Phorbol ester induces a differential effect on the effector function of human allospecific cytotoxic T lymphocyte and natural killer clones.

Six-day allosensitized human peripheral blood lymphocytes treated for 12-18 hr with phorbol 12-myristate 13-acetate (PMA) were found to lose the capacity to kill the specific target in a standard cell-mediated lympholysis (CML) assay, but they were still effective in their ability to kill the tumor cell line K562. We investigated which antigens on the cell membrane involved in alloimmune recognition might be modified by PMA, since it was found that in a lectin-dependent CML assay, the lytic mechanism was not impaired. For this purpose, T3+, T4+ allospecific cytotoxic T lymphocyte (CTL) clones and either T3+ or T3- natural killer (NK) clones were generated from 6-day allostimulated lymphocytes. The results indicated that PMA inhibited the cytolytic function of both alloimmune CTL and NK T3+ clones. In contrast, PMA did not modify the effector cell function of T3- NK clones. Phenotypic analysis of T-cell surface antigens from T3+ clones showed that T3 molecule expression on the cell membrane was reduced by 80-90% after PMA treatment, whereas expression of both accessory T4 molecules, involved in antigen recognition, and receptor for interleukin 2 was increased. Moreover, the loss of function was transitory and could be restored 4 days after PMA treatment when the T3 molecules were fully reexpressed at the cell surface. Taken together, these data strongly suggest that (i) PMA prevents cell-mediated cytotoxicity by modulating the disulfide-linked heterodimer associated to T3 and described as the receptor for antigen on the cell surface of major histocompatibility complex (MHC) and non-MHC specific CTL clones, without affecting the lytic mechanism per se, and (ii) the expression of the receptor for the antigen present on the tumor cell line K562 is not decreased on T3- NK clones after PMA treatment and must be different from that on T3+ T-cell clones.

Separation and further characterization of early and late rosette-forming cells.

Human peripheral blood T-lymphocytes (PBL) can be separated into early rosette-forming cells or active T cells (A-RFC) and late rosette forming cells (L-RFC) according to the time required for formation of rosettes with sheep red blood cells (SRBC). Experimental results showed that treatment of the SRBC with neuraminidase was necessary for the formation of stable early rosettes. The optimum proportion of SRBC to lymphocytes for total rosette formation was 64:1, whereas a 16:1 ratio yielded maximal early rosette formation. The proliferative responses of A-RFC to concanavalin A (Con A) or to alloantigens were less than those seen with L-RFC. The percentage of T gamma cells in A-RFC was significantly lower than in L-RFC. Leukocyte inhibitory factors (LIF) released by the A-RFC fraction after treatment with Con A were more effective inhibitors of the migration of polymorphonuclear leukocytes (PMN) than the factors released from the L-RFC fraction. These results suggest that A-RFC represent a subpopulation of T lymphocytes which are more active in certain lymphocyte responses than another subpopulation which forms rosettes more slowly with sheep red blood cells.

HLA loss variants of a B27+ lymphoblastoid cell line: genetic and cellular characterization.

Variants of a lymphoblastoid cell line, LCL 526 (SB3 MB1 DR1 B44 C5 A2/SB4 MT4 DR4 B27 C2 A24), which lost various HLA specificities were selected with monoclonal antibodies and complement using a method developed by Kavathas et al. (PNAS 77:4251, 1980). Using alpha B27 monoclonals, 8 B27 only loss mutants and 4 B27 haplotype multiple loss mutants were generated. The parental LCL 526 and two of the B27- mutants were used to select alpha B27 CTLs. The selection of six A2 loss, one A2-C5 loss, and 14 A2 haplotype multiple loss variants as well as secondary selection on haplotype loss variants to obtain A null, B null, DR null, and total A, B, C, null variants is also described. The usefulness of these mutants for the study of the relationship between B27 and disease and as two new haplotypes for immunologic, genetic, and molecular research is discussed. These mutants are available to other researchers.

