The rat fructose-1,6-bisphosphatase gene. Structure and regulation of expression.

The gene encoding rat fructose-1,6-bisphosphatase was isolated from a rat genomic Charon 4A library by screening with a cDNA to the rat liver enzyme. Southern blotting of rat genomic DNA showed that there is a single copy of the fructose-1,6-bisphosphatase gene. It extends over 23 kilobases and is composed of seven exons and six introns that range in size from 93 to 267 base pairs and from 1,400 to 11,300 base pairs, respectively. The intron/exon boundary sequences conform to consensus acceptor (GTn) and donor (nAG) sequences, and the exons in the gene appear to code for functional protein domains. The transcription start site, determined by 5'-extension sequencing of mRNA, was assigned to a guanine 119 bases 5' to the translation initiation AUG. The sequence of the gene upstream to the cap site contains characteristic RNA polymerase II promoter-binding sites: a putative TATA box at position -29 and a Sp 1 binding site (GGGGCGGAGA) at position -48. A 1,300-base pair fragment of 5'-flanking sequence containing these elements, ligated upstream from a firefly luciferase reporter gene and transfected into cultured normal rat kidney cells, demonstrated strong promoter activity. The accumulation of fructose-1,6-bisphosphatase mRNA in hepatocytes incubated with cAMP suggests that the gene may be cAMP-responsive, which is consistent with the presence of three consensus cAMP regulatory elements at positions -169, -282, and -698 in the 5'-flanking region of the gene. Expression of the fructose-1,6-bisphosphatase promoter-driven luciferaes gene was 2-3-fold activated by the cyclic nucleotide, suggesting that one or more of these elements may be functional. On the other hand, insulin decreased the expression of the endogenous gene in hepatocytes. Thus, expression of the fructose-1,6-bisphosphatase gene is regulated independently by both cAMP and insulin.

Isolated permeabilized rat heart cells as a model to study heart bioenergetics in situ.

Isolated rat heart cells permeabilized by digitonin were examined as an experimental model to study heart bioenergetics. The cells displayed good parameters of oxidative phosphorylation (acceptor control ratio ca. 8 with pyruvate + malate). High ATPase activity detected in the cells was characterized. The ATPase activity was Ca-dependent (optimum [free Ca] ca. 400 nM), and Mg was necessary for its full activity. Double reciprocal plot l/v vs. 1/[free Ca] at physiological [free Ca] was linear, thus showing free Ca to be a substrate for the ATPase (Km for Ca ca. 149 nM). Double reciprocal plot 1/v vs. 1/[ATP] was also liner, thus showing the ATPase activity could be ascribed to a single enzyme. The ATPase is supposed to represent Ca, Mg-ATPase of sarcoplasmic reticulum. The ATPase activity appeared to be functionally coupled to oxidative phosphorylation of the cells by apparently preferring ATP supplied by mitochondria (KmATP = 74 microM) to external ATP (KmATP = 169 microM; P less than 0.05). Apparent preference by oxidative phosphorylation for ADP supplied by the ATPase (KmATP = 45 microM) to external ADP (KmATP = 152 microM) was also manifested by significant difference (P less than 0.02) in Km.

Ca, Mg-ATPase activity of permeabilised rat heart cells and its functional coupling to oxidative phosphorylation of the cells.

Isolated rat heart cells permeabilised by digitonin were examined as an experimental model to study heart bioenergetics. The cells showed good indices of oxidative phosphorylation (acceptor control ratio about 8 with pyruvate plus malate). The adenosine triphosphatase activity detected in the cells was high and was calcium dependent (optimum [free calcium] about 400 nmol.litre-1); magnesium was necessary for its full activity. Double reciprocal plot l/v vs 1/[free calcium] at physiological free calcium concentrations was linear, thus showing free calcium to be a substrate for the adenosine triphosphatase (Km for calcium about 149 nmol.litre-1). Double reciprocal plot 1/v vs 1/[ATP] was also linear, thus showing that the adenosine triphosphatase activity could be ascribed to a single enzyme. Oxidative phosphorylation and the ATPase activity of the cells appeared to be functionally coupled. This was manifested by apparent preference by oxidative phosphorylation for adenosine diphosphate supplied by the adenosine triphosphatase activity (Km 45 mumol.litre-1) to external adenosine diphosphate (Km 152 mumol.litre-1; p less than 0.02). Apparent preference by the adenosine triphosphatase activity for adenosine triphosphate supplied by mitochondria (Km 74 mumol.litre-1) to external adenosine triphosphate (Km 169 mumol.litre-1) was also manifested by a significant difference in Km values (p less than 0.05).

