Conversion of high grade lymphoma tumor cell line to intermediate grade with TPA and bryostatin 1 as determined by polypeptide analysis on 2D gel electrophoresis.

A high-resolution two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) was used for total cellular polypeptide mapping of two established lymphoma cell lines; MANCA, a Burkitt's line representing a high grade lymphoma (HGL) and WSU-NHL (nodular histiocytic lymphoma) representing an intermediate grade lymphoma (IGL). Gels were digitized and analysed with an image scanning computer using the Elsie 4 system. Polypeptide mapping revealed striking similarities in 1100 polypeptide spots in both types. However, three polypeptides were unique to HGL (Molecular mass/isoelectric point (Mr/PI): 39/4.4, 35/5.6, 33/4.8), and one to IGL (95/4.7). In order to investigate the kinetics of expression of these polypeptides, the two cell lines were treated with two protein kinase C (PKC) activators, tumour promoting 12-O tetradecanoylphorbol 13-acetate (TPA) and bryostatin 1. 2D-PAGE of the treated cells revealed that the HGL line loses its unique polypeptides and expresses a new one. The new polypeptide has the same Mr and PI as that unique to the untreated IGL (95/4.7). TPA or bryostatin 1 treatment of the IGL line for 72 h induced no significant changes. Our data show a unidirectional change from HGL to IGL, supporting the clinical notion that HGL is less differentiated than IGL. It also shows the similarity in the mode of action of bryostatin 1 and TPA in inducing these polypeptide changes.

Bacterial contamination of bone marrow grafts intended for autologous and allogeneic bone marrow transplantation. Incidence and clinical significance.

In a series of 100 bone marrow harvests, the incidence of bacterial contamination of the bone marrow graft was 17 percent. Ex vivo manipulation of some of the grafts prior to infusion may have caused additional bacterial contamination. All isolated bacteria were common skin flora, and no serious sequelae were observed in the patients receiving the culture-positive bone marrow grafts. Samples of harvested bone marrows purposely contaminated with an isolate of Staphylococcus epidermidis demonstrated a bactericidal property that was maximal early after bone marrow collection. Bone marrow collection and ex vivo manipulation may result in considerable bacterial contamination. Procedures must be developed to assure that marrow collection and processing do not result in clinically significant contamination.

Thymus-conditioned medium influences in vitro colony formation by hematopoietic progenitor cells.

Medium conditioned by mouse thymic cells cultured for 7 days in the presence of concanavalin A (Con A) and 5% human serum enhanced erythroid colony formation by CFU-E but not BFU-E. Medium conditioned by thymic cells for less than 7 days or more than 7 days inhibited erythroid colony formation. Enhancement of erythroid colony formation occurred at both suboptimal and saturating concentrations of erythropoietin. The thymus-conditioned medium also stimulated the growth of granulocyte/macrophage colonies by itself and in the presence of a saturating concentration of WEHI-3 conditioned medium. The conditioned medium contained neither burst-promoting activity nor erythropoietin. The erythroid colony-enhancing activity in the thymus-conditioned medium was heat stable, bound to immobilized Con A, lacked protease activity and migrated with an approximate molecular weight of 68,000 on gel filtration.
