The preliminary evaluation of degradation of substance P(SP) fragment's analogue less than Glu SP6-11 in the subcellular fractions from different areas of rat brain.

Peptidase(s) activity of different subcellular fractions isolated from cortex, hippocampus, midbrain, thalamus with hypothalamus, cerebellum and medulla oblongata exerted against less than Glu SP6-11 (3H-Phen8) was evaluated in "low-ionic" and similar (in composition) to both extracellular and intracellular conditions. The incubation of less than Glu SP6-11 with different fractions leaves the hexapeptide undegraded in the studied conditions in most cases. Peptidases activity results in the formation of the first of all C-terminal and exceptionally "internal" labelled products. Labelled N-terminal products were not seen. The most effective degradation in vitro of less than Glu SP6-11 takes place, in the majority of cases, in "low ionic" conditions when compared to those similar to extra or intracellular ones. The biggest total (per 1 g of wet mass) and specific activities against less than Glu SP6-11 can be shown in the hippocampus areas.

Role of theophylline during the action of SP in vitro on the activity of synaptosomal membrane ATPase (EC 3.6.1.3) from different areas of rat brain.

The in vitro action of SP and theophylline on the activities of (Na K) and (Ca Mg) dependent adenosine-5'-triphosphates (ATPase) of crude synaptosomal membranes (SM) of four different areas of rat brain was investigated. The highest activity of (Ca Mg)ATPase was observed in SM from hippocampus. In thalamus with hypothalamus region the highest activity of (Na K)ATPase and the lowest activity of (Ca Mg)ATPase occurred. It was found, that 10 mumol of SP stimulating "alternatively" both (Na K) and (Ca Mg)ATPase activities from different regions may constitute a regulating agent for ionic transport in CNS of rat. Theophylline on concentration of 5 mumol modulates the action of SP on the activities of the investigated enzymes.

Effects of analogues of substance P fragments on the MAO activity in rat brain.

The influence in vitro of analogues of Sp5-11 and SP6-11 substance P fragments on the activity of monoamine oxidase (MAO) in homogenates and crude mitochondrial fractions of rat brain was examined. The rat brain was divided into: I--cerebral cortex, II--hippocampus, III--midbrain, IV--thalamus with hypothalamus, V--cerebellum and VI--medulla oblongata. The obtained results proved that the analogues of SP fragments inhibit selectively the activity of the enzyme in the homogenates of cerebral cortex, hippocampus, midbrain and cerebellum. In the crude mitochondrial fractions the applied analogues of SP fragments caused a slight increase of the enzyme activity. The most significant changes in the activity of MAO were observed in hippocampus homogenate fraction.

Influence of Ca2+ and substance P on the (Ca Mg) ATPase (EC 3.6.1.3) activity of synaptosomes from different areas of rat brain.

The influence of SP in vitro on the (Ca Mg) ATPase activity of synaptosomes from cerebral cortex, hippocampus, midbrain and thalamus with hypothalamus of rats was studied. It was confirmed that SP increases the activity of the enzyme from hippocampus, midbrain and thalamus with hypothalamus. A condition of this stimulation is the maintenance of Ca2+ concentration between 1 and 2 X 10(-4) M. Ca2+ concentration above and below the optimal (0.2 mM), reduced the SP stimulation of (Ca Mg) ATPase activity.

Ca2+ Mg2+ -ATPase activity of synaptosome fraction and synaptosomal membranes from different areas of rat brain.

1. Crude synaptosomal fractions obtained from four areas of rat brain were studied; cerebral cortex, hippocampus, midbrain, thalamus with hypothalamus, using the Cotman & Matthews method (1971) Biochim, biophys. Acta 249, 350-394. 2. The purity of synaptosomal fractions was controlled by electron microscopy, and by determination of some marker enzymes such as: LDH, MAO, AChE and cytochrome-c oxidase. 3. Synaptosomes were disrupted by hypoosmotical shock. 4. Crude synaptosomal membrane preparations indicated on increased Ca2+ Mg2+ -ATPase activity in comparison to the activity of this enzyme in synaptosomal fractions. 5. The incubation of crude synaptosomal membranes with cAMP and theophylline caused the subsequent increase of Ca2+ Mg2+ -ATPase activity, but mainly, in hippocampal region.

[Behavior of urinary estriol level before normal labor].

The levels of oestriol in urine of 30 healthy pregnant women in the last 22 to 25 days of pregnancy were determined and the statistical analysis was performed. It was found that the levels of oestriol in urine were the highest in 19-th and 9-th days before labour. This form of curve of oestriol levels was found to be characteristic for pregnant women before normal labour.
