Subrenal capsule assay of human tumor chemosensitivity.

Breast and colon tumor response to emoxyl, a nitroxyl derivative of daunomycin, was detected using human tumor heterotransplantation under the renal capsule of immunocompetent mice. The substitution of adriamycin by emoxyl in the combined therapy led to enhanced therapeutic efficacy. The evidence of enhanced response of breast tumors to emoxyl obtained during the histologic examination of xenografts is in good agreement with measurements of tumor fragment weight. It is suggested to use a quantitative kinetic index kappa calculated by the method of equivalent exponents for objective evaluation of tumor response to the drugs.

[Sensitivity of human tumor heterografts to a spin-labelled rubomycin derivative].

A degree of emoksil activity has been determined by applying the subrenal capsule assay methodology to fresh surgical explants of the normal immunocompetent mice. The ability of emoksil to inhibit growth of breast and colon tumours xenografts was determined. Emoksil substitution for adriamycin in the combination therapy increases the therapeutic activity. The kinetic criterion kappa calculated by the method of equivalent exponents was suggested to determine the tumour susceptibility to the drugs.

[Effect of the anthracycline antibiotics rubomycin and ruboxyl on the morphology and electric activity of the rat myocardium].

It has been shown that 2 months after discontinuation of rubomycin administration to rats at a dose of 0.7 mg/kg there is a considerable damage of cardiomyocyte morphology manifested in myofibril disintegration and contractions, vacuolization, as well as chromatin coagulation indicative of the inhibition of nuclear synthesis. The experiments on the papillary muscles isolated from the same hearts have demonstrated a depression of Na-conductivity and reduction in the force of isometric contractions. Ruboxyl at a dose of 3 mg/kg caused no changes in the morphology and electrical activity. It is suggested that depression of Na-conductivity is mediated by a decrease in the number of sarcolemmal channels due to the inhibition of the channel-forming protein synthesis.

[Ultrastructure of the ocular ciliary epithelium of the common frog].

The ultrastructure of the adult frog ciliary epithelium cells has definite regional differences. Cells of ciliary epithelium folds near the iris display morphological features characterizing its barrier and secretory functions which lead to the formation of aqueous humor. These are junctional complexes with tight junctions (zonula occludents) in the apical parts of contacting sides of cells of the inner leaf: a great quantity of mitochondria, ribosomes and various vesicles, well developed endoplasmic reticulum in the cytoplasm, much folded basal surface, gap junctions between cells of external and internal leaflets. In the mammalian inner epithelial layer different cell junctions are known to be arranged in a fixed spatial fashion. Unlike, in the frog's epithelium both zonula adherent and desmosomes may be found in any sequence. Tight junctions are formed during metamorphosis, on the place of focal junctions, whereas gap junctions, referred to earlier as "extended", start functioning between cells just on the very early stages of eye morphogenesis (Dabagyan et al., 1979). The epithelium of the posterior part of the ciliary fold and pars plana of the ciliary body have, in addition, the number of morphological sign indicating the cell involvement in the accomodational function of any eye (i. e. a majority of desmosomes binding all cells together and of zonulae adherentes, well developed intracellular skeleton of tonofilament bundles). These features are characteristic of the whole distal part of ciliary epithelium rather than of the place of attachment of zonula fiber only.

[Electron microscopic study of the intercellular junctions during the ciliary epithelial differentiation of the eye of the common frog].

Intercellular junctions in the eye ciliary epithelium were studied in Rana temporaria by means of transmission electron microscopy beginning from the stage of appearance of the light sensitivity in the larval eye and until the completion of metamorphosis. In the ciliary epithelium inner layer the apical parts of adjacent cells are tied by contacts forming the junction complex already at a very early developmental stage. The most apical position in this complex is occupied by the focal junction which appears to be an early stage of zonula occludens; this complex includes also zonula adherens and macula adherens which may follow in any succession focal junction or zonula occludens. No definite order was found in the localization of cell contacts between the outer and inner layers of ciliary epithelium, as well as between the side surfaces of the cells within each layer. Contacts were found everywhere termed as "lengthy" which may be considered as gap junctions. Regional differences were found in the ultrastructure of the ciliary epithelium cells with respect to unequal distribution of functional loads.

[Study of development of ciliary folds and Zinn's zonules in Rana temporaria L. by means of scanning electron microscopy].

The development of ciliary folds and zonul of Zinn has been studied in the eyes of the common frog Rana temporaria L. by means of scanning electron microscopy. The development of ciliary folds begins at the stage 45 by the flexure of the external layer in the ciliary zone. At the stage 46 this process involves the internal layer and the folds become two-layered. The zonules of Zinn form before the folds of internal layer of the ciliary epithelium begin to form as separate bundles of fibers. At the stage 45 they are already distinct. Later the ciliary filaments fold in 2 felt-like layers -- zonula which pass from the equatorial lens zone and attach near orbiculum ciliaris. In the place of attachment the margin of zonul repeats the relief of folds, thus attaching to their whole surface, and individual filaments go farther, to orbiculum ciliaris. All these processes take place in they eye prior to the beginning of metamorphosis.
