Regional displacement by sulpiride of [3H]spiperone binding in vivo. Biochemical and behavioural evidence for a preferential action of limbic and nigral dopamine receptors.

The regional displacement by sulpiride of [3H]spiperone binding in vivo was studied in the rat. A low dose of sulpiride (20 mg/kg) displaced [3H]spiperone binding in certain limbic regions (olfactory tubercle, septum) and the substantia nigra but not in the nucleus accumbens or striatum. At this does sulpiride preferentially blocked apomorphine induced apomorphine Higher doses of sulpiride (150 and 250 mg/kg), which blocked apomorphine induced stereotypes and induced catalepsy were found to displace [3H]spiperone binding in all regions studied including the striatum and the nucleus accumbens. Haloperidol (0.1 and 1.0 mg/kg) displaced [3H]spiperone to approximately the same extent in all regions studied.

Forebrain projections of the ventral tegmentum as studied by axonal transport of [3H]dopamine in the rat.

Twenty-four hours after [3H]dopamine ([3H]DA) injections into different parts of the ventral tegmentum the radioactivity recovered in the forebrain showed a highly regional distribution which corresponded to well-known DA projections. Selective degeneration of DA-neurons with 6-hydroxydopamine (6-OHDA) markedly reduced the axoplasmic transport after [3H]DA injections. The ventral tegmental area (VTA) was found to project primarily to the nucleus accumbens, olfactory tubercle and amygdala. The DA-innervation of the entorhinal cortex and the amygdala derive from cells widely distributed in the VTA and SN.

Presence of NADPH-cytochrome P450 reductase in central catecholaminergic neurones.

The cytochrome P450-containing mixed function oxidases metabolize a variety of endogenous and exogenous compounds including drugs, carcinogens, fatty acids and steroids. Mixed function oxidases have been detected in several tissues, including brain. The enzyme system consists of a lipid fraction (phosphatidylcholine), cytochrome P450 and NADPH-cytochrome P450 reductase. NADPH-cytochrome P450 reductase has been purified to apparent homogeneity and demonstrated to supply reducing equivalents from NADPH to cytochrome P450 (refs 5-7). Detection of NADPH-cytochrome P450 reductase thus represents an indirect means of demonstrating the presence of cytochrome P450. Although the role of cytochrome P450 in the central nervous system (CNS) is not known, it may include such different functions as metabolism of xenobiotics, aromatization of androgens to oestrogens and the formation of catecholoestrogens. Despite the potentially very important role(s) of cytochrome P450 in brain function, its exact regional distribution remains essentially unknown. Using a specific antibody against rat liver NADPH-cytochrome P450 reductase in combination with immunohistochemical techniques, we have now localized this enzyme to define catecholamine (CA)-containing structures of the rat and monkey brain.

Hand function in women and men with early rheumatoid arthritis. A prospective study over three years (the Swedish TIRA project).

OBJECTIVE: To describe the course of hand function in women and men during the first 3 years after diagnosis of recent-onset rheumatoid arthritis (RA), to investigate sex differences in hand function, and to study correlations between and within hand function assessments. METHODS: A total of 276 patients (69% women) with RA of a maximal duration of 12 months were recruited to the study. Hand function was assessed by the Grip Ability Test (GAT) and Signals of Functional Impairment (SOFI). Peak and average grip force over 10 s in the right and left hand was measured by an electronic device. RESULTS: Hand function was affected at diagnosis, but had improved significantly at the 3-months' follow-up and then remained stable (but still affected) in both women and men. As assessed by SOFI, hand function was worse in men than in women, whereas women had significantly lower grip force. GAT, grip force, and SOFI correlated weakly. The average and peak values of grip force correlated strongly, as did the grip force in the right and the left hand. CONCLUSION: Hand function was profoundly affected at diagnosis of RA, but improved significantly within 3 months and remained stable (but still affected) over 3 years. As expected, women on average had significantly lower grip force than men.
