Pepsinogen polymorphism in human gastric mucosa in relation to gastric diseases.

The human stomach mucosa contains two main group of gastric proteinases. Both pepsinogen A (PGA) and pepsinogen C (PGC) consist of molecular variants, isozymogens, which can be separated electrophoretically, PGA was found to consist of five isozymogens (Pg1-Pg2), and PGC of two isozymogens (Pg6 and Pg7). Five hundred zymograms were examined and electrophoretic mobility of pepsinogens from patients with gastric cancer was found to be higher than from other gastric diseases. The ratio of isozymogens Pg3 to Pg5 differs in to great extent in various disease. Patients with ulcer disease have this value higher than 1, but patients with gastric cancer lower than 1. Patients with gastric ulcer have lower occurrence of Pg1 and SMP in antrum. In patients with gastric carcinoma lower concentration of PGA and also ratio PGA to PGC are observed.

[Immunohistologic detection of pepsin and gastricsin in carcinoma of the stomach].

Immunohistological tests for pepsin and gastricsin were carried out in bioptic samples of 51 patients with carcinoma of the stomach. Pepsin was detected in only 2 cases (4%), gastricsin in 26 patients (55%). Compared to the histological picture, the rate of incidence of gastricsin showed no difference between the intestinal and diffuse types of this tumour. In all examined cases, only gelatinous carcinomas were negative. Nor were any differences found in the cardia, the body or antrum. Surprisingly, these enzymes were found in the cytoplasma of neutrophylic granulocytes in the inflammatory infiltrate of the mucosal and tumorous stroma. Intestinal metaplasia of the epithelium was always negative even in the neighbourhood of positive tumours. The detected changes are evidence against the possibility of different histogenesis of the diffuse and intestinal forms of carcinoma of the stomach as well as against the possibility that intestinal metaplasia in a chronic inlammation of the stomach could be regarded as a direct first stage of the intestinal form of carcinoma of the stomach. In cases of metastases of unknown origin, pepsin and gastricsin cannot serve as markers due to their insufficient organ and cell specificities.

[Experimental models of gastric ulcers. Changes in the secretion of gastric glycoproteins in selected types].

The authors discuss possibilities of modelling of gastric ulcers in experiments on small laboratory animals. In four types of acute gastric ulcers and one type of chronic ulcer changes of glycoproteins in gastric mucus were investigated as to glycoproteins a protective effect on the gastric mucosa is ascribed. As indicator of secretion of these substances total hexose and total protein values were used. It was revealed that in homogenates the total hexose values were in all types of ulcers higher than in the control group. Total protein values were elevated in three types of ulcers. Elevated values of these substances were found also in gastric juice in ulcers after ligature of the pylorus.

[Pepsinogen C in the human prostate].

The authors investigated the proteolytic activity of the normal prostate, the prostate with benign hypertrophy and with cancer. Pepsinogen obtained from the normal prostate was isolated on DEAHP pearl cellulose and purified by means of affinite chromatography. Separation on affinite chromatography and by electrophoresis on agar confirmed that the pepsinogen is identical with gastric progastricsin. The proteolytic activity was examined by a modified method of Anson and Mirsky, and with the aid of synthetic substrates it was assessed quantitatively in all three examined groups. The enzyme activity of the carcinomatous tissue practically did not differ from the activity of the normal prostate. In beniming hypertrophy of the prostate the proteolytic activity assessed by the breakdown of haemoglobin is markedly reduced and when tested on the breakdown of synthetic substrates [For-Tyr-Tyr- and Ac-Phe- Tyr (I2)] it is practically zero.

Separation of human pepsin and gastricsin by affinity chromatography with an immobilized synthetic inhibitor.

Pepsin and gastricsin from human gastric juice were separated by affinity chromatography on Sepharose 4B containing the immobilized synthetic inhibitor of aspartic proteinases, Val-D-Leu-Pro-Phe-Phe-Val-D-Leu. These enzymes were bound to the support at low pH, and gastricsin was released at the same pH with buffer containing 20% dioxan. Pepsin was not released under these conditions, but was eluted at higher pH with buffer also containing 20% dioxan. To obtain perfect separations, it is recommended to use diluted samples. Proteinases from the homogenate of human gastric mucosa are isolated on DEAE-cellulose before separation by affinity chromatography. Pepsin and gastricsin from human gastric juice and human gastric mucosa separated on DEAE-cellulose and isolated by affinity chromatography, were electrophoretically pure.