A cytolytic human T lymphocyte clone differentially recognizing HLA-B27 subtypes.

A cytolytic human T cell (CTL) clone, designated F/M-F159, has been produced, the lytic specificity of which distinguishes subtypes of HLA-B27. This was demonstrated in cell-mediated lympholysis (CML) assays of: 1) a panel of target cells from unrelated donors, 75 B27 + and 36 B27-; 2) six families, including 20 B27 + and 14 B27- individuals; and 3) B27 + and B27- variants of a B27+ lymphoblastoid cell line (LCL). Specificity of F/M-F159 for HLA-B27 was confirmed by blocking studies with monoclonal antibodies. Lysis of B27 + targets reactive with the anti-B27 monoclonal antibody B27M2 was 30-104%, while lysis of B27 +, B27M2- targets was 4-22%. Lysis of B27- targets expressing HLA-Bw47, known to be cross-reactive with the B27M2 antibody, was 10 to 19%, while lysis of all other B27- targets was less than or equal to 10%. Clone F/M-F159 lysed B27 + targets, and failed to lyse B27- targets, irrespective of the clinical status of the cell donors. It is concluded that F/M-F159 recognizes an epitope present on the majority of serologically identified HLA-B27 molecules and that this epitope is closely related to, but not identical with, the epitope recognized by the antibody B27M2. These findings are interpreted as supporting a direct role for HLA-B27 in disease pathogenesis.

Helper cell-independent cytotoxic clones in man.

We report here a class of helper cell-independent cytotoxic T cell (HITc) clones in man that can proliferate in response to antigenic stimulation as well as mediate cytotoxicity. HITc appear to be rare among clones derived from primary in vitro allosensitized culture, but constitute the majority of clones derived from cells sensitized to autologous Epstein-Barr virus-transformed lymphoblastoid cell lines. The implications of the derivation and function of HITc clones are discussed.

Nitrofurantoin solubility in aqueous urea and creatinine solutions.

Experiments were carried out to determine the effect of urea and creatinine on the solubility of nitrofurantoin in water at different temperature and pH conditions. The addition of urea to aqueous media increased nitrofurantoin solubility up to a maximum concentration level and then decreased solubility at higher urea concentrations. The amount of urea needed to bring about maximum nitrofurantoin solubility was dependent on temperature and ranged between 1.75 and 2.50%. Spectral studies suggest a possible interaction between urea and nitrofurantoin molecules. Nitrofurantoin solubility increased with an increasing creatinine concentration ranging from 0.05 to 1.6%. Spectral studies indicate a strong interaction between creatinine and nitrofurantoin molecules in solution. The combined effect of urea and creatinine of the solubility of nitrofurantoin could account for the absence of crystalluria with this drug, even though unusually high concentrations in urine have been reported.

Fibrolamellar hepatocellular carcinoma--an atypical MR manifestation.

We report the imaging findings, specifically magnetic resonance pictures of a non-cirrhotic case of fibrolamellar hepatocellular carcinoma in an older patient (60 years). Computed tomography revealed non-enhanced hypodense central scar without notable intratumoral calcification. MR imaging revealed a mass, which was homogeneously hypointense on T1-weighted images (T1WI) and turned hyperintense on T2-weighted images (T2WI). Stellate central scar was depicted with more hypointensity on T1WI, however, it became markedly hyperintense on T2WI. This manifestation is very different from the typical pattern reported in the literature. In gadolinium enhanced T1WI, the mass other than the central scar got enhanced. Fibronodular hyperplasia and fibrolamellar hepatocellular carcinoma should be differentiated with reference to the clinical and imaging findings. Collagenous central scar was described pathologically within the latter. However, a discrepancy between the pathological and magnetic resonance pattern concerning the central scar which was collagenous and corresponding to hyperintensity on T2WI was found. To our knowledge no similar entity has been described in the literatures so far.