Mitochondrial pyruvate carrier--a possible link between gluconeogenesis and ketogenesis in the liver.

Effects of various ketogenic substrates on gluconeogenesis from lactate or alanine were compared. The results suggest that, in intact liver cells, cytoplasmic pyruvate is transported into mitochondria in exchange for intramitochondrially generated acetoacetate. An interrelationship between gluconeogenesis and ketogenesis may thus exist in the liver at the level of mitochondrial pyruvate carrier.

Mutual additivity of hormonal and non-hormonal stimulation of gluconeogenesis.

Possible mutual additivity of the stimulating effects of fatty acids and alpha-adrenergic agents on gluconeogenesis was examined using isolated rat liver cells. Adrenaline or noradrenaline alone stimulated gluconeogenesis from lactate by 34% both in the absence and presence of propranolol. Oleate or acetate alone stimulated gluconeogenesis by 76% and 45%, respectively; propranolol did not influence the effects. Simultaneous administration of alpha-adrenergic agents with oleate or acetate increased gluconeogenesis by 110% and 90-100%, respectively, thus documenting mutual additivity of hormonal and non-hormonal stimulation; propranolol did not affect the mutual additivity of the effects observed.

Evidence for possible interrelationship between gluconeogenesis and ketogenesis?

Effects of ketogenic substrates on gluconeogenesis by rat hepatocytes were examined. D,L-3-Hydroxybutyrate (5 mM) stimulated gluconeogesis from lactate by 41%, the effect being the same as that of 5 mM acetate (43%). Acetoacetate up to 40 mM had no stimulating effect; conversely, it partially or completely abolished the observed stimulating effects of acetate, 3-hydroxybutyrate, or oleate. 3-Hydroxybutyrate also stimulated gluconeogenesis from serine by 70%, the effect being much higher than that of acetate (35%). No effect of 3-hydroxybutyrate or acetate on gluconeogenesis from pyruvate was seen. It is proposed that, in intact liver cells, pyruvate is transported into mitochondria in exchange for intramitochondrially generated acetoacetate, and that the mechanism proposed may be of physiological importance.

Possible interrelationship between gluconeogenesis and ketogenesis in the liver.

Effects of various ketogenic substrates on gluconeogenesis from lactate were examined. D,L-3-Hydroxybutyrate (5 mM) stimulated gluconeogenesis by 41%, the effect being the same as that of 5 mM acetate (49%). No stimulating effect of acetoacetate was observed; conversely, acetoacetate (up to 40 mM) partially or completely abolished the observed stimulating effects of acetate, oleate, and 3-hydroxybutyrate. The results suggest that, in intact liver cells, pyruvate is transported into mitochondria in exchange for acetoacetate and that an interrelationship between gluconeogenesis and ketogenesis at the level of mitochondrial pyruvate carrier may exist in the liver.

Combined effect of lysine and acetate on gluconeogenesis from lactate in isolated hepatocytes.

The effect of lysine combined with acetate on gluconeogenesis from lactate was studied in isolated rat hepatocytes. The combination was found to have a powerful stimulative effect which was significantly greater than that of isolated lysine or acetate. The stimulative effects of the two stimulants were mutually additive and were correlated [both singly and combined] to the free NADH and NAD+ concentration ratio in the cytoplasm.

Human heart aneurysm: biochemical and morphological characteristics.

The microscopic structure of left ventricular wall aneurysms and akinetic zones differs markedly. In samples obtained from akinetic zones, dystrophic changes and marked hypertrophy of muscle cells were observed with dispersed myofibrosis. In 1 case of left ventricular wall aneurysm, a pronounced nonspecific inflammatory reaction in the subepicardial layer was present. In all samples investigated, there were marked morphological signs of activated proteosynthesis, mainly aggregations of ribosomes, both in the muscle cells and in the fibroblasts. Utilization of glucose-14C was highest in samples with a predominant content of muscle cells. Pentose-phosphate shunt activity was highest in samples consisting mainly of fibrous scar tissue. The ability of the tissue homogenate to incorporate glycine-14C into the trichloracetic acid-precipitated protein was higher in samples containing predominantly connective tissue. The highest activity of the pentose-phosphate shunt, together with the highest rate of proteosynthesis, was observed in the sample from the left ventricular wall aneurysm with marked inflammatory infiltration of the tissue.
