5 years after surgery. CONCLUSION: The different histologic types of the peritoneal implants are of prognostic significance and are factors to be considered in determining the treatment project. The diagnostic value of PCNA immunoreactivity in implantations is confirmed.

[Clinicopathological features of human papilloma virus associated squamous cell carcinomas of the vulva].

OBJECTIVE: To investigate the relationship between human papilloma virus (HPV) status and clinicopathological features of vulva carcinoma and its clinical significance. METHODS: Histological sections of 37 cases of vulval carcinoma were subclassified. Standard PCR (HPV types 6/11, 16, 18) were applied to formalin fixed, paraffin wax sections. RESULTS: There were 24 cases of keratinizing squamous cell carcinoma (KSC), 7 cases of basaloid carcinoma (BC) and 6 cases of warty carcinoma (WC). HPVs, Mainly HPV 16, were detected in 10 of the 37 cases, and all of them were women with BC and WC types. Although BC and WC constituted 35% of the total number of cases, the HPV detective rate was significantly higher than that of KSC (83.3% vs 0.0%), and the tumors were frequently accompanied by valvar intra-epithelial neoplasm (VIN). Clinically, women with BC and WC were younger than women with KSC (mean age at diagnosis for WC, BC and KSC were 36.7, 58.9, 63.3 years respectively). Besides, the incidence of lymph node metastasis in BC and WC were lower than that in KSC (7.6% vs 25.0%). BC and WC were more likely to have another invasive or noninvasive squamous neoplasm of the cervix and vagina (3/13 vs 0/24), and recurrence rate was higher (12.5% vs 7.7%). However, their survival rates following surgical treatment of recurrences were also higher. Squamous cell carcinoma of the vulva are etiologically diverse tumors which have different clinical and pathological profiles. CONCLUSION: It is suggested that an etiological classification of squamous carcinoma of the vulva may at least be grouped at present into a small but substantial proportion (BC and WC) related to HPV and the majority portion (KSC) of unknown etiology.

[Reappraisal of the pathological criteria for uterine leiomyosarcoma].

In order to study the pathologic criteria for uterine Leiomyosarcoma, 42 cases of morphologically malignant or uncertain malignant potential uterine smooth muscle tumors which exhibited recurrences or metastasis, or were followed for a minimum of 2 years clinically, were analysed and reappraised. The results showed that in comparison with the leiomyoma group, active mitosis (mitotic index, MI > or = 10/10HPF), significant cytologic atypia and coagulative tumor cell necrosis (CTCN) were more common in the leiomyosarcoma group (71.4%, 92.9% and 71.4% respectively). Whereas 4 of the 14 leiomyosarcomas had MI < or = 4/10HPF, developed recurrences or metastasis, and 7 of the 28 leiomyomas had MI > or = 5/10HPF, including 2 with MI > 10/10HPF were alive and well 5.5-23.0 years after operation; hence a mitotic count is an unreliable prognostic criterion for uterine smooth muscle tumors. The data suggest that a trivariate rule using MI, the degree of atypia and CTCN best identifies the aggressive potential of uterine smooth muscle tumors.

Transmembrane gradient driven phase transitions within vesicles: lessons for drug delivery.

Phase transitions in closed vesicles, i.e., microenvironments defined by the size of the vesicle, its contents, and permeability of its membrane are becoming increasingly important in several scientific disciplines including catalysis, growth of small crystals, cell function studies, and drug delivery. The membrane composed from lipid bilayer is in general impermeable to ions and larger hydrophilic ions. Ion transport can be regulated by ionophores while permeation of neutral and weakly hydrophobic molecules can be controlled by concentration gradients. Some weak acids or bases, however, can be transported through the membrane due to various gradients, such as electrical, ionic (pH) or specific salt (chemical potential) gradients. Upon permeation of appropriate species and reaction with the encapsulated species precipitation may occur in the vesicle interior. Alternatively, these molecules can also associate with the leaflets of the bilayer according to the transmembrane potential. Efficient liposomal therapeutics require high drug to lipid ratios and drug molecules should have, especially when associated with long circulating liposomes, low leakage rates. In this article we present very efficient encapsulation of two drugs via their intraliposomal precipitation, characterize the state of encapsulated drug within the liposome and try to fit the experimental data with a recently developed theoretical model. Nice agreement between a model which is based on chemical potential equilibration of membrane permeable species with experimental data was observed. The high loading efficiencies, however are only necessary but not sufficient condition for effective therapies. If adequate drug retention within liposomes, especially in the case of long-circulating ones, is not achieved, the therapeutic index decreases substantially. Anticancer drug doxorubicin precipitates in the liposome interior in a form of gel with low solubility product and practically does not leak out in blood circulation in the scale of days. With an antibiotic, ciprofloxacin, the high loading efficacy and test tube stability is not reproduced in in vitro plasma leakage assays and in vivo. We believe that the reasons are higher solubility product of precipitated drug in the liposome, larger fraction of neutral molecules due closer pK values of the drug with the pH conditions in the solutions and high membrane permeability of this molecule. High resolution cryoEM shows that encapsulated anticancer agent doxorubicin is precipitated in the form of bundles of parallel fibers while antibiotic ciprofloxacin shows globular precipitate. Doxorubicin gelatin also causes the change of vesicle shape.

Hemolytic, but not cell-invasive activity, of adenylate cyclase toxin is selectively affected by differential fatty-acylation in Escherichia coli.

Adenylate cyclase toxin from Bordetella pertussis requires posttranslational acylation of lysine 983 for the ability to deliver its catalytic domain to the target cell interior and produce cyclic adenosine monophosphate (cell-invasive activity) and to form transmembrane channels (hemolytic activity). When the toxin is expressed in Escherichia coli, it has reduced hemolytic activity, but comparable cell-invasive activity to that of adenylate cyclase toxin from B. pertussis. In contrast to the native protein from B. pertussis, which is exclusively palmitoylated, recombinant toxin from E. coli is acylated at lysine 983 with about 87% palmitoylated and the remainder myristoylated. Furthermore, the recombinant toxin contains an additional palmitoylation on approximately two-thirds of the lysines at position 860. These observations suggest that the site and nature of posttranslational fatty-acylation can be dictated by the bacterial host used for expression and can have a significant, but selective, effect on protein function.

Molecular cloning and expression of alfalfa (Medicago sativa L.) vestitone reductase, the penultimate enzyme in medicarpin biosynthesis.

Medicarpin, the major phytoalexin in alfalfa, is synthesized by way of the isoflavonoid branch of phenylpropanoid metabolism. One of the final steps of medicarpin biosynthesis, from vestitone to 7,2'-dihydroxy-4'-methoxyisoflavanol, is catalyzed by vestitone reductase. A 1245-bp cDNA clone which encodes vestitone reductase was identified utilizing internal amino acid sequence of purified vestitone reductase. When expressed in Escherichia coli, the cloned enzyme exhibits strict substrate stereospecificity for (3R)-vestitone, as was observed for vestitone reductase purified from alfalfa. The calculated molecular weight of the protein (35,918) is similar to that of purified vestitone reductase from alfalfa (38 kDa by SDS-PAGE). The levels of vestitone reductase transcript (1.35 kb) greatly increase within 2 h of elicitor addition to alfalfa cell suspension cultures, preceding the rapid increases in vestitione reductase enzyme activity and medicarpin biosynthesis. In healthy alfalfa plants, the highest levels of transcripts were detected in roots and root nodules, consistent with the synthesis of medicarpin and its conjugate in these tissues. The cloning of the vestitone reductase gene provides a specific tool for the study and manipulation of pterocarpan biosynthesis in legumes.