Depression of experimental gastric ulcers and acute pancreatitis in rats treated by 5-azacytosine, 5-azacytidine and their N-methyl derivatives.

5-Azacytosine, 1-methyl-5-azacytosine and 5-azacytidine administered to rats with a ligated pylorus block gastric secretion, gastric acidity, the extent of hemorrhage and the number and size of gastric defects. The same drugs also depress the development of experimental acute pancreatitis mediated in rats by interstitial administration of 7.5% natrium cholate into the pancreas in vivo. The drugs affected the amount of abdominal fluid and 6 h after the treatment the pathological changes were significantly decreased.

Effects of N4-methyl and N4,N4-dimethyl derivatives of 5-azacytidine on liver RNA synthesis and gastric secretion in rats.

Of different N-alkyl substituted derivatives of 5-azacytidine tested for their action on gastric secretion in rats with ligated pylorus the most effective beside 5-azacytidine were N4-methyl and N4,N4-dimethyl derivatives. The drugs blocked gastric secretion, gastric acidity, the extent of hemorrhage and the number and size of gastric defects. Simultaneously a higher synthesis of RNA in the liver of drug-treated animals was observed.

Effects of cycloheximide and streptovitacin A on protein synthesis and gastric secretion in rats.

Cycloheximide and streptovitacin A administered in vivo to rats display a similar dual effect on the labelling of soluble liver proteins by valine-14C, and result in a similar enhancement of liver uridine kinase activity. On the other hand, in pylorus-ligated rats, both antibiotics markedly depress gastric secretion, acid output, and the level of mucoproteins and proteolytic activity in secreted juice. Streptovitacin A on a molar basis was in all cases 5 approximately 8 times more effective than cycloheximide.

Experimental neurosis and gastric secretion in dogs.

In four dogs (2 males and 2 females from one litter) with established gastric cannula gastric secretion was studied in control experiments and in induced experimental neurosis. Gastric secretion was stimulated by insulin. We monitored in individual 15 min. portions the amount of gastric juice, total HCl output, output of acid gastric proteinases, mucoproteins and some ions. The gastric juice was dialyzed and freeze dried. 50 mg of the lyophilisate was separated on Sephadex G 100. Macromolecular substances were fractionated into glycoproteins (peak I), acid gastric proteinases (peak II) and glycopeptides and polypeptides (peak III). The ratio of these individual macromolecular substances remainded constant in the same dog in all control experiments. However, there were significant differences between individual animals. Induction of experimental neurosis (by collision of the alimentary and avoidance reflex) gave rise to changes not only in the output of HCl and gastric proteinases, but also in the ratio of macromolecular substances. In the series of observed parameters these changes were of a different nature in males and females.

Effect of experimental neurosis on the secretion of acid gastric proteinases in dogs.

In experiments on dogs with gastric cannula we proved that the gastric juice, obtained after intravenous injection of insulin, contained after separation of DEAE Sephadex A-50 5--7 proteolytically active fractions. In four dogs of both sexes the ratio and number of individual fractions remained constant in control experiments. However, there were differences between the animals. After induction of experimental neurosis (by the collision of the alimentary and avoidance reflex) all animals showed the same marked changes in the chromatographic patterns of acid proteinases. There was a change not only in the ratio of two larger groups of proteolytically active fractions but also in the total number of all fractions. These changes persisted for 8--10 weeks after induction of experimental neurosis.

Inhibitory effect of various cytostatics and cycloheximide on acute experimental pancreatitis in rats.

Cycloheximide, 5-azacytidine, and 4-methoxybenzoyl-beta-bromoarylate (Cytembena) block the development of experimental acute pancreatitis in rats when mediated by the administration of 5% bile solution into the pancreas in vivo. Six hours after drug treatment the pathological changes, evaluated macroscopically or using histological sections of the pancreas, were significantly decreased. The drugs affected the amount of abdominal fluid and lowered its lipase and amylase activity. The known inhibitory mechanism of the active drugs and the possible advantage of cycloheximide for clinical use are briefly mentioned.
